Proper Motions of PSRs B1757-24 and B1951+32: Implications for Ages and Associations
Over the last decade, considerable effort has been made to measure the proper
motions of the pulsars B1757-24 and B1951+32 in order to establish or refute
associations with nearby supernova remnants and to understand better the
complicated geometries of their surrounding nebulae. We present proper motion
measurements of both pulsars with the Very Large Array, increasing the time
baselines of the measurements from 3.9 yr to 6.5 yr and from 12.0 yr to 14.5
yr, respectively, compared to previous observations. We confirm the
non-detection of proper motion of PSR B1757-24, and our measurement of (mu_a,
mu_d) = (-11 +/- 9, -1 +/- 15) mas yr^{-1} confirms that the association of PSR
B1757-24 with SNR G5.4-1.2 is unlikely for the pulsar characteristic age of
15.5 kyr, although an association can not be excluded for a significantly
larger age. For PSR B1951+32, we measure a proper motion of (mu_a, mu_d) =
(-28.8 +/- 0.9, -14.7 +/- 0.9) mas yr^{-1}, reducing the uncertainty in the
proper motion by a factor of two compared to previous results. After correcting
to the local standard of rest, the proper motion indicates a kinetic age of ~51
kyr for the pulsar, assuming it was born near the geometric center of the
supernova remnant. The radio-bright arc of emission along the pulsar proper
motion vector shows time-variable structure, but moves with the pulsar at an
approximately constant separation ~2.5", lending weight to its interpretation

Surface structure influences contact killing of bacteria by copper
Copper kills bacteria rapidly by a mechanism that is not yet fully resolved. The antibacterial property of copper has raised interest in its use in hospitals, in place of plastic or stainless steel. On the latter surfaces, bacteria can survive for days or even weeks. Copper surfaces could thus provide a powerful accessory measure to curb nosocomial infections. We here investigated the effect of the copper surface structure on the efficiency of contact killing of Escherichia coli, an aspect which so far has received very little attention. It was shown that electroplated copper surfaces killed bacteria more rapidly than either polished copper or native rolled copper. The release of ionic copper was also more rapid from electroplated copper compared to the other materials. Scanning electron microscopy revealed that the bacteria nudged into the grooves between the copper grains of deposited copper. The findings suggest that, in terms of contact killing, more efficient copper surfaces can be engineered
Calpain- and talin-dependent control of microvascular pericyte contractility and cellular stiffness
Pericytes surround capillary endothelial cells and exert contractile forces modulating microvascular tone and endothelial growth. We previously described pericyte contractile phenotype to be Rho GTPase- and α-smooth muscle actin (αSMA)-dependent. However, mechanisms mediating adhesion-dependent shape changes and contractile force transduction remain largely equivocal. We now report that the neutral cysteine protease, calpain, modulates pericyte contractility and cellular stiffness via talin, an integrin-binding and F-actin associating protein. Digital imaging and quantitative analyses of living cells reveal significant perturbations in contractile force transduction detected via deformation of silicone substrata, as well as perturbations of mechanical stiffness in cellular contractile subdomains quantified via atomic force microscope (AFM)-enabled nanoindentation. Pericytes overexpressing GFP-tagged talin show significantly enhanced contractility (~ two-fold), which is mitigated when either the calpain-cleavage resistant mutant talin L432G or vinculin are expressed. Moreover, the cell-penetrating, calpain-specific inhibitor termed CALPASTAT reverses talin-enhanced, but not Rho GTP-dependent, contractility. Interestingly, our analysis revealed that CALPASTAT, but not its inactive mutant, alters contractile cell-driven substrata deformations while increasing mechanical stiffness of subcellular contractile regions of these pericytes. Altogether, our results reveal that calpain-dependent cleavage of talin modulates cell contractile dynamics, which in pericytes may prove instrumental in controlling normal capillary function or microvascular pathophysiology.National Science Foundation (U.S.) (CAREER Award)American Society for Engineering Education. Symptoms in pediatric asthmatics and air pollution: differences in effects by symptom severity, anti-inflammatory medication use and particulate averaging time.
Experimental research in humans and animals points to the importance of adverse respiratory effects from short-term particle exposures and to the importance of proinflammatory effects of air pollutants, particularly O(subscript)3. However, particle averaging time has not been subjected to direct scientific evaluation, and there is a lack of epidemiological research examining both this issue and whether modification of air pollutant effects occurs with differences in asthma severity and anti-inflammatory medication use. The present study examined the relationship of adverse asthma symptoms (bothersome or interfered with daily activities or sleep) to O(3) and particles (less than or equal to)10 micrometer (PM10) in a Southern California community in the air inversion zone (1200-2100 ft) with high O(3) and low PM (R = 0.3). A panel of 25 asthmatics 9-17 years of age were followed daily, August through October 1995 (n = 1,759 person-days excluding one subject without symptoms). Exposures included stationary outdoor hourly PM10 (highest 24-hr mean, 54 microgram/m(3), versus median of 1-hr maximums, 56 microgram/m(3) and O(3) (mean of 1-hr maximums, 90 ppb, 5 days (greater than or equal to)120 ppb). Longitudinal regression analyses utilized the generalized estimating equations (GEE) model controlling for autocorrelation, day of week, outdoor fungi, and weather. Asthma symptoms were significantly associated with both outdoor O(3) and PM(10) in single pollutant- and co-regressions, with 1-hr and 8-hr maximum PM(10) having larger effects than the 24-hr mean. Subgroup analyses showed effects of current day PM(10) maximums were strongest in 10 more frequently symptomatic (MS) children: the odds ratios (ORs) for adverse symptoms from 90th percentile increases were 2.24 [95% confidence interval (CI), 1.46-3.46] for 1-hr PM10 (47 microgram/m(3); 1.82 (CI, 1.18-2.81) for 8-hr PM10 (36 microgram/m(3); and 1.50 (CI, 0.80-2.80) for 24-hr PM10 (25 microgram/m(3). Subgroup analyses also showed the effect of current day O(subscript)3 was strongest in 14 less frequently symptomatic (LS) children: the ORs were 2.15 (CI, 1.04-4.44) for 1-hr O(3) (58 ppb) and 1.92 (CI, 0.97-3.80) for 8-hr O(3) (46 ppb). Effects of 24-hr PM10 were seen in both groups, particularly with 5-day moving averages (ORs were 1.95 for MS and 4. 03 for LS; p(less than or equal to)0.05). The largest effects were in 7 LS children not on anti-inflammatory medications [5-day, 8-hr PM10, 9.66 (CI, 2.80-33.21); current day, 1-hr O(3), 4.14 (CI, 1.71-11.85)]. Results suggest that examination of short-term particle excursions, medication use, and symptom severity in longitudinal studies of asthma yields sensitive measures of adverse respiratory effects of air pollution

Structure-property relationships in mechanically stimulated Sorghum bicolor stalks
Mechanical properties of plants and underlying structure-property relationships are important for agricultural purposes as well as for biomimetic concepts. In this study, the effect of mechanical stimulation on morphology and bending properties of the stalk was investigated for Sorghum bicolor (Poaceae), a widely used drought-tolerant biomass grass. An experimental set-up allowing for defined growth and mechanical perturbation (flexing) during a defined growth period was designed. Mechanical properties of individual internodes of the stalk were determined by three-point bending tests. We found that the three investigated lines showed differences in their general bending strength in the non-stimulated condition. However, similar high range of bending strength values was measured for all plant lines after they underwent the mechanical stimulation procedure. The anatomy of internode cross-sections was examined to evaluate structure-property relationships. An increased thickness of the outer sclerenchymatous tissue was observed for internodes with higher bending strength values. Dried internodes fail under lower strains but showed higher bending strength. These findings show that mechanosensitivity in sorghum is dependent on genetic as well as environmental factors. The experimental system presented here offers new straight-forward possibilities for S. bicolor line selection for applications requiring mechanical strength of the stalk
Ground state cooling, quantum state engineering and study of decoherence of ions in Paul traps
We investigate single ions of in Paul traps for quantum
information processing. Superpositions of the S electronic ground state
and the metastable D state are used to implement a qubit. Laser light
on the S D transition is used for the
manipulation of the ion's quantum state. We apply sideband cooling to the ion
and reach the ground state of vibration with up to 99.9% probability. Starting
from this Fock state , we demonstrate coherent quantum state
manipulation. A large number of Rabi oscillations and a ms-coherence time is
observed. Motional heating is measured to be as low as one vibrational quantum

Use of marginal organs in kidney transplantation for marginal recipients: too close to the margins of safety?
<p>Abstract</p> <p>Objective</p> <p>Due to organ shortage, average waiting time for a kidney in Germany is about 4 years after start of dialysis. Number of kidney grafts recovered can only be maintained by accepting older and expanded criteria donors. The aim of this study was to analyse the impact of donor and recipient risk on kidney long-term function.</p> <p>Methods</p> <p>All deceased kidney transplantations were considered. We retrospectively studied 332 patients between 2002 and 2006; divided in 4 groups reflecting donor and recipient risk.</p> <p>Results</p> <p>Non-marginal recipients were less likely to receive a marginal organ (69 of 207, 33%) as compared to marginal recipients, of whom two-thirds received a marginal organ (p < 0.0001). Graft function significantly differed between the groups, but detrimental effect of marginal recipient status on eGFR after 12 months (-6 ml/min/1.73 qm, 95% CI -2 to -9) was clearly smaller than the effect of marginal donor status (-10 ml/min/1.73 qm, 95% CI -7 to -14).</p> <p>Conclusions</p> <p>As we were able to show expanded criteria donor has a far bigger effect on long-term graft function than the "extra risk" recipient. Although there have been attempts to define groups of recipients who should be offered ECD kidneys primarily the discussion is still ongoing.</p
Tactile defensiveness and impaired adaptation of neuronal activity in the Fmr1 knockout mouse model of autism
Sensory hypersensitivity is a common symptom in autism spectrum disorders (ASDs), including Fragile X Syndrome (FXS), and frequently leads to tactile defensiveness. In mouse models of ASDs, there is mounting evidence of neuronal and circuit hyperexcitability in several brain regions, which could contribute to sensory hypersensitivity. However, it is not yet known whether or how sensory stimulation might trigger abnormal sensory processing at the circuit level or abnormal behavioral responses in ASD mouse models, especially during an early developmental time when experience-dependent plasticity shapes such circuits. Using a novel assay, we discovered exaggerated motor responses to whisker stimulation in young Fmr1 knockout (KO) mice (postnatal days (P) 14-16), a model of FXS. Adult Fmr1 KO mice actively avoided a stimulus that was innocuous to wild-type controls, a sign of tactile defensiveness. Using in vivo two-photon calcium imaging of Layer 2/3 barrel cortex neurons expressing GCaMP6s, we found no differences between wild-type and Fmr1 KO mice in overall whisker-evoked activity, though 45% fewer neurons in young Fmr1 KO mice responded in a time-locked manner. Notably, we identified a pronounced deficit in neuronal adaptation to repetitive whisker stimulation in both young and adult Fmr1 KO mice. Thus, impaired adaptation in cortical sensory circuits is a potential cause of tactile defensiveness in autism
Primary Care Provider Awareness and Attitudes Regarding Updated BRCA1/2 Screening Recommendations and Genetic Testing
Detection of BRCA1/2 gene mutations opens the door for breast cancer prevention; however, many high-risk individuals are not screened until after their first cancer diagnosis. The United States Preventive Services Task Force (USPSTF) updated its BRCA1/2 screening recommendations in 2019. This study explores provider awareness of this update and attitudes towards BRCA1/2 genetic testing. A survey was sent to Obstetrics/ Gynecology (Ob/Gyn), Internal Medicine (IM), and Family Medicine (FM) providers at Thomas Jefferson University Hospitals (TJUH). 66% of responders felt familiar with BRCA1/2 screening guidelines, and 38% were aware of the 2019 USPSTF update. Ob/Gyn providers were more familiar with BRCA1/2 screening recommendations compared to IM and FM providers (p= 0.003). PCP concerns centered around the potential for negative psychological and financial impact, unnecessary testing, and test result uncertainty. Results highlight the need to increase provider awareness of BRCA1/2 screening measures and guidelines while addressing concerns regarding the impact of genetic testing on patients
