HCV derived from sera of HCV-infected patients induces pro-fibrotic effects in human primary fibroblasts by activating GLI2
Hepatitis C virus (HCV) infection is a leading cause of liver fibrosis, especially in developing countries. The process is characterized by the excess accumulation of ECM that may lead, over time, to hepatic cirrhosis, liver failure and also to hepatocarcinoma. The direct role of HCV in promoting fibroblasts trans-differentiation into myofibroblasts, the major fibrogenic cells, has not been fully clarified. In this study, we found that HCV derived from HCV-infected patients infected and directly induced the trans-differentiation of human primary fibroblasts into myofibroblasts, promoting fibrogenesis. This effect correlated with the activation of GLI2, one of the targets of Hedgehog signaling pathway previously reported to be involved in myofibroblast generation. Moreover, GLI2 activation by HCV correlated with a reduction of autophagy in fibroblasts, that may further promoted fibrosis. GLI2 inhibition by Gant 61 counteracted the pro-fibrotic effects and autophagy inhibition mediated by HCV, suggesting that targeting HH/GLI2 pathway might represent a promising strategy to reduce the HCV-induced fibrosis
Modified mattress sutures vs running sutures in uterine closure: which is better?
Background: Conventional closure of uterus has been known to bear risk of scar dehiscence and scar rupture in subsequent pregnancies and thus, a study was conducted to compare the outcome of uterine closure with modified mattress manner and running manner and to establish a better method of uterine closure during caesarean section. Objective was to compare the conventional single layer running sutures and single layer modified mattress sutures for closure of uterus in caesarean section and find out which method is superior.Methods: This prospective interventional study was carried out in Dhiraj Hospital, a tertiary care hospital in Vadodara. 60 pregnant women in the study criteria were equally divided randomly into 2 groups. Uterine closure was done in single layered sutures, one by running sutures and other group by modified mattress sutures.Results: Uterine scar thickness on 8th day and 6 months post-operatively was significantly more in single layered suturing by modified mattress suture compared to running suture (p <0.05).Conclusions: Uterine closure by single layered modified mattress suture is better in comparison to conventional single layer running suture
H++: a server for estimating pK(a)s and adding missing hydrogens to macromolecules
The structure and function of macromolecules depend critically on the ionization (protonation) states of their acidic and basic groups. A number of existing practical methods predict protonation equilibrium pK constants of macromolecules based upon their atomic resolution Protein Data Bank (PDB) structures; the calculations are often performed within the framework of the continuum electrostatics model. Unfortunately, these methodologies are complex, involve multiple steps and require considerable investment of effort. Our web server provides access to a tool that automates this process, allowing both experts and novices to quickly obtain estimates of pKs as well as other related characteristics of biomolecules such as isoelectric points, titration curves and energies of protonation microstates. Protons are added to the input structure according to the calculated ionization states of its titratable groups at the user-specified pH; the output is in the PQR (PDB + charges + radii) format. In addition, corresponding coordinate and topology files are generated in the format supported by the molecular modeling package AMBER. The server is intended for a broad community of biochemists, molecular modelers, structural biologists and drug designers; it can also be used as an educational tool in biochemistry courses

Macroscopic coherence of a single exciton state in a polydiacetylene organic quantum wire
We show that a single exciton state in an individual ordered conjugated
polymer chain exhibits macroscopic quantum spatial coherence reaching tens of
microns, limited by the chain length. The spatial coherence of the k=0 exciton
state is demonstrated by selecting two spatially separated emitting regions of

Stark Spectroscopy and Radiative Lifetimes in Single Self-Assembled CdTe Quantum Dots
We present studies on Coulomb interactions in single self-assembled CdTe
quantum dots. We use a field effect structure to tune the charge state of the
dot and investigate the impact of the charge state on carrier wave functions.
The analysis of the quantum confined Stark shifts of four excitonic complexes
allows us to conclude that the hole wave function is softer than electron wave
function, i. e. it is subject to stronger modifications upon changing of the
dot charge state. These conclusions are corroborated by time-resolved
photoluminescence studies of recombination lifetimes of different excitonic
complexes. We find that the lifetimes are notably shorter than expected for
strong confinement and result from a relatively shallow potential in the
valence band. This weak confinement facilitates strong hole wave function
redistributions. We analyze spectroscopic shifts of the observed excitonic
complexes and find the same sequence of transitions for all studied dots. We
conclude that the universality of spectroscopic shifts is due to the role of
Coulomb correlations stemming from strong configuration mixing in the valence

On a Symmetry Argument for the Guidance Equation in Bohmian Mechanics
Bohmian mechanics faces an underdetermination problem: when it comes to solving the measurement problem, alternatives to the Bohmian guidance equation work just as well as the official guidance equation. DĂŒrr, Goldstein, and ZanghĂŹ have argued that of the candidate guidance equations, the official guidance equation is the simplest Galileanâinvariant candidate. This symmetry argumentâif it workedâwould solve the underdetermination problem. But the argument does not work. It fails because it rests on assumptions about how Galilean transformations (especially boosts) act on the wavefunction that are (in this context) unwarranted. My discussion has larger morals about the physical significance of certain mathematical results (like, for example, Wignerâs theorem) in nonâorthodox interpretations of quantum mechanics
