    The ecology and evolution of female-specific ornamentation in the dance flies (Diptera: Empidinae)

    Elaborate morphological ornaments can evolve if they increase the reproductive success of the bearer during competition for mates. However, ornament evolution is incredibly rare in females, and the type and intensity of selection required to develop female-specific ornamentation is poorly understood. The main goals of my thesis are to clarify the relationship between the type and intensity of sexual selection that drives the evolution of female ornamentation, and investigate alternative hypotheses that might be limiting or contributing to the development of female ornaments. I investigated the ecology and evolution of female-specific ornaments within and between species of dance flies from the subfamily Empidinae (Diptera: Empididae). The dance flies display incredible mating system diversity including those with elaborate female-specific ornaments, lek-like mating swarms, aerial copulation and nuptial gift giving. To elucidate the form of sexual selection involved in female-ornament evolution, I experimentally investigated the role of sexual conflict in the evolution of multiple female- specific ornaments in the species Rhamphomyia longicauda. Through manipulative field experiments, I found that variation in the attractiveness of two ornaments displayed by females indicates that sexual conflict, causing a coevolutionary arms race, is an important force in the evolution of multiple extravagant female ornaments. Using R. longicauda again, I tested for a role of functional load-lifting constraints on the aerial mating ability of males who paired with females displaying multiple large ornaments. I found no evidence of functional constraints influencing the mating opportunities of elaborately ornate females, but instead discovered a relationship consistent with positive assortative mating for mass. Biased sex ratios are predicted to increase the intensity of sexual selection in a population, which in turn, is predicted to influence the evolution of ornamentation. I measured the incidence and prevalence of vertically transmitted symbiotic bacteria that has been observed to distort the sex ratio in other Dipteran hosts. While my survey revealed that symbionts occur at high incidence and variable prevalence across dance fly hosts, I found no effect of symbiont infection levels on population sex ratios, or female- specific ornament evolution. Further investigation into the relationship between sex ratios and female-ornament evolution using the comparative method revealed that the operational sex ratio (OSR) of a population did not predict continuous measures of female ornamentation across species. However, female-ornament evolution did predict male relative testis investment across species indicating that female ornaments likely indicate increased levels of polyandry. My thesis reveals that sexual selection theory developed to describe male-specific ornament evolution cannot readily be translated to apply to females. I show that male mate choice, rather than functional constraints or ecological associations with bacteria, is likely driving the evolution of female-specific ornaments. I also identify sexual conflict as an important selective force in the evolution of female-specific ornaments

    Are sex ratio distorting endosymbionts responsible for mating system variation among dance flies (Diptera: Empidinae)?

    Maternally inherited bacterial endosymbionts are common in many arthropod species. Some endosymbionts cause female-biased sex ratio distortion in their hosts that can result in profound changes to a host's mating behaviour and reproductive biology. Dance flies (Diptera: Empidinae) are well known for their unusual reproductive biology, including species with female-specific ornamentation and female-biased lek-like swarming behaviour. The cause of the repeated evolution of female ornaments in these flies remains unknown, but is probably associated with female-biased sex ratios in individual species. In this study we assessed whether dance flies harbour sex ratio distorting endosymbionts that might have driven these mating system evolutionary changes. We measured the incidence and prevalence of infection by three endosymbionts that are known to cause female-biased sex ratios in other insect hosts (Wolbachia, Rickettsia and Spiroplasma) across 20 species of dance flies. We found evidence of widespread infection by all three symbionts and variation in sex-specific prevalence across the taxa sampled. However, there was no relationship between infection prevalence and adult sex ratio measures and no evidence that female ornaments are associated with high prevalences of sex-biased symbiont infections. We conclude that the current distribution of endosymbiont infections is unlikely to explain the diversity in mating systems among dance fly species. ©2017 Murray et al.     Sexual selection on multiple female ornaments in dance flies

    Sex-specific ornaments typically occur in males, but they can also develop in females. While there are several models concerning the evolution of male-specific ornaments, it is not clear how, or under what circumstances, those models apply to female-specific ornament evolution. Here, we present a manipulative field experiment that explores the theoretical ā€˜trait spaceā€™ of multiple female-specific ornaments to study how these unusual traits evolved. We measured the attractiveness of two female-specific ornaments (pinnate leg scales and inflatable abdominal sacs) in the dance fly Rhamphomyia longicauda in a wild mating swarm. We found significant directional preferences for larger ornaments of both types; however, variation in one of the ornaments (abdominal sacs) was almost three times more effective at improving attractiveness. The abdominal ornament was consistently effective in increasing attractiveness to males regardless of leg ornament expression, while leg ornament size was only effective if abdominal ornaments were very small. These results are consistent with predictions from a sexual conflict model of ornament expression in supporting the probable role of deception in the evolution of female-specific ornaments among dance flies. Sexual conflict can be an important force in generating elaborate sex-specific ornaments in females as well as males

    The role of functional constraints in nonrandom mating patterns for a dance fly with female ornaments

    Most hypotheses to explain nonā€random mating patterns invoke mate choice, particularly in species that display elaborate ornaments. However, conflicting selection pressures on traits can result in functional constraints that can also cause nonā€random mating patterns. We tested for functional loadā€lifting constraints during aerial copulation in Rhamphomyia longicauda, a species of dance fly that displays multiple extravagant femaleā€specific ornaments that are unusual among sexual traits because they are under stabilizing selection. R. longicauda males provide females with a nuptial gift before engaging in aerial mating, and the male bears the entire weight of the female and nuptial gift for the duration of copulation. In theory, a male's ability to carry females and nuptial gifts could constrain pairing opportunities for the heaviest females, as reported for nonā€ornamented dance flies. In concert with directional preferences for large females with mature eggs, such a loadā€lifting constraint could produce the stabilizing selection on female size previously observed in this species. We therefore tested whether wildā€caught male R. longicauda collected during copulation were experiencing loadā€lift limitations by comparing the mass carried by males during copulation with the male's wing loading traits. We also performed permutation tests to determine whether the loads carried by males during copulation were lighter than expected. We found that heavier males are more often found mating with heavier females suggesting that while R. longicauda males do not experience a loadā€lift constraint, there is a strong relationship of assortative mating by mass. We suggest that active male mate choice for intermediately adorned females is more likely to be causing the nonā€random mating patterns observed in R. longicauda

    A Proline-Rich Structural Protein of the Surface Sheath of Larval Brugia Filarial Nematode Parasites

    Both cDNA and genomic DNA sequences have been isolated which encode a proline-rich precursor protein of the sheath from microfilariae, the first stage larvae of the filarial nematode parasites Brugia pahangi andBrugia malayi. This 22-kDa protein is soluble only under reducing conditions and is extensively cross- linked by both disulfide and nonreducible bonds. Immunogold electron microscopy shows that the protein is localized exclusively in the sheath, a vestigial remnant of the eggshell, which is retained by and encloses the mature microfilaria. Analysis by Western blotting confirms that the protein is expressed only in microfilariae and adult female worms, although transcripts are detectable only in adult females. The deduced amino acid sequence contains a short N-terminal hydrophobic putative leader sequence, a central repetitive domain that contains 14 copies of a degenerate 5-amino acid repeat with the consensus sequence Met- Pro-Pro-Gln-Gly, and a C-terminal prolinerich do- main flanked by clusters of cysteine residues. These clusters can be aligned with cysteine residues implicated in cross-linking of a family of cuticular collagens originally identified in Caenorhabditiselegans but which extends to other nematodes

    Competition for access to mates predicts female-specific ornamentation and male investment in relative testis size

    Sexually selected ornaments are highly variable and the factors that drive variation in ornament expression are not always clear. Rare instances of femaleā€specific ornament evolution (such as in some dance fly species) are particularly puzzling. While some evidence suggests that such rare instances represent straightforward reversals of sexual selection intensity, the distinct nature of tradeā€offs between ornaments and offspring pose special constraints in females. To examine whether competition for access to mates generally favours heightened ornament expression, we built a phylogeny and conducted a comparative analysis of Empidinae dance fly taxa that display femaleā€specific ornaments. We show that species with more femaleā€biased operational sex ratios in lekā€like mating swarms have greater female ornamentation, and in taxa with more ornate females, male relative testis investment is increased. These findings support the hypothesis that ornament diversity in dance flies depends on female receptivity to mates, which is associated with contests for nutritious nuptial gifts provided by males. Moreover, our results suggest that increases in female receptivity lead to higher levels of sperm competition among males. The incidence of both heightened preā€mating sexual selection on females and postā€mating selection on males contradicts assertions that sexā€roles are straightforwardly reversed in dance flies

    Prioritising Infectious Disease Mapping.

    BACKGROUND: Increasing volumes of data and computational capacity afford unprecedented opportunities to scale up infectious disease (ID) mapping for public health uses. Whilst a large number of IDs show global spatial variation, comprehensive knowledge of these geographic patterns is poor. Here we use an objective method to prioritise mapping efforts to begin to address the large deficit in global disease maps currently available. METHODOLOGY/PRINCIPAL FINDINGS: Automation of ID mapping requires bespoke methodological adjustments tailored to the epidemiological characteristics of different types of diseases. Diseases were therefore grouped into 33 clusters based upon taxonomic divisions and shared epidemiological characteristics. Disability-adjusted life years, derived from the Global Burden of Disease 2013 study, were used as a globally consistent metric of disease burden. A review of global health stakeholders, existing literature and national health priorities was undertaken to assess relative interest in the diseases. The clusters were ranked by combining both metrics, which identified 44 diseases of main concern within 15 principle clusters. Whilst malaria, HIV and tuberculosis were the highest priority due to their considerable burden, the high priority clusters were dominated by neglected tropical diseases and vector-borne parasites. CONCLUSIONS/SIGNIFICANCE: A quantitative, easily-updated and flexible framework for prioritising diseases is presented here. The study identifies a possible future strategy for those diseases where significant knowledge gaps remain, as well as recognising those where global mapping programs have already made significant progress. For many conditions, potential shared epidemiological information has yet to be exploited

    Vitamin B 12 and Folate status of older New Zealand women

    The primary aim of this study was to assess the biochemical vitamin B 12 and folate status of a representative group of elderly women (70-80 y) living in Dunedin, New Zealand. A second aim was to determine the prevalence of hyperhomocysteinaemia and to explore the determinants of homocysteine (hcy) concentration in this population. A cross-sectional study was carried out between June and August of 2000. Two hundred and fifty women were randomly selected from the 1998 electoral roll. Fasting blood samples were analysed for folate, vitamin B 12 , total hcy, creatinine, and haematological parameters. Of the women selected, 87 did not respond, 37 were not traceable, 23 were not eligible or had died, and 103 agreed to participate. The overall response rate was 46%. Based on a cut-off of 150 pmol/L for serum B 12 , 13 % of participants would be classified as having sub-optimal vitamin B 12 status. Of the women, 3 and 5 %, respectively, had low serum (<6.6 nmol/L) and erythrocyte folate (<317 nmol/L) concentrations. No participant had megaloblastic anaemia. The prevalence of hyperhomocysteinaemia (>15 Āµmol/L) in this population was 18%. Hyperhomocysteinaemia in this group may be partly explained by renal insufficiency because there was a significant association between serum creatinine and plasma hcy (P<0.001). Blood folate levels but not serum B 12 were significantly inversely associated with hcy. In conclusion, there was a moderately high prevalence of hyperhomocysteinaemia and suboptimal plasma vitamin B 12 concentrations but not low blood folate concentrations in this elderly female population

    Multiple and Multidimensional life transitions in the context of life-limiting health conditions:Longitudinal study focussing on perspectives of Young Adults, Families and Professionals

    Background: There is a dearth of literature that investigates life transitions of young adults (YAs) with life-limiting conditions, families and professionals. The scant literature that is available has methodological limitations, including not listening to the voice of YAs, collecting data retrospectively, at one time point, from one groupā€™s perspective and single case studies. The aim of this study was to address the gaps found in our literature review and provide a clearer understanding of the multiple and multi-dimensional life transitions experienced by YAs and significant others, over a period of time. Methods: This qualitative study used a longitudinal design and data were collected using semi-structured interviews over a 6-month period at 3 time points. Participants included 12 YAs with life-limiting conditions and their nominated significant others (10 family members and 11 professionals). Data were analysed using a thematic analysis approach. Results: Life transitions of YA and significant others are complex; they experience multiple and multi-dimensional transitions across several domains. The findings challenge the notion that all life transitions are triggered by health transitions of YAs, and has highlighted environmental factors (attitudinal and systemic) that can be changed to facilitate smoother transitions in various aspects of their lives. Conclusions: This study makes a unique and significant contribution to literature. It provides evidence and rich narratives for policy makers and service providers to change policies and practices that are in line with the needs of YAs with life-limiting conditions as they transition to adulthood. Families and professionals have specific training needs that have not yet been met fully
