    Recent advances in the application of pulsed light processing for improving food safety and increasing shelf life

    peer-reviewedBackground New technologies of non-thermal disinfection such as pulsed light (PL) have emerged lately as an alternative to traditional (thermal and chemical) disinfection and preservation methods. PL can be used to decontaminate a great variety of foods as well as to decontaminate contact surfaces, thus improving safety in foods and extending their shelf life. Moreover, this technology can prevent or reduce some of the detrimental effects of traditional methods on nutrients and bioactive compounds of food products. Scope and approach The combination of PL with other techniques such as ultraviolet light (UV), thermosonication (TS), pulsed electric fields (PEF), manothermosonication (MTS), etc., can improve the effectiveness of the decontamination process. Therefore, in this review, some of the most relevant studies evaluating the potential application of PL treatments to decontaminate food samples, and its impact of nutritional and physicochemical quality parameters will be discussed. Key findings and conclusions PL treatments are suitable for microbial decontamination in transparent drinks and for surface contaminated foods without complex microstructures. They also can be used for meat, fish and their by-products However, it is still necessary to evaluate the appropriate treatment conditions (number of light flashed, voltage, distance between sample and flash light, spectral range of light flashes and contamination) for each food and microorganism separately to improve the effectiveness and minimize the appearance of negative attributes reducing the quality of product as, in some cases, PL can have a negative effect on the photosensitive compounds and sensory characteristics of food products

    Improving the lipid profile in hypercholesterolemia-induced rabbit by supplementation of germinated brown rice.

    It is imperative that there be a diet designed specifically to improve lipid profile in order to impede the progress of atherosclerosis. Because rice is a staple food in Asia, it will be chosen as the diet of interest. This study sets out to discover whether consumption of different processed rice diets may result in a change of the lipid profile. The experiment was done on male New Zealand white rabbits after 10 weeks of treatment with diet containing 0.5% cholesterol. The experimental diets include white rice (WR), brown rice (BR), and germinated brown rice (GBR). Among them, rabbits fed a GBR diet demonstrated significantly lower levels of total cholesterol (TC), low-density lipoprotein (LDL), LDL/HDL, and atherogenic index (AI) and a higher level of high-density lipoprotein (HDL). Results from atherosclerotic plaque assessment further support the findings. The level of malondialdehyde (MDA), which acts as an indicator for oxidative stress, was also reduced by GBR diet. The positive change in lipid profile in the rabbits fed GBR appeared to correspond with the higher amounts of γ-oryzanol, tocopherol, and monounsaturated fatty acid (MUFA) content

    Evaluation of commercial soy sauce koji strains of Aspergillus oryzae for γ-aminobutyric acid (GABA) production

    In this study, four selected commercial strains of Aspergillus oryzae were collected from soy sauce koji. These A. oryzae strains designated as NSK, NSZ, NSJ and NST shared similar morphological characteristics with the reference strain (A. oryzae FRR 1675) which confirmed them as A. oryzae species. They were further evaluated for their ability to produce γ-aminobutyric acid (GABA) by cultivating the spore suspension in a broth medium containing 0.4 % (w/v) of glutamic acid as a substrate for GABA production. The results showed that these strains were capable of producing GABA; however, the concentrations differed significantly (P < 0.05) among themselves. Based on the A. oryzae strains, highest GABA concentration was obtained from NSK (194 mg/L) followed by NSZ (63 mg/L), NSJ (51.53 mg/L) and NST (31.66 mg/L). Therefore, A. oryzae NSK was characterized and the sequence was found to be similar to A. oryzae and A. flavus with 99 % similarity. The evolutionary distance (K nuc) between sequences of identical fungal species was calculated and a phylogenetic tree prepared from the K nuc data showed that the isolate belonged to the A. oryzae species. This finding may allow the development of GABA-rich ingredients using A. oryzae NSK as a starter culture for soy sauce production

    Impact of tartrazine and curcumin on mineral status, and thyroid and reproductive hormones disruption in vivo

    Endocrine disrupters (EDCs) are naturally occurring or man-made substances that either mimic or obstruct the functions of oestrogens and androgens, thyroid hormones, as well as microminerals in the body. The present work aimed to evaluate the effects of oral administration of tartrazine and curcumin, synthetic and natural dyes, respectively, on thyroid hormones (T3, T4, and TSH), female reproductive hormones (oestrogen, progesterone, LH, and FSH), and minerals (iron, copper, zinc, sodium, potassium, and chloride) in plasma, liver, and kidney of female rats after 15, 30, and 45 d of treatment. The rats were treated with admissible daily intake (ADI) and 10× ADI (9.6 and 96 mg/kg/body weight for tartrazine, 3.85 and 38.5 mg/kg/body weight for curcumin, respectively). Results showed significant changes in thyroid and female reproductive hormones, especially, in the tartrazine-treated groups as compared to the control. Low and high doses of tartrazine and curcumin significantly (p < 0.05) decreased iron, copper, and zinc concentrations in plasma, whereas, the concentrations of sodium and copper in liver and kidney increased. Both tartrazine and curcumin, at ADI and 10× ADI, resulted in lower LH levels after 30 and 45 d of treatment. After 30 d, low and high dose of tartrazine significantly decreased T4, oestrogen, and FSH levels; whereas, progesterone level increased. The results demonstrated that hormone secretion and mineral content in tissues are severely affected at ADI and higher concentrations of tartrazine and curcumin. These observations suggested that lower doses of these dyes might be a safer option for their usage in foods and pharmaceuticals

    Low expression of LncRNA-CAF attributed to the high expression of HIF1A in esophageal squamous cell carcinoma and gastric cancer patients

    Purpose: Cancer-associated fibroblasts (CAFs) are major components of tumor microenvironment that stimulate ESCC and GC progression. The LncRNA-CAF, FLJ22447, is located in the vicinity of HIF1A, while their association remains unclear. This study aims to assess the FLJ22447 expression in the ESCC and GC patients and evaluate its association with the HIF1A gene. Methods: Fresh ESCC and GC tumor samples and their adjacent non-tumor tissues were collected from patients who underwent surgery in Imam Khomeini Hospital, Tehran, Iran. The expression of FLJ22447, HIF1A, and VEGF was evaluated using qRT-PCR test. The association of their expression with tumor clinicopathological features in ESCC patients was assessed. System biology tools were then applied for the possible biological subsequences of the FLJ22447. Results: A significant reduction in FLJ22447 expression was observed in ESCC and GC tissues than adjacent non-tumor tissues, while, the expression of HIF1A and VEGF were increased. Low expression of FLJ22447 was significantly correlated with HIF1A (P = 2.4e�73, R = 0.63) and VEGF (P = 0.00019, R = 0.15) expression. A significant relationship was detected between the high expression of HIF1A and tumor stages (I�II) and it was related to the reduced survival of ESCC patients. Conversely, increased VEGF expression was linked to the advanced stages (III�IV) and metastasis in ESCC. The analysis of FLJ22447-interacted proteins showed that MYC, JUN, SMRCA4, PPARG, AR, FOS, and CEBPA are the hub genes. These proteins were implicated in the cancer related pathways. Among them, SPI1, E2F1, TCF7L2, and STAT1 were significantly expressed in esophageal and gastric cancers that were functionally involved in the proliferation, apoptosis, and angiogenesis pathways in cancer. Conclusion: The results suggested that FLJ22447 may have a regulatory function on the HIF1A expression. We identified the FLJ22447-interacted proteins and their molecular function in cancer pathogenesis. Further research emphasis is to realize the association of FLJ22447 with its protein partners in progression of cancer. These may provide an insight into the FLJ22447 activity that could introduce it as a potential value in tumor gene therapy. © 2021, The Author(s), under exclusive licence to Springer Nature B.V
