Design, fabrication, assembly and delivery of a laboratory prototype of a residual gas analyzer
The design, development, and testing of a wide mass range residual gas analyzer which will be one component of an integrated real time contamination monitor system are described. The instrument has been developed and tested to the laboratory prototype phase, demonstrating the performance that can be expected from a flight instrument of similar design. The instrument's analyzer is of the quadrupole type and a cold cathode ion source is employed as the ionizer. The associated electronics supply all necessary operating and mass sweep voltages for the ionizer, analyzer and electron multiplier ion detector. The instrument features a very fast linear electrometer with automatic range changing. The full mass range of 2 to 300 amu is automatically and repetitively scanned every sixty seconds and suitable telemetry outputs are provided for intensity and mass identification as well as a digital identification of the electrometer range
A High-Quality Professional Development for Teachers of Grades 3–6 for Implementing Engineering into Classrooms
With the increasing emphasis on integrating engineering into K-12 classrooms to help meet the needs of our complex and multidisciplinary society, there is an urgent need to investigate teachers\u27 engineering-focused professional development experiences as they relate to teacher learning, implementation, and student achievement. This study addresses this need by examining the effects of a professional development program focused on engineering integration, and how teachers chose to implement engineering in their classrooms as a result of the professional development. 198 teachers in grades 3–6 from 43 schools in 17 districts participated in a yearlong professional development program designed to help integrate the new state science standards, with a focus on engineering, into their teaching. Posters including lesson plans and student artifacts were used to assess teachers\u27 engineering practices and the implementation in their classrooms. Results indicated that the majority of the teachers who participated in the professional development were able to effectively implement engineering design lessons in their classrooms suggesting that the teachers\u27 success in implementing engineering lessons in their classroom was closely related to the structure of the professional development program
Matter effects in the D0-D0bar system
We discuss the impact of matter effects in the D0-D0bar system. We show that
such effects could, in principle, be measured, but that they cannot be used to
probe the mass difference x_D or the lifetime difference y_D. This occurs
because the mixing effects and the matter effects decouple at short times. We
also comment briefly on the B systems.Comment: 6 pages, RevTe

Pairing of Competitive and Topologically Distinct Regulatory Modules Enhances Patterned Gene Expression
Biological networks are inherently modular, yet little is known about how modules are assembled to enable coordinated and complex functions. We used RNAi and time series, whole-genome microarray analyses to systematically perturb and characterize components of a Caenorhabditis elegans lineage-specific transcriptional regulatory network. These data are supported by selected reporter gene analyses and comprehensive yeast one-hybrid and promoter sequence analyses. Based on these results, we define and characterize two modules composed of muscle- and epidermal-specifying transcription factors that function together within a single cell lineage to robustly specify multiple cell types. The expression of these two modules, although positively regulated by a common factor, is reliably segregated among daughter cells. Our analyses indicate that these modules repress each other, and we propose that this cross-inhibition coupled with their relative time of induction function to enhance the initial asymmetry in their expression patterns, thus leading to the observed invariant gene expression patterns and cell lineage. The coupling of asynchronous and topologically distinct modules may be a general principle of module assembly that functions to potentiate genetic switches.Molecular and Cellular Biolog
Optimal entanglement witnesses for continuous-variable systems
This paper is concerned with all tests for continuous-variable entanglement
that arise from linear combinations of second moments or variances of canonical
coordinates, as they are commonly used in experiments to detect entanglement.
All such tests for bi-partite and multi-partite entanglement correspond to
hyperplanes in the set of second moments. It is shown that all optimal tests,
those that are most robust against imperfections with respect to some figure of
merit for a given state, can be constructed from solutions to semi-definite
optimization problems. Moreover, we show that for each such test, referred to
as entanglement witness based on second moments, there is a one-to-one
correspondence between the witness and a stronger product criterion, which
amounts to a non-linear witness, based on the same measurements. This
generalizes the known product criteria. The presented tests are all applicable
also to non-Gaussian states. To provide a service to the community, we present
the documentation of two numerical routines, FULLYWIT and MULTIWIT, which have
been made publicly available.Comment: 14 pages LaTeX, 1 figure, presentation improved, references update
The role of pathology in an investigation of an outbreak of West Nile encephalitis in New York, 1999.
An outbreak of encephalitis occurred in New York City in late August 1999, the first caused by West Nile virus in North America. Histopathologic and immunopathologic examinations performed on human autopsy materials helped guide subsequent laboratory and epidemiologic investigations that led to identification of the etiologic agent
The First Human Epitope Map of the Alphaviral E1 and E2 Proteins Reveals a New E2 Epitope with Significant Virus Neutralizing Activity
Although the murine immune response to Venezuelan equine encephalitis virus (VEEV) is well-characterized, little is known about the human antibody response to VEEV. In this study we used phage display technology to isolate a panel of 11 VEEV-specfic Fabs from two human donors. Seven E2-specific and four E1-specific Fabs were identified and mapped to five E2 epitopes and three E1 epitopes. Two neutralizing Fabs were isolated, E2-specific F5 and E1-specific L1A7, although the neutralizing capacity of L1A7 was 300-fold lower than F5. F5 Fab was expressed as a complete IgG1 molecule, F5 native (n) IgG. Neutralization-escape VEEV variants for F5 nIgG were isolated and their structural genes were sequenced to determine the theoretical binding site of F5. Based on this sequence analysis as well as the ability of F5 to neutralize four neutralization-escape variants of anti-VEEV murine monoclonal antibodies (mapped to E2 amino acids 182–207), a unique neutralization domain on E2 was identified and mapped to E2 amino acids 115–119
Cell-cell adhesion regulates Merlin/NF2 interaction with the PAF complex
The PAF complex (PAFC) coordinates transcription elongation and mRNA processing and its CDC73/parafibromin subunit functions as a tumour suppressor. The NF2/Merlin tumour suppressor functions both at the cell cortex and nucleus and is a key mediator of contact inhibition but the molecular mechanisms remain unclear. In this study we have used affinity proteomics to identify novel Merlin interacting proteins and show that Merlin forms a complex with multiple proteins involved in RNA processing including the PAFC and the CHD1 chromatin remodeller. Tumour-derived inactivating mutations in both Merlin and the CDC73 PAFC subunit mutually disrupt their interaction and growth suppression by Merlin requires CDC73. Merlin interacts with the PAFC in a cell density-dependent manner and we identify a role for FAT cadherins in regulating the Merlin-PAFC interaction. Our results suggest that in addition to its function within the Hippo pathway, Merlin is part of a tumour suppressor network regulated by cell-cell adhesion which coordinates post-initiation steps of the transcription cycle of genes mediating contact inhibition
