    How can I tell how successful this course has been? is becoming an increasingly common question in tertiary education. This interest in tertiary teaching and learning is reflected in the fact that one-half of all Australian universities now have tertiary teaching units. There are a number of reasons why evaluation is important. Firstly, discrepancies between the actual and the ideal situation can be detected, causes identified and corrective measures instituted at all stages of the evaluative model, to serve the interests of increased efficiency and improved staff and student satisfaction. Secondly, courses which are continually being evaluated are better able to meet changing demands from students, from the society which the educational institution is serving, and also better able to adjust to internal changes in staff numbers and expertise. Thirdly, evaluation properly conducted can provide a statement of accountability

    A clone encoding carboxymethyl cellulase activity was isolated during functional screening of a human gut metagenomic library using Lactococcus lactis MG1363 as heterologous host. The insert carried a glycoside hydrolase family 9 (GH9) catalytic domain with sequence similarity to a gene from Coprococcus eutactus ART55/1. Genome surveys indicated a limited distribution of GH9 domains among dominant human colonic anaerobes. Genomes of C. eutactus-related strains harboured two GH9-encoding and four GH5-encoding genes, but the strains did not appear to degrade cellulose. Instead, they grew well on β-glucans and one of the strains also grew on galactomannan, galactan, glucomannan and starch. Coprococcus comes and Coprococcus catus strains did not harbour GH9 genes and were not able to grow on β-glucans. Gene expression and proteomic analysis of C. eutactus ART55/1 grown on cellobiose, β-glucan and lichenan revealed similar changes in expression in comparison to glucose. On β-glucan and lichenan only, one of the four GH5 genes was strongly upregulated. Growth on glucomannan led to a transcriptional response of many genes, in particular a strong upregulation of glycoside hydrolases involved in mannan degradation. Thus, β-glucans are a major growth substrate for species related to C. eutactus, with glucomannan and galactans alternative substrates for some strains

    Exogenously added GPI-anchored tissue inhibitor of matrix metal loproteinase-1 (TIMP-1) displays enhanced and novel biological activities

    The family of tissue inhibitors of metalloproteinases (TIMPs) exhibits diverse physiological/biological functions including the inhibition of active matrix metalloproteinases, regulation of proMMP activation, cell growth, and the modulation of angiogenesis. TIMP-1 is a secreted protein that can be detected on the cell surface through its interaction with surface proteins. The diverse biological functions of TIMP-1 are thought to lie, in part, in the kinetics of TIMP-1/MMP/surface protein interactions. Proteins anchored by glycoinositol phospholipids (GPIs), when purified and added to cells in vitro, are incorporated into their surface membranes. A GPI anchor was fused to TIMP-1 to generate a reagent that could be added directly to cell membranes and thus focus defined concentrations of TIMP-1 protein on any cell surface independent of protein-protein interaction. Unlike native TIMP-1, exogenously added GPI-anchored TIMP-1 protein effectively blocked release of MMP-2 and MMP-9 from osteosarcoma cells. TIMP-1-GP1 was a more effective modulator of migration and proliferation than TIMP-1. While control hTIMP-1 protein did not significantly affect migration of primary microvascular endothelial cells at the concentrations tested, the GPI-anchored TIMP-1 protein showed a pronounced suppression of endothelial cell migration in response to bFGF. In addition, TIMP-1-GPI was more effective at inducing microvascular endothelial proliferation. In contrast, fibroblast proliferation was suppressed by the agent. Reagents based on this method should assist in the dissection of the protease cascades and activities involved in TIMP biology. Membrane-fixed TIMP-1 may represent a more effective version of the protein for use in therapeutic expression

    Motor subtype as a predictor of future working memory performance in idiopathic Parkinson\u27s disease

    Parkinson’s disease is a progressive neurodegenerative disorder associated with reduced spatial and verbal working memory ability. There are two established motor subtypes of PD, tremor dominant (TD) and postural instability and gait difficulty (PIGD). This study used structural equation modelling to explore the longitudinal relationship between the two subtypes and working memory assessed at a 2-year follow-up. The study comprised 84 males and 30 females (N = 114), aged between 39 and 85 (M = 64.82, SD = 9.23) with confirmed PD. There was no significant relationship between motor subtype at Time 1 and working memory at Time 2. Postural symptom severity at Time 1 predicted Time 2 spatial working memory for the PIGD subtype (p = .011) but not the TD subtype. Tremor symptoms were not associated with Time 2 working memory in either subtype. Predictive significance of Time 1 postural symptoms only in the PIGD subtype suggests an interaction between symptom dominance (subtype) and symptom severity that future subtyping should consider. This study demonstrates a predictive relationship between postural difficulties and working memory performance assessed at a 2-year follow-up. Establishing physical symptoms as predictors of cognitive change could have significant clinical importance

    Variation in xylem structure from tropics to tundra: Evidence from vestured pits

    Bordered pits play an important role in permitting water flow among adjacent tracheary elements in flowering plants. Variation in the bordered pit structure is suggested to be adaptive in optimally balancing the conflict between hydraulic efficiency (conductivity) and safety from air entry at the pit membrane (air seeding). The possible function of vestured pits, which are bordered pits with protuberances from the secondary cell wall of the pit chamber, could be increased hydraulic resistance or minimized vulnerability to air seeding. These functional hypotheses have to be harmonized with the notion that the vestured or nonvestured nature of pits contains strong phylogenetic signals (i.e., often characterize large species-rich clades with broad ecological ranges). A literature survey of 11,843 species covering 6,428 genera from diverse climates indicates that the incidence of vestured pits considerably decreases from tropics to tundra. The highest frequencies of vestured pits occur in deserts and tropical seasonal woodlands. Moreover, a distinctly developed network of branched vestures is mainly restricted to warm habitats in both mesic and dry (sub)tropical lowlands, whereas vestures in woody plants from cold and boreal arctic environments are usually minute and simple. A similar survey of the frequency of exclusively scalariform perforation plates illustrates that the major ecological trend of this feature is opposite that of vestured pits. These findings provide previously undescribed insights suggesting that vessels with vestured pits and simple perforation plates function as an efficient hydraulic system in plants growing in warm environments with periodical or continuous drought stress. NaturalisPlant science

    Research on the Geography of Agricultural Change: Redundant or Revitalized?

    Future research directions for agricultural geography were the subject of debate in Area in the late 1980s. The subsequent application of political economy ideas undoubtedly revived interest in agricultural research. This paper argues that agricultural geography contains greater diversity than the dominant political economy discourse would suggest. It reviews ‘other’ areas of agricultural research on policy, post-productivism, people, culture and animals, presenting future suggestions for research. They should ensure that agricultural research continues revitalized rather than redundant into the next millennium
