Effect of high pressure on surfactin production by Bacillus subtilis: implications for its application by the oil industry
Surfactin, a lipopeptide biosurfactant produced by Bacillus subtilis strains, exhibits extraordinary surface
active properties, as well as stability at a wide range of temperatures and salinities, making it useful to replace
the chemical surfactants in many industrial applications. The oil industry can take advantage of its application
to increase the productivity of oil reservoirs, through a technology known as microbial enhanced oil recovery
(MEOR). However, in order to make this technology advantageous from an economic point of view, the
surfactin-producing strains must be able of growing and producing the biosurfactant inside the oil reservoirs.
In this work, B. subtilis #573, isolated from an oil reservoir, was evaluated regarding its ability of producing
surfactin under oxygen limited conditions at high pressure. A central composite design (CCD) was used to
model the effect of pressure (3.8-46.2 bar) and temperature (35.3-46.7°C) on surfactin production. The results
obtained demonstrated that pressure (in the range studied) did not exhibit a negative effect on surfactin
production by this isolate, whereas temperatures higher than 45°C reduced its production. For most of the
different combinations of pressure and temperature assayed, surfactin production was observed after 24 h,
and the surface tension was reduced to values bellow 26.5 mN/m. At 41°C and 47 bar, B. subtilis #573
produced 31 ± 2 mg of surfactin per liter after 24 h, reducing the surface tension to 25.6 ±0.6 mN/m. These
results were similar to those achieved at the same temperature at atmospheric pressure (26.0 ± 0.3 mN/m
and 27 ± 3 mg surfactin/L). The surfactin produced in both cases exhibited a critical micelle concentration
value around 15 mg/L, and the chemical characterization (through UHPLC-MS) demonstrated the production
of similar percentages of the different surfactin isoforms (C12-, C13-, C14-, C15- and C16-surfactin) in both
conditions. Finally, the applicability of B. subtilis #573 in MEOR was studied in sand-pack columns. In assays
performed at 41°C and 47 bar, additional oil recoveries around 14% were obtained after 14 days in in situ
assays. These results demonstrate the applicability of B. subtilis #573 in in situ oil recovery processes.info:eu-repo/semantics/publishedVersio
Subjective perception of sexual achievement - an exploratory study on people with overweight
The literature reports that Body Mass Index (BMI) changes are directly
related to sexual interest. Individuals with overweight and obesity selfreport
fewer sexual partners and are less likely to have a romantic relationship
compared to their non-obese counterparts.
The objectives of this work are to know the level of perceived sexual
achievement, in overweight people with the Binge Eating Scale
(BES) and to study the association between age and the perception
of quality of sex in people with overweight are the objectives of
this work.
An exploratory and quantitative study was developed, based on a
sample of 218 patients of both genders, aged between 18 and
65 years. Data was collected in various hospitals of northern and central
Portugal with the Sexual Satisfaction Index (SSI) being used for
this purpose.
Of all participants, 64.5 % expressed overweight. On a scale of 0 to
100, the SSI was, on average, 26.7 (SD = 14.6). On average, Individuals
with severe poor scores on the BES registered the highest level of
sexual dissatisfaction. There were, statistically, significant differences
in the SSI, among individuals with and without overweight. Dissatisfaction
is greater the more advanced the age of participants. Contrary
to what is reported in the literature, there was no difference
between individuals with and without overweight according to the
number of sexual partners.
The results obtained with this research are consistent with the literature,
except for those concerning the number of sexual partners.info:eu-repo/semantics/publishedVersio

Structural and functional measures of invertebrate and fungal communities as predictors of eutrophication
To assess how eutrophication affects leaf-litter decomposition and the associated biota
in streams, we examined i) the structure of invertebrate and fungal communities and ii)
leaf mass loss of different plant species. Leaves of alder, chestnut, eucalyptus, plane tree
and oak were placed in coarse-mesh bags and immersed in 6 low-order streams along an
eutrophication gradient during 38 days. Additional benthic invertebrate samples were
collected with a hand-net. The values of the IBMWP biotic index applied to benthic
invertebrates increased from low to intermediate levels of eutrophication and then
dropped sharply at high and very high levels of eutrophication. Cluster analysis applied
to leaf-associated invertebrate and fungal communities separated the streams according
to the eutrophication level. The % of shredders on leaves decreased, whereas the % of
oligochaetes increased along the eutrophication gradient. Invertebrate biomass and
density, as well as fungal biomass and reproduction increased along the eutrophication
gradient, but decreased in the most eutrophic stream. Alder leaves decomposed faster
than other leaf species. A hump-shaped relationship was established between leaf mass
loss and the eutrophication gradient for all leaf species, suggesting that leaf
decomposition is a valuable tool to assess changes in stream water quality. Results
suggested that both structural and functional measures of invertebrate and fungal
communities are good predictors of stream eutrophication when assessing the ecological
integrity of streams.FEDER-POFC-COMPETE and FCT supported this study (PEst-C/BIA/UI4050/2011

Effect of NaCl on the aggregation behavior of rhamnolipids and implications in their biological activity
RICI7 - VII Iberian Meeting on Colloids and InterfacesRhamnolipids are a class of glycolipid biosurfactants produced mainly by Pseudomonas aeruginosa strains. Their structure comprises one or two rhamnose molecules linked to one or two -hydroxy fatty acids which can differ in the chain length and the degree of saturation. Rhamnolipids display a similar or better performance when compared with chemical surfactants, and exhibit higher biodegradability and lower toxicity, which make them potential alternatives to the synthetic surfactants in several applications [1]. Among them, rhamnolipids can be used as an alternative to the chemical fungicides to prevent the contamination and deterioration of agriculture products by fungi. In this work, the antifungal activity of rhamnolipids produced by P. aeruginosa #112 was evaluated against Aspergillus niger MUM 92.13 and Aspergillus carbonarius MUM 05.18. These species are among the most common fungal contaminants, and represent a health risk due to the production of mycotoxins. P. aeruginosa #112 produced eight different rhamnolipid congeners (including mono- and di-rhamnolipids). The growth inhibition percentages obtained for both fungi with the crude rhamnolipid mixture (3 g/L) were between 22 and 28%. Subsequently, it was studied the effect of different electrolytes in the antifungal activity of rhamnolipids. It was found that the growth of A. carbonarius MUM 05.18 and A. niger MUM 92.13 was completely inhibited by a combination of the crude rhamnolipid mixture (3 g/L) and NaCl (at concentrations between 375 and 875 mM). In order to establish a relationship between the antifungal activity and the aggregation behavior of rhamnolipids, the size distribution of the self-assembled structures formed by the crude rhamnolipid mixture in aqueous solution was studied at different NaCl concentrations through Dynamic Light Scattering. In the absence of NaCl, rhamnolipids formed micelles with an average size around 300 nm. The addition of NaCl resulted in an increase in the size of the structures formed, and for the highest NaCl concentration tested (875 mM), the average size of the self-assembled vesicles was higher than 2 µm. The formation of these large vesicles by rhamnolipids in the presence of NaCl was confirmed through Confocal Scanning Laser Microscopy. The results obtained demonstrated a relationship between the antifungal activity of rhamnolipids and their aggregation behavior, which opens the possibility to improve their biological activities for application in different fields.info:eu-repo/semantics/publishedVersio

Rhamnolipids inhibit aflatoxins production in Aspergillus flavus by causing structural damages in the fungal hyphae and down-regulating the expression of their biosynthetic genes
Aflatoxins are hepatotoxic and carcinogenic fungal secondary metabolites that usually contaminate crops and represent a serious health hazard for humans and animals worldwide. In this work, the effect of rhamnolipids (RLs) produced by Pseudomonas aeruginosa #112 on the growth and aflatoxins production by Aspergillus flavus MUM 17.14 was studied in vitro. At concentrations between 45 and 1500Â mg/L, RLs reduced the mycelial growth of A. flavus by 2340% and the production of aflatoxins by 93.999.5%. Purified mono-RLs and di-RLs exhibited a similar inhibitory activity on fungal growth. However, the RL mixture had a stronger inhibitory effect on aflatoxins production at concentrations up to 190Â mg/L, probably due to a synergistic effect resulting from the combination of both congeners. Using transmission electron microscopy, it was demonstrated that RLs damaged the cell wall and the cytoplasmic membrane of the fungus, leading to the loss of intracellular content. This disruptive phenomenon explains the growth inhibition observed. Furthermore, RLs down-regulated the expression of genes aflC, aflE, aflP and aflQ involved in the aflatoxins biosynthetic pathway (6.4, 44.3, 38.1 and 2.0-fold, respectively), which is in agreement with the almost complete inhibition of aflatoxins production. Overall, the results herein gathered demonstrate for the first time that RLs could be used against aflatoxigenic fungi to attenuate the production of aflatoxins, and unraveled some of their mechanisms of action. This study was supported by the Portuguese Foundation for Science
