    Penelitian ini bertujuan mengetahui keberadaan antibodi dari bakteri brucella pada sapi perah di Kecamatan Purwoharjo Kabupaten Banyuwangi menggunakan Rose Bengal Test (RBT). Metode yang digunakan dalam penelitian ini adalah metode analisis diskriptif dengan pendekatan kuantitatif, yaitu metode yang menggambarkan keberadaan antibodi Brucella pada sapi perah dengan cepat. Informasi pendukung diperoleh dari survei peternak dengan teknik wawancara. Sampel ditentukan dengan rumus slovin dengan ukuran sampel sebanyak 75 sampel dari 294 ekor sapi pada populasi sapi perah di Kecamatan Purwoharjo. Hasil penelitian menunjukan 2 sampel dinyatakan positif Brucella dari 75 sampel. Hasil penelitian ini dapat disimpulkan bahwa reaktor Brucella ditemukan pada sapi perah di Kecamatan Purwoharjo Kabupaten Banyuwangi yang berpotensi menyebabkan penyakit Brucellosis. Harapan dari penelitian ini adalah data yang diperoleh dapat diuji ulang dan diuji lebih lanjut dalam penelitian selanjutnya sehingga hasil dapat dikonfirmasi

    The reproductive characteristics of a precocious line of E. tenella sporozoite were investigated in chicken embryos and the implications of the findings appraised. While it was possible to reproduce E. tenella in chicken embryos, other species could not be propagated efficiently. The results of this expriments showed that the reproductive index of this line of E. tenella in chickens, the propagation in eggs is presently more efficient and yields are variab1e. There was considerable vation in the oocyst yields among experiments, although the yields within experiment among individual embryos did not vary greatly. It should be possible to achieve higher oocyst yields enhanced embryo-adaptation (leading to decreased embryonic mortality) through increased serial passage in eggs, thereby maintaining immunogenicity in the vaccinated host.     Isolation of Actinomycetes from mangrove sediments at Ujung Pangkah, Gresik, Indonesia

    This research was conducted to get actinomycetes isolate from mangrove sediments at Ujung Pangkah in Gresik-Indonesia. Sediment samples were taken from mangrove rhizosphere in Ujung Pangkah on 3 different location sampling areas. Ten grams of soil sample was accurately weighed and transferred to add 100 mL of sterile NaCl 0.9% mixed well for 10 minutes. The resultant solution was serially diluted up to 10 -12 (10 -3, 10 -4 until 10 -12) with NaCl 0.9%. One millilitre of each intermediate dilution (10 -3, 10 -4 until 10 -12) was added to 10 mL of sterile molten Starch Casein Agar medium which has been supplemented using both chloramphenicol and griseofulvin each one 0.05 ppm individually in separate flasks. The plates were incubated for the growth of actinomycetes colonies at 28◦C and observed intermittently during incubation. After 7 days of incubation, the colonies showing the characteristics of actinomycetes (rough, chalky, powderyappearance radiating growth and leathery texture) were observed. Identification of actinomycetes isolates was conducted by observing macroscopic characteristic of the colony, microscopic of conidial, bacterial cell and the ability of bacteria to resist from acid alcohol. Identification was done based on Bergey’s manual of determinative if bacteriology. The result of this research obtained 9 different isolates actinomycetes. The 9 isolate was identified as 3 genera from actinomycetes. The genera of isolate obtained are Micromonospora, Nocardia and Streptomyces. Key words: Actinomycetes isolate, Incubation, Bacteriolog

    Antifulgal potency agints Candida albicans (ATCC 10231) and its Activity as biosurfactant of WNA 4.1.13 fermented growth of Sediment from mangrove Wonorejo Surabaya

    The purpose of this study was to identify the genus of bacteria that produces biosurfactants and its potency as antifungal to Candida albicans (ATCC 10231). WNA bacteria 4.1.13 were isolated from Wonorejo mangrove sediments in Surabaya. The bacteria were identified by examining the morphology and and cell colony.Cell morphology identification was carried out by Gram staining and spores identification. Biosurfactant activity was carried out by oil spreading test, drop collapse test and parafilm test. Antifungal activity was performed by agar diffusion method. The results showed that WNA 4.1.13 isolates from Surabaya Wonorejo mangrove sediments were rod shaped, including Gram positive and had endospores located at the center of bacterial cells. Based on the morphological character of the colony and genus cell bacterial isolates WNA 4.1.13 including Bacillus sp. The result showed that in the broth collapse test, the droplets were flat, forming a clean zone in the oil spreading test and widened diameter in the parafilm test. Antifungal activity test showed inhibition zones formed by fermentation broth isolates of WNA 4.1.13. In conclusion, the fermentation of broth of WNA 4.1.13 isolates from the Wonorejo Surabaya have biosurfactant activity and antifungal potency against Candida albicans (ATCC 10231).     Potensi Ekstrak Daun Afrika (Vernonia Amygdalina Delile) sebagai Antibakterial terhadap Bakteri Escherichia Coli ATCC 25922

    Tujuan dari penelitian ini adalah untuk mengetahui potensi ekstrak metanol daun Afrika (Vernonia amygdalina Delile) sebagai antibakteri terhadap bakteri Escherichia coli ATCC 25922. Isolat E. coli dibiakan pada media Muller Hinton Agar (MHA). Uji antibakteri menggunakan metode difusi sumuran. Kontrol negatif (K-) pada lubang sumuran hanya diberikan pengencer ekstrak CMC Na 0,5%. Kontrol positif (K+) pada lubang sumuran diberi dengan eugenol. Perlakuan (P1, P2, P3, P4) diberikan ekstrak metanol daun Afrika (Vernonia amygdalina Delile) dengan masing - masing konsentrasi 100%, 75%, 50% dan 25%. Rancangan penelitian menggunakan metode Rancangan Acak Lengkap (RAL), dengan enam perlakuan dan empat pengulangan. Data dianalisis dengan ANOVA, dilanjutkan dengan uji Duncan. Hasil analisis zona hambat menunjukkan, K+ memiliki perbedaan yang nyata dengan semua perlakuan K-, P1, P2, P3, dan P4. Serta K- ditemukan tidak berbeda nyata dengan perlakuan P1, P2, P3 dan P4. Hasil penelitian menunjukkan bahwa setiap perlakuan tidak menghasilkan zona hambat sehingga tidak berbeda nyata dengan kontrol negatif (CMC Na 0,5%)

    Deteksi Residu Antibiotik Oksitetrasiklin pada Susu Kambing Peranakan Etawah di Kelurahan Kalipuro, Banyuwangi dengan Uji Bioassay

    Oksitetrasiklin diketahui sebagai salah satu antibiotik yang sering digunakan dalam menangani kasus mastitis pada hewan ternak. Penelitian ini merupakan penelitian deskriptif untuk mengetahui ada atau tidaknya residu oksitetrasiklin pada susu kambing Peranakan Etawah (PE) di Kelurahan Kalipuro, Banyuwangi. Sampel susu di koleksi kemudian dilakukan pengujian residu antibiotik dengan metode uji bioassay menggunakan bakteri standar Bacillus cereus ATCC 11778. Penelitian yang dilakukan pada 26 sampel susu yang di ambil secara acak berasal dari ternak kambing perah PE di dapatkan hasil yang negatif. Hal ini di tunjukkan dengan tidak terbentuknya zona hambat di sekitar well yang diisi sampel susu. Berbeda dengan kontrol postif oksitetrasiklin dengan konsentrasi 1 µg/ml yang menghasilkan zona hambatan sekitar 18 mm. Dapat disimpulkan bahwa 100% dari 26 jumlah sampel susu kambing PE yang di ambil secara acak dan di periksa dari Kelurahan Kalipuro, Banyuwangi tidak mengandung residu antibiotik oksitetrasiklin
