Toward a script theory of guidance in computer-supported collaborative learning
This article presents an outline of a script theory of guidance for computer-supported collaborative learning (CSCL). With its four types of components of internal and external scripts (play, scene, role, and scriptlet) and seven principles, this theory addresses the question how CSCL practices are shaped by dynamically re-configured internal collaboration scripts of the participating learners. Furthermore, it explains how internal collaboration scripts develop through participation in CSCL practices. It emphasizes the importance of active application of subject matter knowledge in CSCL practices, and it prioritizes transactive over non-transactive forms of knowledge application in order to facilitate learning. Further, the theory explains how external collaboration scripts modify CSCL practices and how they influence the development of internal collaboration scripts. The principles specify an optimal scaffolding level for external collaboration scripts and allow for the formulation of hypotheses about the fading of external collaboration scripts. Finally, the article points towards conceptual challenges and future research questions
The H.E.S.S. central data acquisition system
The High Energy Stereoscopic System (H.E.S.S.) is a system of Imaging
Atmospheric Cherenkov Telescopes (IACTs) located in the Khomas Highland in
Namibia. It measures cosmic gamma rays of very high energies (VHE; >100 GeV)
using the Earth's atmosphere as a calorimeter. The H.E.S.S. Array entered Phase
II in September 2012 with the inauguration of a fifth telescope that is larger
and more complex than the other four. This paper will give an overview of the
current H.E.S.S. central data acquisition (DAQ) system with particular emphasis
on the upgrades made to integrate the fifth telescope into the array. At first,
the various requirements for the central DAQ are discussed then the general
design principles employed to fulfil these requirements are described. Finally,
the performance, stability and reliability of the H.E.S.S. central DAQ are
presented. One of the major accomplishments is that less than 0.8% of

Detection of very-high-energy gamma-ray emission from the vicinity of PSR B1706-44 with H.E.S.S
The energetic pulsar PSR B1706-44 and the adjacent supernova remnant (SNR)
candidate G 343.1-2.3 were observed by H.E.S.S. during a dedicated
observational campaign in 2007. A new source of very-high-energy (VHE; E > 100
GeV) gamma-ray emission, HESS J1708-443, was discovered with its centroid at
RA(J2000) = 17h08m10s and Dec(J2000) = -44d21', with a statistical error of 3
arcmin on each axis. The VHE gamma-ray source is significantly more extended
than the H.E.S.S. point-spread function, with an intrinsic Gaussian width of
0.29 +/- 0.04 deg. Its energy spectrum can be described by a power law with a
photon index Gamma = 2.0 +/- 0.1 (stat) +/- 0.2 (sys). The integral flux
measured between 1-10 TeV is ~17% of the Crab Nebula flux in the same energy
range. The possible associations with PSR B1706-44 and SNR G343.1-2.3 are

Potential Neutrino Signals from Galactic Gamma-Ray Sources
The recent progress made in Galactic gamma-ray astronomy using the High
Energy Stereoskopic System (H.E.S.S.) instrument provides for the first time a
population of Galactic TeV gamma-rays, and hence potential neutrino sources,
for which the neutrino flux can be estimated. Using the energy spectra and
source morphologies measured by H.E.S.S., together with new parameterisations
of pion production and decay in hadronic interactions, we estimate the signal
and background rates expected for these sources in a first-generation water
Cherenkov detector (ANTARES) and a next-generation neutrino telescope in the
Mediterranean Sea, KM3NeT, with an instrumented volume of 1 km^3. We find that
the brightest gamma-ray sources produce neutrino rates above 1 TeV, comparable
to the background from atmospheric neutrinos. The expected event rates of the
brightest sources in the ANTARES detector make a detection unlikely. However,
for a 1 km^3 KM3NeT detector, event rates of a few neutrinos per year from
these sources are expected, and the detection of individual sources seems
possible. Although generally these estimates should be taken as flux upper
limits, we discuss the conditions and type of gamma-ray sources for which the
neutrino flux predictions can be considered robust.Comment: 20 pages, 4 figures; v2: ERROR in energy scale of KM3NeT effective

Electrostatic colloid-membrane complexation
We investigate numerically and on the scaling level the adsorption of a
charged colloid on an oppositely charged flexible membrane. We show that the
long ranged character of the electrostatic interaction leads to a wrapping
reentrance of the complex as the salt concentration is varied. The membrane
wrapping depends on the size of the colloid and on the salt concentration and
only for intermediate salt concentration and colloid sizes we find full
wrapping. From the scaling model we derive simple relations for the phase
boundaries between the different states of the complex, which agree well with
the numerical minimization of the free energy.Comment: 7 page, 11 figure

Role of a Transbilayer pH Gradient in the Membrane Fusion Activity of the Influenza Virus Hemagglutinin: Use of the R18 Assay to Monitor Membrane Merging
It had been suggested that influenza virus-mediated membrane fusion might be dependent on a pH gradient across a target membrane. We have designed experiments in which this issue could be addressed. Two populations of liposomes were prepared, both simulating the plasma membrane of target cells, but with the pH of the internal aqueous medium buffered either at pH 7.4 (physiological cytosol pH) or at pH 5.0 (endosomal pH at which influenza virus displays maximal fusion activity). By monitoring fusion using the R18 assay, we found that the internal pH of the target liposomes did not influence membrane merging as mediated by the influenza virus hemagglutinin, thus demonstrating that a transmembrane pH gradient is not required in this fusion process
Heart failure is independently associated with white matter lesions: Insights from the population-based LIFE-Adult Study
Aims: White matter lesions (WML) are common structural alterations in the white matter of the brain and their prevalence increases with age. They are associated with cerebral ischaemia and cognitive dysfunction. Patients with heart failure (HF) are at risk for cognitive decline. We hypothesized that the presence and duration of HF are associated with WML. Methods and results: The LIFE-Adult Study is a population-based study of 10 000 residents of Leipzig, Germany. WML were quantitated in 2490 participants who additionally underwent cerebral MRI using the Fazekas score. Mean age was 64 years, and 46% were female; 2156 (86.6%) subjects had Fazekas score of 0-1, and 334 (13.4%) had Fazekas score of 2-3. Thirty participants had a medical history of HF, 1019 had hypertension, and 51 had a history of stroke. Median left ventricular ejection fraction of the participants with HF was 57% (interquartile ranges 54-62). Age, troponin T, NT-proBNP, body mass index, history of acute myocardial infarction, stroke, HF, and diabetes were positively associated with WML in univariate analysis. On multivariate analysis, age, hypertension, stroke, and HF were independently associated with WML. The odd's ratio for the association of WML (Fazekas 2-3) with HF was 2.8 (95% CI 1.2-6.5; P = 0.019). WML increased with longer duration of HF (P = 0.036 for trend). Conclusions: In addition to age, hypertension, and stroke, the prevalence and duration of HF are independently associated with WML. This observation sets the stage to investigate the prognostic value of WML in HF and the impact of HF therapies on WML
