    Effect of obesity and metabolic syndrome on plasma oxysterols and fatty acids in human

    BACKGROUND: Obesity and the related entity metabolic syndrome are characterized by altered lipid metabolism and associated with increased morbidity risk for cardiovascular disease and cancer. Oxysterols belong to a large family of cholesterol-derived molecules known to play crucial role in many signaling pathways underlying several diseases. Little is known on the potential effect of obesity and metabolic syndrome on oxysterols in human. OBJECTIVES: In this work, we questioned whether circulating oxysterols might be significantly altered in obese patients and in patients with metabolic syndrome. We also tested the potential correlation between circulating oxysterols and fatty acids. METHODS: 60 obese patients and 75 patients with metabolic syndrome were enrolled in the study along with 210 age- and sex-matched healthy subjects, used as control group. Plasma oxysterols were analyzed by isotope dilution GC/MS, and plasma fatty acids profiling was assessed by gas chromatography coupled with flame ionization detection. RESULTS: We found considerable differences in oxysterols profiling in the two disease groups that were gender-related. Compared to controls, males showed significant differences only in 4α- and 4β-hydroxycholesterol levels in obese and metabolic syndrome patients. In contrast, females showed consistent differences in 7-oxocholesterol, 4α-hydroxycholesterol, 25-hydroxycholesterol and triol. Concerning fatty acids, we found minor differences in the levels of these variables in males of the three groups. Significant changes were observed in plasma fatty acid profile of female patients with obesity or metabolic syndrome. We found significant correlations between various oxysterols and fatty acids. In particular, 4β-hydroxycholesterol, which is reduced in obesity and metabolic syndrome, correlated with a number of saturated and mono-unsaturated fatty acids that are end-products of de novo lipogenesis. CONCLUSIONS: Our data provide the first evidence that obesity and metabolic syndrome are associated with major, gender-specific, changes in circulating oxysterols and fatty acids. These findings suggest a metabolic link between oxysterols and fatty acids, and that oxysterols may contribute to the epidemic diseases associated with obesity and metabolic syndrome in female

    Quantifying the relationship between mechanical loading and the skeletal response in pre- and early-pubertal girls

    PURPOSEBefore exercise prescription for bone health can be recommended, the relationship between mechanical loading characteristics and the skeletal response need to be quantified. We asked i) does moderate impact exercise result in a greater gain in BMC than low impact exercise, ii) what are the loading characteristics associated with a moderate and low impact exercise program and does this differ from non-structured play?, and iii) does loading history affect the osteogenic response to a moderate or low impact program? METHODSSixty-eight pre- and early-pubertal girls (aged 8.9 +/- 0.2 yrs) were randomized to take part in a moderate or low impact exercise program 3 times/wk for 8.5 mnths. The number and type of loads associated with the exercise classes and non-structured play (recess) were assessed from video footage. The magnitude of load was assessed using a pedar in-sole mobile system. Hours of moderate and high impact organized sport were assessed from a physical activity questionnaire. RESULTSThe moderate and low impact exercise programs consisted of -400 impacts per class, but the jumping, hopping and dynamic activities performed during the moderate impact program produced forces ranging from 2 to 4 times body weight (BW) compared to -1 BW for the low impact program. Moderate impact exercise resulted in a 2.7% greater gain in BMC at the tibia compared to the low impact exercise. The moderate impact exercise program consisted of fewer low impacts (1-2 BW) and a higher number of moderate impacts (2-4BW) compared to those typically performed during non-structured play. There were greater gains in BMC in subjects participating in the moderate versus the low impact exercise programs who participated in 2 to 3 hours of moderate impact sports outside school (2.5% to 4.5%, p CONCLUSIONApproximately 400 impacts ranging 2-4 BW, 3 times/wk was enough stimuli to result in an osteogenic response in normally active girls; even in those actively involved in moderate impact sports outside school.<br /

    Endothelial cells, endoplasmic reticulum stress and oxysterols

    Oxysterols are bioactive lipids that act as regulators of lipid metabolism, inflammation, cell viability and are involved in several diseases, including atherosclerosis. Mounting evidence linked the atherosclerosis to endothelium dysfunction; in fact, the endothelium regulates the vascular system with roles in processes such as hemostasis, cell cholesterol, hormone trafficking, signal transduction and inflammation. Several papers shed light the ability of oxysterols to induce apoptosis in different cell lines including endothelial cells. Apoptotic endothelial cell and endothelial denudation may constitute a critical step in the transition to plaque erosion and vessel thrombosis, so preventing the endothelial damaged has garnered considerable attention as a novel means of treating atherosclerosis. Endoplasmic reticulum (ER) is the site where the proteins are synthetized and folded and is necessary for most cellular activity; perturbations of ER homeostasis leads to a condition known as endoplasmic reticulum stress. This condition evokes the unfolded protein response (UPR) an adaptive pathway that aims to restore ER homeostasis. Mounting evidence suggests that chronic activation of UPR leads to cell dysfunction and death and recently has been implicated in pathogenesis of endothelial dysfunction. Autophagy is an essential catabolic mechanism that delivers misfolded proteins and damaged organelles to the lysosome for degradation, maintaining basal levels of autophagic activity it is critical for cell survival. Several evidence suggests that persistent ER stress often results in stimulation of autophagic activities, likely as a compensatory mechanism to relieve ER stress and consequently cell death.     Gut Microbiota in Celiac Disease: Is There Any Role for Probiotics?

    Celiac disease (CD) is an immune-mediated disorder initiated by the ingestion of gluten in genetically predisposed individuals. Recent data shows that changes in the gut microbiome composition and function are linked with chronic inflammatory diseases; this might also be the case for CD. The main aim of this manuscript is to discuss our present knowledge of the relationships between gut microbiota alterations and CD and to understand if there is any role for probiotics in CD therapy. PubMed was used to search for all of the studies published from November 2009 to November 2019 using key words such as “Celiac Disease” and “Microbiota” (306 articles), “Celiac Disease” and “Gastrointestinal Microbiome” (139), and “Probiotics” and “Celiac Disease” (97 articles). The search was limited to articles published in English that provided evidence-based data. Literature analysis showed that the gut microbiota has a well-established role in gluten metabolism, in modulating the immune response and in regulating the permeability of the intestinal barrier. Promising studies suggest a possible role of probiotics in treating and/or preventing CD. Nevertheless, human trials on the subject are still scarce and lack homogeneity. A possible role was documented for probiotics in improving CD-related symptoms, modulating the peripheral immune response and altering the fecal microbiota, although the results were not consistent in all of the studies. No evidence was found that probiotic administration might prevent CD onset. Knowledge of the role of intestinal bacteria in the development of CD opens new possibilities for its treatment through probiotic administration, even though further studies are needed to better clarify whether probiotics can help treat or prevent the disease and to define which probiotics to use, at what dose and for how long

    Analisi del campo di deformazione superficiale della Regione Campania da dati SAR (PS-InSAR)

    Il lavoro presenta un’applicazione della tecnica dei diffusori permanenti (Permanent Scatterers) finalizzata alla caratterizzazione del campo di deformazione superficiale della Regione Campania. Per effettuare l’analisi sono state processate, tramite la tecnica PS-InSARTM, 1078 scene SAR ERS1 ed ERS2 relative al periodo giugno 1992 - gennaio 2001. Sulla base della velocità media annua di spostamento di 1.691.740 PS con coerenza maggiore di 0.65 sono state prodotte mappe di velocità media di deformazione mediante l’utilizzo di una procedura articolata nei seguenti punti: a) filtraggio dei dati effettuato sulla base dei valori di coerenza; b) produzione di mappe di velocità media di deformazione in coordinate SAR; c) generazione di mappe di velocità media di deformazione secondo le componenti verticale ed Est-Ovest; d) validazione di risultati mediante confronto con dati prodotti con differenti metodiche d’indagine. L’analisi dei risultati ottenuti ha consentito di identificare aree caratterizzate da distinti stili deformativi. Aree in subsidenza sono risultate essere, per il periodo di interesse, oltre all’area vulcanica dei Campi Flegrei, le aree costiere dei bacini dei fiumi Volturno e Sele ed il settore centrale dell’isola d’Ischia. Aree con significativi valori di sollevamento sono localizzate nel settore settentrionale del Monte Somma, nel settore centrale della Piana Campana, nel Sannio e nel Cilento. Significativi valori di velocità di spostamento in direzione EW si riscontrano, oltre che nell’area dei Campi Flegrei, nel settore appenninico del bacino del Sele e nell’area interposta tra il Somma-Vesuvio e la catena appenninica. Il quadro deformativo globale risulta essere caratterizzato da un andamento complesso, governato da una molteplicità di processi di differente natura (tettonici, vulcano-tettonici, esogeni ed antropici)

    Up-regulation of prostaglandin biosynthesis by leukotriene C4 in elicited mice peritoneal macrophages activated with lipopolysaccharide/interferon-gamma

    Leukotrienes (LT) and prostaglandins (PG) are proinflammatory mediators generated by the conversion of arachidonic acid via 5-lipoxygenase (5-LO) and cyclooxygenase (COX) pathways. It has long been proposed that the inhibition of the 5-LO could enhance the COX pathway leading to an increased PG generation. We have found that in in vitro models of inflammation, such as mice-elicited peritoneal macrophages activated with lipopolysaccharide (LPS)/interferon- Îł (IFN-Îł), the deletion of the gene encoding for 5-LO or the enzyme activity inhibition corresponded to a negative modulation of the COX pathway. Moreover, exogenously added LTC4, but not LTD4, LTE 4, and LTB4, was able to increase PG production in stimulated cells from 5-LO wild-type and knockout mice. LTC4 was not able to induce COX-2 expression by itself but rather potentiated the action of LPS/IFN-Îł through the extracellular signal-regulated kinase-1/2 activation, as demonstrated by the use of a specific mitogen-activated protein kinase (MAPK) kinase inhibitor. The LT-induced increase in PG generation, as well as MAPK activation, was dependent by a specific ligand-receptor interaction, as demonstrated by the use of a cys-LT1 receptor antagonist, although also a direct action of the antagonist used, on PG generation, cannot be excluded. Thus, the balance between COX and 5-LO metabolites could be of great importance in controlling macrophage functions and consequently, inflammation and tumor promotion

    Experimental testing of 3D printed polymeric heat exchangers

    Unlike conventional manufacturing technologies, additive manufacturing and 3D printing empower engineers with much more design freedom. Heat exchangers with complex internal channels or lattice structures can be designed for layerwise manufacturing by maximizing the surface to volume ratio. Low-weight polymeric heat exchangers are employed in aviation and aerospace applications. For increasing the thermal performance of polymers, additives can be used such as graphene. In this study, a Grafylon filament is used for the production of a simple heat exchanger by 3D printing. The heat exchanger is composed of two external shells and an interior duct with a two-stage 45-degree bend. For watertight purposes, the duct is manufactured by selective laser sintering of polyamide powder. Two replicas of the shells are fabricated by 3D printing of Grafylon and acrylonitrile butadiene styrene (ABS) respectively. The thermal performance of the two materials is experimentally tested and compared also to numerical simulations. The results of the study show that the Grafylon filament provides enhanced thermal performance to 3D printed heat exchangers of polymeric material
