    Comparative virulence of Salmonella cholerae-suis var kunzendorf strains

    Seventy-two field strains of smooth Salmonella cholerae-suis var kunzendorf were examined for susceptibility to killing by antibody-complement (Ab-C) after they were exposed to Tris-EDTA, lysozyme, or saline solution. Considerable differences were found among the strains with some being susceptible to killing under all conditions and some being competely resistant. Evidence was obtained to indicate that Tris-EDTA may inhibit the growth of S. cholerae-suis in addition to its documented effect on the release of lipopolysaccharide from the cell wall of gram-negative bacteria. Twenty-two of these strains were further evaluated for their susceptibility to killing by porcine peripheral blood polymorphonuclear neutrophils (PMNs) under various conditions. Susceptibility to PMN killing was found to correlate well with the Ab-C susceptibility of these strains. The mouse virulence of these strains was examined. Eight of the strains were selected for further evaluation in the pig. With one exception, the virulence of these strains for pigs was well predicted by the results of the in vitro Ab-C bacteriolysis. Mouse virulence was also found to correlate with pig virulence but was somewhat less accurate. The immune response of the pig to one of the strains of S. cholerae-suis was evaluated with and without the presence of the immunosuppressive drug cyclophosphamide. In addition the effects of CY on the delayed hypersensitivity (DH) reaction to Mycobacterium avium were examined. For this, pigs were sensitized with M. avium sensitinogen. Pigs given 20 mg/kg of CY on days 0, 2 and 4 and infected with 10(\u276) S. cholerae-suis on day 0 were more severely affected than those given the same dose of CY and infected on day 4. The DH response in the former group was equivalent to the nonsensitized group and thus was severely depressed. The DH reaction of pigs infected on day 4 was also decreased compared to the sensitized controls. Mortality rates and serological responses were compared for the various groups of pigs. The virulence and immunogenicity of several mutants of S. cholerae-suis were evaluated in mice. An aro(\u27-) galE mutant of strain 38 proved to be reduced in virulence from the parent strain but provided only moderate protection against homologous challenge. Thymidine-requiring mutants of strains 9, 33, 38, 51 and 61 were observed to be avirulent for mice. All provided as much or more protection against challenge than the parent strain

    Motor Axon Synapses on Renshaw Cells Contain Higher Levels of Aspartate than Glutamate

    Motoneuron synapses on spinal cord interneurons known as Renshaw cells activate nicotinic, AMPA and NMDA receptors consistent with co-release of acetylcholine and excitatory amino acids (EAA). However, whether these synapses express vesicular glutamate transporters (VGLUTs) capable of accumulating glutamate into synaptic vesicles is controversial. An alternative possibility is that these synapses release other EAAs, like aspartate, not dependent on VGLUTs. To clarify the exact EAA concentrated at motor axon synapses we performed a quantitative postembedding colloidal gold immunoelectron analysis for aspartate and glutamate on motor axon synapses (identified by immunoreactivity to the vesicular acetylcholine transporter; VAChT) contacting calbindin-immunoreactive (-IR) Renshaw cell dendrites. The results show that 71% to 80% of motor axon synaptic boutons on Renshaw cells contained aspartate immunolabeling two standard deviations above average neuropil labeling. Moreover, VAChT-IR synapses on Renshaw cells contained, on average, aspartate immunolabeling at 2.5 to 2.8 times above the average neuropil level. In contrast, glutamate enrichment was lower; 21% to 44% of VAChT-IR synapses showed glutamate-IR two standard deviations above average neuropil labeling and average glutamate immunogold density was 1.7 to 2.0 times the neuropil level. The results were not influenced by antibody affinities because glutamate antibodies detected glutamate-enriched brain homogenates more efficiently than aspartate antibodies detecting aspartate-enriched brain homogenates. Furthermore, synaptic boutons with ultrastructural features of Type I excitatory synapses were always labeled by glutamate antibodies at higher density than motor axon synapses. We conclude that motor axon synapses co-express aspartate and glutamate, but aspartate is concentrated at higher levels than glutamate

    Emissions from Smoldering Combustion of Biomass Measured by Open-Path Fourier Transform Infrared Spectroscopy

    Biomass samples from a diverse range of ecosystems were burned in the Intermountain Fire Sciences Laboratory open combustion facility. Midinfrared spectra of the nascent emissions were acquired at several heights above the fires with a Fourier transform infrared spectrometer (FTIR) coupled to an open multipass cell. In this report, the results from smoldering combustion during 24 fires are presented including production of carbon dioxide, carbon monoxide, methane, ethene, ethyne, propene, formaldehyde, 2-hydroxyethanal, methanol, phenol, acetic acid, formic acid, ammonia, hydrogen cyanide, and carbonyl sulfide. These were the dominant products observed, and many have significant influence on atmospheric chemistry at the local, regional, and global scale. Included in these results are the first optical, in situ measurements of smoke composition from fires in grasses, hardwoods, and organic soils. About one half of the detected organic emissions arose from fuel pyrolysis which produces white smoke rich in oxygenated organic compounds. These compounds deserve more attention in the assessment of fire impacts on the atmosphere. The compound 2-hydroxyethanal is a significant component of the smoke, and it is reported here for the first time as a product of fires. Most of the observed alkane and ammonia production accompanied visible glowing combustion. NH3 is normally the major nitrogen-containing emission detected from smoldering combustion of biomass, but from some smoldering organic soils, HCN was dominant. Tar condensed on cool surfaces below the fires accounting for ∌1% of the biomass burned, but it was enriched in N by a factor of 6–7 over the parent material, and its possible role in postfire nutrient cycling should be further investigated

    Emissions of Formaldehyde, Acetic Acid, Methanol, and Other Trace Gases from Biomass Fires in North Carolina Measured by Airborne Fourier Transform Infrared Spectroscopy

    Biomass burning is an important source of many trace gases in the global troposphere. We have constructed an airborne trace gas measurement system consisting of a Fourier transform infrared spectrometer (FTIR) coupled to a “flow-through” multipass cell (AFTIR) and installed it on a U.S. Department of Agriculture Forest Service King Air B-90. The first measurements with the new system were conducted in North Carolina during April 1997 on large, isolated biomass fire plumes. Simultaneous measurements included Global Positioning System (GPS); airborne sonde; particle light scattering, CO, and CO2; and integrated filter and canister samples. AFTIR spectra acquired within a few kilometers of the fires yielded excess mixing ratios for 10 of the most common trace gases in the smoke: water, carbon dioxide, carbon monoxide, methane, formaldehyde, acetic acid, formic acid, methanol, ethylene, and ammonia. Emission ratios to carbon monoxide for formaldehyde, acetic acid, and methanol were each 2.5±1%. This is in excellent agreement with (and confirms the relevance of) our results from laboratory fires. However, these ratios are significantly higher than the emission ratios reported for these compounds in some previous studies of “fresh” smoke. We present a simple photochemical model calculation that suggests that oxygenated organic compounds should be included in the assessment of ozone formation in smoke plumes. Our measured emission factors indicate that biomass fires could account for a significant portion of the oxygenated organic compounds and HOx present in the tropical troposphere during the dry season. Our fire measurements, along with recent measurements of oxygenated biogenic emissions and oxygenated organic compounds in the free troposphere, indicate that these rarely measured compounds play a major, but poorly understood, role in the HOx, NOx, and O3 chemistry of the troposphere

    Pharmacokinetics of tulathromycin in pregnant ewes (Ovis aries) challenged with Campylobacter jejuni

    The purpose of this study was to evaluate the pharmacokinetics of tulathromycin in the plasma and maternal and fetal tissues of pregnant ewes when administered within 24 hours of a single, IV Campylobacter jejuni (C. jejuni) challenge. Twelve, pregnant ewes between 72–92 days of gestation were challenged IV with C. jejuni IA3902 and then treated with 1.1 ml/45.36 kg of tulathromycin subcutaneously 18 hours post-challenge. Ewes were bled at predetermined time points and euthanized either at a predetermined time point or following the observation of vaginal bleeding or abortion. Following euthanasia, tissues were collected for bacterial culture, pharmacokinetics and histologic examination. The maximum (geometric) mean tulathromycin plasma concentration was estimated at 0.302 ÎŒg/mL, with a peak level observed at around 1.2 hours. The apparent systemic clearance of tulathromycin was estimated at 16.6 L/h (or 0.28 L/kg/h) with an elimination half-life estimated at approximately 22 hours. The mean tissue concentrations were highest in the uterus (2.464 ÎŒg/g) and placentome (0.484 ÎŒg/g), and were lowest in fetal liver (0.11 ÎŒg/g) and fetal lung (0.03 ÎŒg/g). Compared to previous reports, results of this study demonstrate that prior IV administration of C. jejuni appeared to substantially alter the pharmacokinetics of tulathromycin, reducing both the peak plasma concentrations and elimination half-life. However, additional controlled trials are required to confirm those observations

    Estimation of withdrawal interval recommendations following administration of fenbendazole medicated feed to ring-necked pheasants (Phasianus colchicus)

    IntroductionPrescribing fenbendazole medicated feed for pheasants in the USA is considered extra-label drug use under CPG Sec 615.115, and a safe estimated withdrawal interval (WDI) must be applied following administration to this minor food-producing species. This study sought to determine the pharmacokinetic and residue depletion profile for fenbendazole and its major metabolites to estimate a WDI for pheasants following fenbendazole administration as an oral medicated feed.MethodPheasants (n = 32) were administered fenbendazole as an oral medicated feed (100 ppm) for 7 days. Fenbendazole, fenbendazole sulfoxide, and fenbendazole sulfone (FBZ-SO2) in liver and muscle samples were analyzed using HPLC-UV. Tissue WDIs were estimated using FDA, European Medicines Agency (EMA), and half-life multiplication methods for US poultry tolerances, EMA maximum residue limits, and the analytical limit of detection (LOD; 0.004 ppm). Terminal tissue elimination half-lives (T1/2) were estimated by non-compartmental analysis using a naĂŻve pooled data approach.ResultsThe tissue T1/2 was 14.4 h for liver, 13.2 h for thigh muscle, and 14.1 h for pectoral muscle. The maximum estimated withdrawal interval was 153 h (7 days) for FBZ-SO2 in pectoral muscle using the FDA tolerance method (95% confidence interval for the 99th percentile of the population), and the LOD as the residue limit.DiscussionThe results from this study support the use of FBZ-SO2 as the marker residue in the liver of pheasants and the provision of evidence based WDIs following the extra-label administration of fenbendazole medicated feed (100 ppm) for 7 days

    Safety Evaluation of Lasalocid Use in Chinese Ring-Necked Pheasants (Phasianus colchicus)

    Coccidiosis remains a significant threat to the welfare of game farm–reared pheasants in the United States. Although lasalocid has been demonstrated to be effective against pheasant specific coccidia, information regarding its safety in this species is lacking. The purpose of this study was to gather data on the safety of lasalocid when fed to Chinese ring-necked pheasants at one, two, and three times the recommended high dose of lasalocid used for prevention of coccidiosis in other poultry at three times the normal treatment period. Pheasant chicks (approximately 1 day-old; n  =  160) were randomly blocked by sex into four treatment groups and given their respective diets continuously for 6 wk. No significant differences were observed in overall feed consumption, weight gain, feed conversion rates, clinical pathology measurements, or tissue gross and histopathologic evaluations between controls and treatment groups associated with lasalocid administration. Based on the results of this study it appears that lasalocid fed at the recommended rate of 125 ppm is safe in Chinese ring-necked pheasants

    Toward accurate CO2 and CH4 observations from GOSAT

    The column-average dry air mole fractions of atmospheric carbon dioxide and methane (XCO2 and XCH4) are inferred from observations of backscattered sunlight conducted by the Greenhouse gases Observing SATellite (GOSAT). Comparing the first year of GOSAT retrievals over land with colocated ground-based observations of the Total Carbon Column Observing Network (TCCON), we find an average difference (bias) of-0.05% and-0.30% for XCO2 and XCH4 with a station-to-station variability (standard deviation of the bias) of 0.37% and 0.26% among the 6 considered TCCON sites. The root-mean square deviation of the bias-corrected satellite retrievals from colocated TCCON observations amounts to 2.8 ppm for XCO2 and 0.015 ppm for X CH4. Without any data averaging, the GOSAT records reproduce general source/sink patterns such as the seasonal cycle of XCO2 suggesting the use of the satellite retrievals for constraining surface fluxes.
