YA literature allows adolescents to mirror themselves in books, safely discussing problems in their lives through conversations about how characters handle/mishandle problems. Novels provide maps to navigate risks and issues experienced by teens. The presenter, a former middle-grades teacher and author of No More “Us” and “Them,” teaches Bibliotherapy and will share Young Adult novels/memoirs and strategies that focus discussions on risks contemporary adolescents face

The war for the future

The following essay was born out of the authors’ participation in the Hamburg (Insecurity) Sessions 2019: Un-Cancelling the Future, and the discussions that took place in the workshop on Future Weapons and Emerging Technologies. The workshop participants critically addressed such problems as the future of defence technologies and their sociotechnical environments, cybersecurity and surveillance proliferation and the improvised instruments of insurgency. Our task was to weave the ideas and insights of the workshop’s participants into a vision of the world in 2040 and use it to motivate an analysis of the technoscientific imaginaries emerging in the present. From the workshop presentations and discussions, we identified three key areas of that allowed us to imagine the outlines and interactions of global security and technoscientific practice in 2040: ecopolitics: the exploitation of ecological systems for strategic ends; technonationalism: the use of advanced technologies to pursue racialised and nationalistic geopolitical agendas; and the security continuum: the extension of conflict modes to all aspects of social life and the open-source proliferation of security tools and techniques

No Sunday Business\u27: Navigating Religious Rules and Business Opportunities in the Shipyard Mennonite Settlement, Belize

Within the Old Colony Mennonite settlements of Belize, the relationship between religious and economic practices entails a constant navigation of the acceptable, where threats of worldliness come from technology and from contact with outsiders. This paper takes as its focus the business of a butcher in Shipyard settlement, whose daily work testifies to a navigation of both of these potential threats. This entrepreneur uses technologies of energy, transportation, and communication—operated in part by an outside worker—to extend the radius of his meat business. The tense environment of Shipyard’s religious diversity frames our discussion of these observations, leading us to reconsider our understanding of the Ordnung and its relation to business activity. To understand the entrepreneur’s skillful navigation of rules and opportunities, we use the term “social capital” (Bourdieu 1986; Portes 2010) to reflect on the paradoxical relationship between religious rules and entrepreneurial space—and to consider how the Ordnung can be seen as a spacious (rather than a constrictive) place for Mennonite entrepreneurs

Controlling dendritic cell function by targeting iron-modulating proteins identified through transcriptional analysis

Dendritic cells (DC) are key orchestrators of the body’s immune responses. DC control T cell proliferation and differentiation while displaying cognate antigen (Ag) to Ag-specific T cells. DC cytokine secretion is central to the differentiation of T cells into distinct, functional subsets. Stimulation with bacteria-derived molecules such as lipopolysaccharide (LPS) through Toll-like receptor 4 (TLR4) leads to interleukin-12 (IL-12) production by DC and supports T helper type 1 (Th1) generation in interacting cluster of differentiation 4 (CD4+) T cells. Fungal interactions with the Dectin-1 receptor yields IL-6, IL-23, and IL-1β production by DC to mediate Th17 cell polarization. DC, especially those exposed to IL-33, support the generation of IL-5- and IL-13- secreting Th2 cells. However, there are no identified DC-produced cytokines which have been shown to directly underlie Th2 polarization. The aim of this project was to identify novel genes and signaling pathways by which CD4+ T cell polarization is directed following DC TLR4/ST2 (IL1RL1) ligation. Our preliminary data demonstrated that IL-33 and LPS stimulate the expression of several iron-modulating proteins in DC, such as lipocalin 2 (Lcn2), hemoglobin-α, and cytochrome b5 reductase 3 (Cyb5R3), which act as a previously unappreciated DC nitric oxide (NO) sequestration system. Given these findings, we hypothesized that TLR4/ST2 ligation on DC causes expression of iron-modulating proteins which sequester NO. We found that while TLR4/ST2 receptor ligation on DC induces a NO sequestration system in DC, it involves the expression of hemoglobin-α and potentially other redundant reductase proteins, but not Cyb5R3. Instead, we determined that Cyb5R3 acts as a regulator of DC stimulatory capacity toward CD4+ T cells. Specifically, Cyb5R3 functions as a negative regulator of glycolysis. In Cyb5R3-expressing cells, unstimulated and IL-33-stimulated cells favor oxidative phosphorylation (OXPHOS) while LPS stimulation leads a shift towards glycolysis. Finally, Cyb5R3-deficient DC are both more inflammatory and more stimulatory, producing greater amounts of IL-12 and expressing more stimulatory surface markers, ultimately leading to Th1 cell polarization of naïve T cells, especially upon DC stimulation with LPS
