    Different approaches for dealing with rare variants in family-based genetic studies: application of a Genetic Analysis Workshop 17 problem

    Rare variants are becoming the new candidates in the search for genetic variants that predispose individuals to a phenotype of interest. Their low prevalence in a population requires the development of dedicated detection and analytical methods. A family-based approach could greatly enhance their detection and interpretation because rare variants are nearly family specific. In this report, we test several distinct approaches for analyzing the information provided by rare and common variants and how they can be effectively used to pinpoint putative candidate genes for follow-up studies. The analyses were performed on the mini-exome data set provided by Genetic Analysis Workshop 17. Eight approaches were tested, four using the trait’s heritability estimates and four using QTDT models. These methods had their sensitivity, specificity, and positive and negative predictive values compared in light of the simulation parameters. Our results highlight important limitations of current methods to deal with rare and common variants, all methods presented a reduced specificity and, consequently, prone to false positive associations. Methods analyzing common variants information showed an enhanced sensibility when compared to rare variants methods. Furthermore, our limited knowledge of the use of biological databases for gene annotations, possibly for use as covariates in regression models, imposes a barrier to further research

    Este trabalho problematiza, a partir de uma perspectiva sócio-histórica, formas de enfrentamento à violência a que estão submetidos adolescentes e jovens de grupos populares urbanos no Brasil. Considera-se a violência como um fenômeno complexo de grande relevância para diversas instâncias sociais. A vulnerabilidade daqueles adolescentes e jovens, expressa por inúmeros índices relacionados à violência, tem alcançado patamares alarmantes no nosso país, num contexto de políticas públicas que são, em grande parte, insuficientes, fragmentadas e/ou inadequadas. Assim, apresenta-se o relato e a análise de uma intervenção social calcada na educação e na defesa dos direitos decorrentes da cidadania, para adolescentes e jovens vulneráveis socialmente, tomando-se como referência uma região composta por bairros pobres e carentes de infra-estrutura social numa cidade média do interior do estado de São Paulo. Trabalhou-se em diferentes projetos com ações pautadas na abordagem interdisciplinar, por meio de três eixos bases: Violência Escolar; Violência Urbana; e Violação de Direitos e Comunidade. Os resultados alcançados refletem elaborações coletivas acerca das ações destinadas aos jovens de grupos populares urbanos e suas alternativas, buscando produzir estratégias de enfrentamento dessas questões em espaços públicos, desde a instituição escolar historicamente constituída para essa população, assim como as instituições mais recentes que respondem à lógica contraditória e complexa da assistência ao direito. Cria-se, portanto, subsídios para políticas públicas cujo impacto se dê na direção de promover a diminuição da desigualdade, da discriminação e da violência a que está sujeita a maioria dos adolescentes e jovens no Brasil.<br>This article searches, from a social historical point of view, for ways of facing the violence that afflicts teenagers and youngsters of urban popular groups in Brazil. Violence is a complex phenomenon of great importance in many social substrates. The vulnerability of those teenagers and youngsters, as shown by a plurality of indexes, has reached unacceptable highs in Brazil, where the public policies, so far, have proven to be inadequate, insufficient and fragmented. Thus, the article reports and analyzes a social intervention based on education and on the defense of human rights deriving from citizenship, targeted on socially vulnerable teenagers and youngsters, specifically those living in a deprived region lacking basic social infrastructure located in a medium-sized town in the state of São Paulo. Different projects dealt with actions based on an interdisciplinary approach, according to the following aspects: a) School violence; b) Urban violence; c) Human rights violation and violence in the community. The results showed a collective frame related to actions concerning teenagers and youngsters of urban popular groups and their alternatives. Strategies for dealing with these matters in public spaces are formulated, from regular schools to more recent institutions concerning the contradictory logic of social rights. Therefore, subsidies are provided for public policies in order to reduce the differences, discrimination and violence that afflict teenagers and youngsters in Brazil
