    Model choice: An Operational Comparison of Stochastic Streamflow Models for Droughts

    The rapid development of stochastic or operational hydrology over the past 10 years has led to the need for some comparative analyses of the currently available long-term persistence models. Five annual stochastic streamflow generation models (autoregressive, autoregressive-moving-average (ARMA), ARMA-Markov, fast fractional Gaussian noise, and broken line) are compared on their ability to preserve drought-related time series properties and annual statistics. Using Monte Carlo generation procedures and comparing the average generated statistics and drought or water supply properties, a basis is established to evalute model performance on four different Utah study streams. A seasonal disaggregation model is applied to each of the generated annual models for each of the four study streams at a monthly disaggregation level. A model choice strategy is presented for the water resources engineer to select an annual stochastic streamflow model based on values of the historic time series\u27 lag-one serial correlation and Hurst coefficient. Procedures are presented for annual and seasonal model parameter estimatino, calibration, and generation. Techniques are included such as normality, trend-analysis, and choice of model. User oriented model parameter estimation techniques that are easy and efficient to use are presented in a systematic manner. The ARMA-Markov and ARMA models are judged to be the best overall models in terms of preserving the short and long term persistence statistics for the four historic time series studied. The broken line model is judged to be the best model in terms of minimizing the evonomic regret as determined by an agricultural crop production function. Documentation and listings of the computer programs that were used for the stochastic models\u27 parameter estimation, generation, and camparison techniques are presented in a supplementary appendix

    An Immature Retroviral RNA Genome Resembles a Kinetically Trapped Intermediate State

    Retroviral virions initially assemble in an immature form that differs from that of the mature infectious particle. The RNA genomes in both immature and infectious particles are dimers, and interactions between the RNA dimer and the viral Gag protein ensure selective packaging into nascent immature virions. We used high-sensitivity selective 2′-hydroxyl acylation analyzed by primer extension (SHAPE) to obtain nucleotide-resolution structural information from scarce, femtomole quantities of Moloney murine leukemia virus (MuLV) RNA inside authentic virions and from viral RNA extracted from immature (protease-minus) virions. Our secondary structure model of the dimerization and packaging domain indicated that a stable intermolecular duplex known as PAL2, previously shown to be present in mature infectious MuLV particles, was sequestered in an alternate stem-loop structure inside immature virions. The intermediate state corresponded closely to a late-folding intermediate that we detected in time-resolved studies of the free MuLV RNA, suggesting that the immature RNA structure reflects trapping of the intermediate folding state by interactions in the immature virion. We propose models for the RNA-protein interactions that trap the RNA in the immature state and for the conformational rearrangement that occurs during maturation of virion particles

    Insights into Starch Coated Nanozero Valent Iron-Graphene Composite for Cr(VI) Removal from Aqueous Medium

    Embedding nanoparticles into an inert material like graphene is a viable option since hybrid materials are more capable than those based on pure nanoparticulates for the removal of toxic pollutants. This study reports for the first time on Cr(VI) removal capacity of novel starch stabilized nanozero valent iron-graphene composite (NZVI-Gn) under different pHs, contact time, and initial concentrations. Starch coated NZVI-Gn composite was developed through borohydrate reduction method. The structure and surface of the composite were characterized by scanning electron microscopy (SEM), X-ray diffraction spectroscopy (XRD), Fourier transform infrared spectroscopy (FTIR), Brunauer-Emmett-Teller (BET), and point of zero charge (pHpzc). The surface area and pHpzc of NZVI-Gn composite were reported as 525 m2 g−1 and 8.5, respectively. Highest Cr(VI) removal was achieved at pH 3, whereas 67.3% was removed within first few minutes and reached its equilibrium within 20 min obeying pseudo-second-order kinetic model, suggesting chemisorption as the rate limiting process. The partitioning of Cr(VI) at equilibrium is perfectly matched with Langmuir isotherm and maximum adsorption capacity of the NZVI-Gn composite is 143.28 mg g−1. Overall, these findings indicated that NZVI-Gn composite could be utilized as an efficient and magnetically separable adsorbent for removal of Cr(VI)

    Influence of culture medium on in-vitro biofilm formation by Candida species

    Objectives: Objective of this study was to establish an in vitro biofilm on the 96 well plates and to determine the efficacy of three different culture media on biofilm formation of Candida albicans and C. tropicalis Methods: A 96 well sterile, polystyrene plate was inoculated using 10^6 cell/ml of C. albicans and C. tropicalis suspensions and the growth rate of planktonic cells was determined by measuring the absorbance (OD492) at 2 hour intervals. Adhesion of Candidial cells to initiate the biofilm formation in the presence of three culture media (Yeast Nitrogen Base (YNB) supplemented with 100 mM glucose, Sabouraud Dextrose Broth (SDB) and RPMI1640) was quantified using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and Crystal Violet (CV) assay after 90 minutes. Biofilms of C. albicans, C. tropicalis and 1:1 co-biofilms were developed and the growth rates were quantified at 24 hours’ time intervals. Scanning electron microscope (SEM) was performed to assess the architecture. Results: Planktonic cells of both C. albicans and C. tropicalis showed maximum growth with SDB. C. albicans and co-biofilm adhesion were significantly facilitated with RPMI1640 and the best medium for C. tropicalis adhesion was YNB. Biofilms showed the maximum growth rate in RPMI 1640. C. tropicalis exhibited the minimum growth with all three culture media.Conclusions: The maximum growth rate for planktonic C. albicans and C. tropicalis was achieved with SDB. However RPMI 1640 was the best medium for growth of biofilms

    Objectives: This study was aimed to determine the efficacy of four routine laboratory culture media onbiofilm formation of Pseudomonas aeruginosa, Staphylococcus aureus and Enterococcus feacalis.Methods: A sterile flat bottom 96 well plate was inoculated using 0.5 McFarland equivalent standardcell suspension of P. aeruginosa, S. aureus and E. feacalis and the growth rate of planktonic cells wasquantified by measuring the optical density (OD492) at two hour intervals. Influence of culture mediumon adhesion of bacteria as an initial step of biofilm formation in the presence of four culture media(Nutrient broth (NB), Brain Heart Infusion (BHI) broth, Luria-Bertani (LB) broth and RPMI 1640) wasquantified using MTT (3-[4, 5- dimethylthiazole-2-yl]-2, 5- diphenyltetrazolium bromide) assay after90 minutes adhesion. Biofilms of P. aeruginosa, S. aureus, E. feacalis and their 1:1 mixed biofilmswere developed and the growth was quantified using MTT metabolic activity at 24 hour time intervals.Scanning electron microscopy (SEM) was performed to assess the ultrastructure.Results: On comparing the relative growth of the bacteria in different culture media, the maximumgrowth of all three planktonic cultures was achieved using BHI broth. All mono species and mixedspecies cultures exhibited their maximum adhesion in the presence of RPMI 1640. All biofilm exhibitedthe maximum growth in BHI broth. SEM imaging had shown the enhanced growth of ultrastructure ofthe biofilm with the presence of BHI broth.Conclusions: The maximum planktonic and biofilm growth was achieved with BHI broth. However,bacterial adhesion was enhanced in the presence of RPMI 1640

    Reproductive health for refugees by refugees in Guinea III: maternal health

    BACKGROUND: Maternal mortality can be particularly high in conflict and chronic emergency settings, partly due to inaccessible maternal care. This paper examines associations of refugee-led health education, formal education, age, and parity on maternal knowledge, attitudes, and practices among reproductive-age women in refugee camps in Guinea. METHODS: Data comes from a 1999 cross-sectional survey of 444 female refugees in 23 camps. Associations of reported maternal health outcomes with exposure to health education (exposed versus unexposed), formal education (none versus some), age (adolescent versus adult), or parity (nulliparous, parous, grand multiparous), were analysed using logistic regression. RESULTS: No significant differences were found in maternal knowledge or attitudes. Virtually all respondents said pregnant women should attend antenatal care and knew the importance of tetanus vaccination. Most recognised abdominal pain (75%) and headaches (24%) as maternal danger signs and recommended facility attendance for danger signs. Most had last delivered at a facility (67%), mainly for safety reasons (99%). Higher odds of facility delivery were found for those exposed to RHG health education (adjusted odds ratio 2.03, 95%CI 1.23-3.01), formally educated (adjusted OR 1.93, 95%CI 1.05-3.92), or grand multipara (adjusted OR 2.13, 95%CI 1.21-3.75). Main reasons for delivering at home were distance to a facility (94%) and privacy (55%). CONCLUSIONS: Refugee-led maternal health education appeared to increase facility delivery for these refugee women. Improved knowledge of danger signs and the importance of skilled birth attendance, while vital, may be less important in chronic emergency settings than improving facility access where quality of care is acceptable

    The role and scope of practice of midwives in humanitarian settings:a systematic review and content analysis

    Abstract Background Midwives have an essential role to play in preparing for and providing sexual and reproductive health (SRH) services in humanitarian settings due to their unique knowledge and skills, position as frontline providers and geographic and social proximity to the communities they serve. There are considerable gaps in the international guidance that defines the scope of practice of midwives in crises, particularly for the mitigation and preparedness, and recovery phases. We undertook a systematic review to provide further clarification of this scope of practice and insights to optimise midwifery performance. The review aimed to determine what SRH services midwives are involved in delivering across the emergency management cycle in humanitarian contexts, and how they are working with other professionals to deliver health care. Methods Four electronic databases and the websites of 33 organisations were searched between January and March 2017. Papers were eligible for inclusion if they were published in English between 2007 and 2017 and reported primary research pertaining to the role of midwives in delivering and performing any component of sexual and/or reproductive health in humanitarian settings. Content analysis was used to map the study findings to the Minimum Initial Service Package (MISP) for SRH across the three phases of the disaster management cycle and identify how midwives work with other members of the health care team. Results Fourteen studies from ten countries were included. Twelve studies were undertaken in conflict settings, and two were conducted in the context of the aftermath of natural disasters. We found a paucity of evidence from the research literature that examines the activities and roles undertaken by midwives across the disaster management cycle. This lack of evidence was more apparent during the mitigation and preparedness, and recovery phases than the response phase of the disaster management cycle. Conclusion Research-informed guidelines and strategies are required to better align the scope of practice of midwives with the objectives of multi-agency guidelines and agreements, as well as the activities of the MISP, to ensure that the potential of midwives can be acknowledged and optimised across the disaster management cycle

    Microbial cycling of isoprene, the most abundantly produced biological volatile organic compound on Earth

    Isoprene (2-methyl-1,3-butadiene), the most abundantly produced biogenic volatile organic compound (BVOC) on Earth, is highly reactive and can have diverse and often detrimental atmospheric effects, which impact on climate and health. Most isoprene is produced by terrestrial plants, but (micro)algal production is important in aquatic environments, and the relative bacterial contribution remains unknown. Soils are a sink for isoprene, and bacteria that can use isoprene as a carbon and energy source have been cultivated and also identified using cultivation-independent methods from soils, leaves and coastal/marine environments. Bacteria belonging to the Actinobacteria are most frequently isolated and identified, and Proteobacteria have also been shown to degrade isoprene. In the freshwater-sediment isolate, Rhodococcus strain AD45, initial oxidation of isoprene to 1,2-epoxy-isoprene is catalyzed by a multicomponent isoprene monooxygenase encoded by the genes isoABCDEF. The resultant epoxide is converted to a glutathione conjugate by a glutathione S-transferase encoded by isoI, and further degraded by enzymes encoded by isoGHJ. Genome sequence analysis of actinobacterial isolates belonging to the genera Rhodococcus, Mycobacterium and Gordonia has revealed that isoABCDEF and isoGHIJ are linked in an operon, either on a plasmid or the chromosome. In Rhodococcus strain AD45 both isoprene and epoxy-isoprene induce a high level of transcription of 22 contiguous genes, including isoABCDEF and isoGHIJ. Sequence analysis of the isoA gene, encoding the large subunit of the oxygenase component of isoprene monooxygenase, from isolates has facilitated the development of PCR primers that are proving valuable in investigating the ecology of uncultivated isoprene-degrading bacteria
