    Response of Siratro (Macroptilium atropurpureum Urb. Rabaceae) to Vesicular-arbuscular Mycorrhizal Fungi and Rhizobium sp. in Sterilized Soil

    The effect of inoculation of six species of vesicular-arbuscular mycorrhizal fungi viz., Gigaspora margarita, Glomus mosseae, G. monosporum, G. versiformis, G. fasciculatum, G. deserticola and Rhizobium sp. on growth and NPK content in siratro (Macroptilium atropurpureum) was studied. The mycorrhizal species were isolated from the rhizosphere forest soils of the Western Ghats and multiplied in pot culture with Sorghum bicolor as the host plant. Rhizobium sp. isolated from fresh nodules of siratro was cultured and maintained in yeast extract mannitol broth. The influence of VAM fungi and Rhizobium sp. either singly or in combination on growth and nutrient contents of siratro showed statistically significant increase after only 60 days of plant growth. Dual inoculation with VAM fungi and Rhizobium increased plant nodule number and tissue NPK content.     Action Mechanism of Inhibin α-Subunit on the Development of Sertoli Cells and First Wave of Spermatogenesis in Mice

    Inhibin is an important marker of Sertoli cell (SC) activity in animals with impaired spermatogenesis. However, the precise relationship between inhibin and SC activity is unknown. To investigate this relationship, we partially silenced both the transcription and translation of the gene for the α-subunit of inhibin, Inha, using recombinant pshRNA vectors developed with RNAi-Ready pSIREN-RetroQ-ZsGreen Vector (Clontech Laboratories, Mountain View, Calif). We found that Inha silencing suppresses the cell-cycle regulators Cyclin D1 and Cyclin E and up-regulates the cell-cycle inhibitor P21 (as detected by Western blot analysis), thereby increasing the number of SCs in the G1 phase of the cell cycle and decreasing the amount in the S-phase of the cell cycle (as detected by flow cytometry). Inha silencing also suppressed Pdgfa, Igf1, and Kitl mRNA levels and up-regulated Tgfbrs, Inhba, Inhbb, Cyp11a1, Dhh, and Tjp1 mRNA levels (as indicated by real-time polymerase chain reaction [PCR] analysis). These findings indicate that Inha has the potential to influence the availability of the ligand inhibin and its antagonist activin in the SC in an autocrine manner and inhibit the progression of SC from G1 to S. It may also participate in the development of the blood–testis barrier, Leydig cells, and spermatogenesis through its effect on Dhh, Tjp1, Kitl, and Pdgfa. Real-time PCR and Western blot analyses of Inha, Inhba, and Inhbb mRNA and Inha levels over time show that Inha plays an important role in the formation of round spermatid during the first wave of spermatogenesis in mice

    Effects of cast-bracing of the knee on physical performance in healthy subjects. Veldhuizen JW, Verstappen FT, Koene FM, Greep JM. Effects of application of a hinged cast-brace on thigh muscle strength and physical performance tests were studied in eight healthy volunteers. The cast-brace was applied to one leg for four weeks. The subjects were free to move around. The extra loading by the cast-brace was determined in a progressive uphill walk test on a treadmill with measurements of oxygen uptake, heart rate and plasma lactate concentration. Submaximal oxygen uptake in the uphill walk test was raised significantly (average 9%) on the day after application. After four weeks of cast-brace wearing the submaximal oxygen uptake in the uphill walk test had decreased, but remained elevated (average 4%) even one day after removal. Heart rate was significantly higher (average 7%) during cast application and after removal. Plasma lactate concentration, however, was not influenced. To investigate the effect of four weeks cast-bracing various performance tests to judge the thigh muscle function were taken before application and after removal. No significant changes in peak torque of knee flexion and extension, in physiologic variables at submaximal running pace during treadmill exercise, maximal running speed, 60 m dash, or in jump height were found.
