    Spikelets in pyramidal neurons: generating mechanisms, distinguishing properties, and functional implications

    Spikelets are small spike-like depolarizations that are found in somatic recordings of many neuron types. Spikelets have been assigned important functions, ranging from neuronal synchronization to the regulation of synaptic plasticity, which are specific to the particular mechanism of spikelet generation. As spikelets reflect spiking activity in neuronal compartments that are electrotonically distinct from the soma, four modes of spikelet generation can be envisaged: (1) dendritic spikes or (2) axonal action potentials occurring in a single cell as well as action potentials transmitted via (3) gap junctions or (4) ephaptic coupling in pairs of neurons. In one of the best studied neuron type, cortical pyramidal neurons, the origins and functions of spikelets are still unresolved; all four potential mechanisms have been proposed, but the experimental evidence remains ambiguous. Here we attempt to reconcile the scattered experimental findings in a coherent theoretical framework. We review in detail the various mechanisms that can give rise to spikelets. For each mechanism, we present the biophysical underpinnings as well as the resulting properties of spikelets and compare these predictions to experimental data from pyramidal neurons. We also discuss the functional implications of each mechanism. On the example of pyramidal neurons, we illustrate that several independent spikelet-generating mechanisms fulfilling vastly different functions might be operating in a single cell

    Comparison of Therapeutic Effects between Pulsed and Continuous Wave 810-nm Wavelength Laser Irradiation for Traumatic Brain Injury in Mice

    Background and Objective Transcranial low-level laser therapy (LLLT) using near-infrared light can efficiently penetrate through the scalp and skull and could allow non-invasive treatment for traumatic brain injury (TBI). In the present study, we compared the therapeutic effect using 810-nm wavelength laser light in continuous and pulsed wave modes in a mouse model of TBI. Study Design/Materials and Methods TBI was induced by a controlled cortical-impact device and 4-hours post-TBI 1-group received a sham treatment and 3-groups received a single exposure to transcranial LLLT, either continuous wave or pulsed at 10-Hz or 100-Hz with a 50% duty cycle. An 810-nm Ga-Al-As diode laser delivered a spot with diameter of 1-cm onto the injured head with a power density of 50-mW/cm2 for 12-minutes giving a fluence of 36-J/cm2. Neurological severity score (NSS) and body weight were measured up to 4 weeks. Mice were sacrificed at 2, 15 and 28 days post-TBI and the lesion size was histologically analyzed. The quantity of ATP production in the brain tissue was determined immediately after laser irradiation. We examined the role of LLLT on the psychological state of the mice at 1 day and 4 weeks after TBI using tail suspension test and forced swim test. Results The 810-nm laser pulsed at 10-Hz was the most effective judged by improvement in NSS and body weight although the other laser regimens were also effective. The brain lesion volume of mice treated with 10-Hz pulsed-laser irradiation was significantly lower than control group at 15-days and 4-weeks post-TBI. Moreover, we found an antidepressant effect of LLLT at 4-weeks as shown by forced swim and tail suspension tests. Conclusion The therapeutic effect of LLLT for TBI with an 810-nm laser was more effective at 10-Hz pulse frequency than at CW and 100-Hz.     Three set of experiments were performed in an enclosed space (open-field) and in an open space (elevated platform). The surface of the open-field and the elevated platform were divided in nine equal squares. Rats were exposed (without previous habituation) in a unique session (experiment 1) or three consecutive sessions (experiment 2) either to an open-field (enclosed space) or to an elevated platform (open space) with and without an object on the centre of the field. In experiment 3, rats were exposed (without previous habituation) either to an enclosed or an open space on five consecutive sessions, one session a day. They were tested in an object recognition test in sessions 1, 3 and 5. In sessions 2 and 4, no objects were present. In experiment 1, we recorded the latency, frequency and duration of entries into different areas of the field. In experiment 3, we recorded the latency, frequency and duration of contacts with objects in addition to entries into different areas of the field. The first experiment demonstrates that rats exposed for the first time to an enclosed or an open space do not express neophobia toward novel objects in the absence of walls that surround an open-field. They crossed frequently into and spent more time in areas occupied with an object than in unoccupied areas. After two sessions of habituation to an empty open space or an empty enclosed space, the latency of first approach to a novel object is reduced while the frequency and duration of approaches are increased. The third experiment on object recognition confirmed that rats do not avoid novel objects; they made frequent visit and spent more time in the corner of the field occupied with an object than in empty corners. Recording of crossings provided detailed information about the patterns of exploratory behavior of rats but failed to reveal discrimination between novel and familiar objects which was evident in both open and enclosed space with recording of contacts with objects on the fifth exposure

    MK-801 increases baseline level of anxiety in mice introduced to a spatial memory task without prior habituation

    c57BL/6J mice were introduced to a nine arm radial maze without prior habituation and trained in the acquisition of a working memory task in 16 sessions, one session per day. In this maze mice need to climb onto an upward inclined bridge in order to reach and cross onto an arm. They received in each session an i.p. injection of MK-801 (0.1 mg/kg) 30 min before training or immediately after training. MK-801 pretreated mice made significantly more entries onto the bridges, fewer entries onto the arms and took significantly longer time to make a first arm visit compared to saline and MK-801 post-treated mice during the first 3 session blocks (4 sessions per block). These results indicate that MK-801 induced anxiety which was extended throughout the first 3 session blocks. MK-801 pre-treated mice made also significantly more errors and required more sessions to reach the criterion compared to saline and MK-801 post-treated mice. Administration of MK-801 after training did not affect the acquisition of the task. The present results indicate that MK-801 pre-treatment impaired the acquisition of a spatial task and this can be accounted for by its effect on the baseline level of anxiety which was elevated. The introduction of mice to the acquisition of the task without prior habituation demonstrates that a drug treatment can affect learning and memory by increasing and/or prolonging anxiety. Such effect may be confounded with learning and memory performance and not detected with pre-habituation training procedures, particularly when the number of sessions is determined a-priori

    Structural modifications of swift-ion-bombarded metallic glasses studied by high-energy X-ray synchrotron radiation

    Modifications of structural arrangements including relaxation processes were investigated in swift-heavy-ion-bombarded Fe73Cu1Nb3Si16B7 metallic glass. The irradiation of as-quenched amorphous ribbons was accomplished with 3 MeV u−1 (u is atomic mass unit) and 11.1 MeV u−1 Au ions with total fluences of up to 1 × 1013 ions cm−2. Specimens irradiated with 3 MeV u−1 ions were subsequently annealed for 4 h at 350 °C. Ion-irradiated samples including those heat-treated were studied using in situ temperature-dependent diffraction of high-energy X-ray synchrotron radiation (wavelength of 0.020689 nm). Cyclic heating–cooling regimes were applied during the measurements. A tendency towards a higher degree of disorder was revealed as a function of the ion fluence in both types of alloys. During the first heating–cooling cycle, a remarkable hysteresis is observed in the position and line width of the first diffraction peak, especially in the as-quenched irradiated alloys. The hysteresis increases with rising ion fluence but vanishes completely after the first heating–cooling cycle. No hysteresis is found in the irradiated and subsequently annealed alloys. The irradiation-induced structural modifications have some features similar to the effects caused by mechanical deformation

    The main purpose of this study is to determine whether the tourist's social distance perceptions towards the local people are reducing thanks to tourism and the causes of tourist's social distance perceptions. The Social Distance Scale of Bogardus was employed to prepare the questionnaires that were used in the study. Domestic tourists (n = 400) visiting the Southeastern Anatolia Region (before and after their visit) and people who had not visited the region (n = 263) before were asked to complete the questionnaire.
