Topological jamming of spontaneously knotted polyelectrolyte chains driven through a nanopore
The advent of solid state nanodevices allows for interrogating the
physico-chemical properties of a polyelectrolyte chain by electrophoretically
driving it through a nanopore. Salient dynamical aspects of the translocation
process have been recently characterized by theoretical and computational
studies of model polymer chains free from self-entanglement. However,
sufficiently long equilibrated chains are necessarily knotted. The impact of
such topological "defects" on the translocation process is largely unexplored,
and is addressed in this study. By using Brownian dynamics simulations on a
coarse-grained polyelectrolyte model we show that knots, despite being trapped
at the pore entrance, do not "per se" cause the translocation process to jam.
Rather, knots introduce an effective friction that increases with the applied
force, and practically halts the translocation above a threshold force. The
predicted dynamical crossover, which is experimentally verifiable, is of
relevance in applicative contexts, such as DNA nanopore sequencing.Comment: 6 pages; 7 figure
Housing system and welfare of buffalo (Bubalus bubalis) cows
Twenty-eight buffalo cows were used to evaluate the effect of housing system on a range of behavioural and physiological variables. Fourteen cows were group-housed in a loose open-sided barn with a concrete floor and 10 m2 per head as space allowance (group IS). Fourteen others were group-housed in a similar barn but they could also benefit from an outdoor yard with 500 m2 per head as space allowance, free access to potholes for wallowing and spontaneous vegetation (group TS). Animals were subjected to six sessions of instantaneous scan sampling at 10-day intervals. Behavioural variables were expressed as proportions of subjects observed in each category of posture and activity. Phytohaemagglutinin (PHA) was used to perform a skin test based on non-specific delayed type hypersensitivity, whereas 20 mg of ovalbumin were injected subcutaneously to evaluate humoral immune response. Blood samples for evaluation of cortisol concentration were collected immediately prior to exogenous porcine ACTH injection and 1, 2 and 4 h after. The metabolic status of the animals and milk production were also monitored. The proportion of idling animals was higher in group IS than in group TS (P< 0·001). More IS buffalo cows were observed eating at the manger than TS animals (P< 0·001). A higher proportion of TS animals were observed in the sun (P< 0·001). Grazing and bathing activities were recorded only for TS animals. Our findings suggest that buffalo cows kept in intensive conditions and having no access to ample yards and potholes may extend their periods of idling with negative effects on the state of welfare. Immune responses, metabolite concentrations and milk production were not affected by treatment, whereas cortisol levels were higher in IS animals (P< 0·05). The provision of a housing system similar to natural conditions was able to improve the welfare of buffalo cows as indicated by the expression of some species-specific natural behaviours. Such conditions were also associated with lower adrenal cortex response to ACTH injection, possibly as a consequence of the higher degree of initiative allowed to TS cows
Analysis of unmitigated large break loss of coolant accidents using MELCOR code
In the framework of severe accident research activity developed by ENEA, a MELCOR nodalization of a generic Pressurized Water Reactor of 900 MWe has been developed. The aim of this paper is to present the analysis of MELCOR code calculations concerning two independent unmitigated large break loss of coolant accident transients, occurring in the cited type of reactor. In particular, the analysis and comparison between the transients initiated by an unmitigated double-ended cold leg rupture and an unmitigated double-ended hot leg rupture in the loop 1 of the primary cooling system is presented herein. This activity has been performed focusing specifically on the in-vessel phenomenology that characterizes this kind of accidents. The analysis of the thermal-hydraulic transient phenomena and the core degradation phenomena is therefore here presented. The analysis of the calculated data shows the capability of the code to reproduce the phenomena typical of these transients and permits their phenomenological study. A first sequence of main events is here presented and shows that the cold leg break transient results faster than the hot leg break transient because of the position of the break. Further analyses are in progress to quantitatively assess the results of the code nodalization for accident management strategy definition and fission product source term evaluation
Just Chill! An experimental approach to stereotypical attributions regarding young activists
Climate change is a crucial issue, which is mobilized by activists. However, activists are targeted with negative stereotypes, hindering their influence. Young activists are environmentally conscious, but the stereotypical attributions assigned to them are unknown, with competing predictions in the literature (for being activist vs being young). In two studies, we aimed at experimentally examining the stereotypical dimensions that are ascribed to activists (youth vs adult) based on the Stereotype Content Model (SCM), as well as a morality/trustworthiness dimension. Considering that activists are generally considered as high-competent, but low-warm, while youth are considered the opposite (low-competence and high-warmth), we hypothesized the impacts on morality/trustworthiness. Greta Thunberg and Jane Fonda were the personalities used in Study 1 (N = 276), randomly assigned to participants while keeping the same discourse excerpt. Thunberg was penalized in all stereotypical dimensions. In Study 2 (N = 228), fictional characters (teenager or adult) were used instead. As hypothesized, no differences were found in the warmth or competence dimensions, only in the morality/trustworthiness dimension, penalizing the young activist. These results highlight the importance of studying environmental activists considering different social categories in stereotypical appraisals. They also contribute to a better understanding of general resistance towards activists, as well as the factors that are detrimental to their social influence.info:eu-repo/semantics/publishedVersio
