    Formal Training Methods in Selected North Dakota Retail Stores

    STATEMENT OF THE PROBLEM It was the primary goal of this thesis to examine in detail the formal training practices of selected North Dakota retailers. Another goal of this study was to gain insight into the opinions of North Dakota retail executives concerning the value of formal training to their employees. METHODOLOGY 1 A sample was taken of durable goods retail firms located in the nine major trade centers of North Dakota. Information was collected through the use of personal interviews with retail executives. RESULTS It was found that approximately 80% of the North Dakota chain retailers used formal training, as did 13% of independent retailers. Of the three categories of employees which received training-salespeople, non-selling employees, and management trainees — salespeople were more often enrolled in formal training programs. Considerable emphasis was placed on evaluation of the trainee\u27s progress by North Dakota retail executives. Supervisors\u27 evaluations and professional shopping service reports were commonly used. The primary objective of all North Dakota training programs was increased productivity of employees. CONCLUSION The acceptance of formal training as a useful tool of retailing was related to store size, ownership of the firm, and management attitude. Not all North Dakota retailers can use formal training, due to the small size of many of the stores. Several firms included in the survey were of sufficient size to benefit from formal training, but did not use it. Lack of acceptance of useful tools of retailing such as formal training by retail firms may lead to lowered profitability

    Apoptosis-Specific Protein (ASP) Identified in Apoptotic Xenopus Thymus Tumor Cells

    A novel apoptosis-specific protein (ASP) has recently been identified in the cytoplasm of apoptotic mammalian cells. This paper investigates whether ASP is found in Xenopus thymus tumor-derived lymphoid cell lines undergoing apoptosis and also in apoptotic, nontransformed splenocytes. Cultured Xenopus tumor lymphoid cells induced to undergo, apoptosis by serum deprivation or treatment with the calcium ionophore, ionomycin, displayed altered morphology typical of apoptotic cells, as judged by flow cytometric light-scatter characteristics and by fluorescence microscopy of acridine-orange-stained cells. Flow cytometry of permeabilized cells and fluorescence microscopy of acetone-fixed cytospins revealed that apoptotic Xenopus tumor cells, especially those displaying loss or condensation of DNA, displayed increased expression of epitopes recognized by a rabbit polyclonal antibody against ASP. Flow cytometry confirmed that ASP is also expressed in splenocytes induced to apoptose by culture in ionomycin or following concanavalin A stimulation. No increased expression of ASP was seen when lymphoid tumor cells or splenocytes were induced into necrosis by overdose with the antifungal agent amphotericin B. Western blotting with antibody against ASP identified the emergence of several protein bands in cell lysates from apoptotic, but not necrotic, Xenopus tumor cells. The new and simple methodology for identifying apoptotic cells described here is likely to be of value to those studying immune system development and associated programmed cell death in Xenopus

    Latent heat must be visible in climate communications

    C.R.'s portion of the work was carried out at the Jet Propulsion Laboratory, California Institute of Technology, under a contract with the National Aeronautics and Space Administration (80NM0018D0004). This work used JASMIN, the UK collaborative data analysis facility.Anthropogenic forcing is driving energy accumulation in the Earth system, including increases in the sensible heat content of the atmosphere, as measured by dry-bulb temperature—the metric that is almost universally used for communications about climate change. The atmosphere is also moistening, though, representing an accumulation of latent heat, which is partly concealed by dry-bulb temperature trends. We highlight that, consistent with basic theory, latent heat gains are outpacing sensible heat gains over about half of the Earth's surface. The difference is largest in the tropics, where global “hotspots” of total heat accumulation are located, and where regional disparities in heating rates are very poorly represented by dry-bulb temperatures. Including latent heat in climate-change metrics captures this heat accumulation and therefore improves adaptation-relevant understanding of the extreme humid heat and precipitation hazards that threaten these latitudes so acutely. For example, irrigation can lower peak dry-bulb temperatures, but amplify latent heat content by a larger margin, intensifying dangerous heat stress. Based on a review of the research literature, our Perspective therefore calls for routine use of equivalent temperature, a measure that expresses the combined sensible and latent heat content of the atmosphere in the familiar units of °C or K. We recognize that dry-bulb air temperature must remain a key indicator of the atmospheric state, not least for the many sectors that are sensitive to sensible heat transfer. However, we assert here that more widespread use of equivalent temperature could improve process understanding, public messaging, and adaptation to climate change.Publisher PDFPeer reviewe

    Replication of the association of HLA-B7 with Alzheimer's disease: a role for homozygosity?

    BACKGROUND: There are reasons to expect an association with Alzheimer's disease (AD) within the HLA region. The HLA-B & C genes have, however, been relatively understudied. A geographically specific association with HLA-B7 & HLA-Cw*0702 had been suggested by our previous, small study. METHODS: We studied the HLA-B & C alleles in 196 cases of 'definite' or 'probable' AD and 199 elderly controls of the OPTIMA cohort, the largest full study of these alleles in AD to date. RESULTS: We replicated the association of HLA-B7 with AD (overall, adjusted odds ratio = 2.3, 95% confidence interval = 1.4–3.7, p = 0.001), but not the previously suggested interaction with the ε4 allele of apolipoprotein E. Results for HLA-Cw*0702, which is in tight linkage disequilibrium with HLA-B7, were consistent with those for the latter. Homozygotes of both alleles appeared to be at particularly high risk of AD. CONCLUSION: HLA-B7 and HLA-Cw*0702 are associated with AD in the Oxford population. Because of the contradictions between cohorts in our previous study, we suggest that these results may be geographically specific. This might be because of differences between populations in the structure of linkage disequilibrium or in interactions with environmental, genetic or epigenetic factors. A much larger study will be needed to clarify the role of homozygosity of HLA alleles in AD risk

    GNOSIS: the first instrument to use fibre Bragg gratings for OH suppression

    GNOSIS is a prototype astrophotonic instrument that utilizes OH suppression fibres consisting of fibre Bragg gratings and photonic lanterns to suppress the 103 brightest atmospheric emission doublets between 1.47-1.7 microns. GNOSIS was commissioned at the 3.9-meter Anglo-Australian Telescope with the IRIS2 spectrograph to demonstrate the potential of OH suppression fibres, but may be potentially used with any telescope and spectrograph combination. Unlike previous atmospheric suppression techniques GNOSIS suppresses the lines before dispersion and in a manner that depends purely on wavelength. We present the instrument design and report the results of laboratory and on-sky tests from commissioning. While these tests demonstrated high throughput and excellent suppression of the skylines by the OH suppression fibres, surprisingly GNOSIS produced no significant reduction in the interline background and the sensitivity of GNOSIS and IRIS2 is about the same as IRIS2. It is unclear whether the lack of reduction in the interline background is due to physical sources or systematic errors as the observations are detector noise-dominated. OH suppression fibres could potentially impact ground-based astronomy at the level of adaptive optics or greater. However, until a clear reduction in the interline background and the corresponding increasing in sensitivity is demonstrated optimized OH suppression fibres paired with a fibre-fed spectrograph will at least provide a real benefits at low resolving powers.Comment: 15 pages, 13 figures, accepted to A

    Trace element geochemistry (Li, Ba, Sr, and Rb) using Curiosity's ChemCam: Early results for Gale crater from Bradbury Landing Site to Rocknest

    The ChemCam instrument package on the Mars rover, Curiosity, provides new capabilities to probe the abundances of certain trace elements in the rocks and soils on Mars using the laser-induced breakdown spectroscopy technique. We focus on detecting and quantifying Li, Ba, Rb, and Sr in targets analyzed during the first 100 sols, from Bradbury Landing Site to Rocknest. Univariate peak area models and multivariate partial least squares models are presented. Li, detected for the first time directly on Mars, is generally low (100 ppm and >1000 ppm, respectively. These analysis locations tend to have high Si and alkali abundances, consistent with a feldspar composition. Together, these trace element observations provide possible evidence of magma differentiation and aqueous alteration.     Detection of Rift Valley Fever Virus RNA in Formalin-Fixed Mosquitoes by In Situ Hybridization (RNAscope®).

    Rift Valley fever virus (RVFV) causes a zoonotic mosquito-borne haemorrhagic disease that emerges to produce rapid large-scale outbreaks in livestock within sub-Saharan Africa. A range of mosquito species in Africa have been shown to transmit RVFV, and recent studies have assessed whether temperate mosquito species are also capable of transmission. In order to support vector competence studies, the ability to visualize virus localization in mosquito cells and tissue would enhance the understanding of the infection process within the mosquito body. Here, the application of in situ hybridization utilizing RNAscope® to detect RVFV infection within the mosquito species, Culex pipiens, derived from the United Kingdom was demonstrated. Extensive RVFV replication was detected in many tissues of the mosquito with the notable exception of the interior of ovarian follicles

    PRAXIS: low thermal emission high efficiency OH suppressed fibre spectrograph

    PRAXIS is a second generation instrument that follows on from GNOSIS, which was the first instrument using fibre Bragg gratings for OH background suppression. The Bragg gratings reflect the NIR OH lines while being transparent to light between the lines. This gives a much higher signal-noise ratio at low resolution but also at higher resolutions by removing the scattered wings of the OH lines. The specifications call for high throughput and very low thermal and detector noise so that PRAXIS will remain sky noise limited. The optical train is made of fore-optics, an IFU, a fibre bundle, the Bragg grating unit, a second fibre bundle and a spectrograph. GNOSIS used the pre-existing IRIS2 spectrograph while PRAXIS will use a new spectrograph specifically designed for the fibre Bragg grating OH suppression and optimised for 1470 nm to 1700 nm (it can also be used in the 1090 nm to 1260 nm band by changing the grating and refocussing). This results in a significantly higher transmission due to high efficiency coatings, a VPH grating at low incident angle and low absorption glasses. The detector noise will also be lower. Throughout the PRAXIS design special care was taken at every step along the optical path to reduce thermal emission or stop it leaking into the system. This made the spectrograph design challenging because practical constraints required that the detector and the spectrograph enclosures be physically separate by air at ambient temperature. At present, the instrument uses the GNOSIS fibre Bragg grating OH suppression unit. We intend to soon use a new OH suppression unit based on multicore fibre Bragg gratings which will allow increased field of view per fibre. Theoretical calculations show that the gain in interline sky background signal-noise ratio over GNOSIS may very well be as high as 9 with the GNOSIS OH suppression unit and 17 with the multicore fibre OH suppression unit.Comment: SPIE conference proceedings 915
