Metabolism of vandetanib by cytochrome P450 expressed in prokaryotic systems

Při léčbě nádorových onemocnění, nesoucích konkrétní molekulárně genetické nebo morfologické znaky, se v poslední době začíná uplatňovat biologicky cílená léčba neboli cílená molekulární terapie. Vandetanib je perorálně užívané protinádorové léčivo patřící do skupiny tyrosinkinasových inhibitorů. Tyto inhibitory blokují receptory signálních drah, a tak inhibují růst, zvyšují buněčnou smrt a omezují šíření rakoviny. V dubnu 2011 byl vandetanib schválen americkým úřadem FDA pro léčbu progresivní či symptomatické medulární rakoviny štítné žlázy. Využívá se u lidí s metastázovanou nebo lokálně pokročilou rakovinou, kterou nelze operovat. V předkládané bakalářské práci byl studován metabolismus vandetanibu. Práce rozšiřuje poznatky o kinetice oxidace vandetanibu na N-desmethylvandetanib lidskými rekombinantními cytochromy P450 3A4 exprimovanými v membráně E. coli (Bactosomy). Studován byl vliv přítomnosti cytochromu b5 a vliv hladiny aktivity NADPH:cytochrom P450 reduktasy na aktivitu cytochromů P450 3A4. Za použití Bactosomů byl identifikován demethylovaný metabolit vandetanibu N-desmethylvandetanib, který byl separován vysokoúčinnou kapalinovou chromatografií (HPLC). Studium enzymové kinetiky naznačuje, že na oxidaci vandetanibu má vliv jak hladina aktivity NADPH:CYP reduktasy, tak i přítomnost cyt...1 Abstract Recently, biologically targeted treatment by another name targeted molecular therapies have begun to be used in the treatment of cancers bearing specific molecular genetic or morphological traits. Vandetanib is an oral anticancer drug that belongs to a group of tyrosine kinases inhibitors. These inhibitors block signal pathway receptors, thereby inhibit growth, stimulate cell death and reduce the spread of cancer. Vandetanib was approved in April 2011 by the US FDA for a treatment of progressive or symptomatic medullary thyroid cancer. It is used in patients with metastatic or inoperable locally advanced cancer. The metabolism of vandetanib was studied in this thesis. Specifically, the kinetics of vandetanib oxidation to N-desmethylvandetanib by human recombinant cytochromes P450 3A4 expressed in the membrane of E. coli (Bactosomes). The effect of the presence of cytochrome b5 and the effect of the level of NADPH: cytochrome P450 reductase activity on the activity of cytochrome P450 3A4 were studied. The demethylated metabolite of vandetanib, N-desmethylvandetanib, was identified and separated by high performance liquid chromatography (HPLC). The outdoor geography of high school students in Prague

Tato bakalářská práce se zaměřuje na volnočasové outdoorové aktivity studentů středních škol v Praze. Venkovní aktivity se dynamicky rozvíjejí, protože zahrnují všestranný pohyb a uskutečňují se venku pod širým nebem. Tyto aktivity jsou velice oblíbené také u mládeže. Cílem této práce je zjistit, jestli studenti středních škol v Praze tráví svůj volný čas aktivně nebo spíše pasivně, kolik mají denně v průměru hodin volného času a jakým outdoorovým aktivitám se věnují. Dále práce řeší otázky, s kým studenti tráví svůj volný čas, zda mají raději kolektivní či individuální sporty, letní či zimní atd. Metodika práce je postavena na kvantitativním výzkumu - dotazníkovém šetření. Dotazování se uskutečnilo na dvou gymnáziích, dvou středních odborných školách a dvou středních odborných učilištích v Praze. V závěru jsou vysloveny další náměty a návrhy pro další výzkum.This bachelor thesis focuses on the leisure time outdoor activities of the high school students in Prague. Outdoor activities spread dynamically, because they involve complex activities which take place outdoors. These activities are especially favoured by teenagers. The aims of this thesis is to reveal if high school students spend their leisure time actively or passively, how much leisure time they have and which outdoor activities are the most popular. More, the thesis also answers the questions with whom students spend their leisure time, if they prefer collective or individual sports, winter or summer sports etc. Methodologically the thesis stems from the quantitative methods - a questionnaire survey. The survey was conducted in two gymnasia, two high schools and two vocational training schools in Prague. Travelogues of Karel Čapek

Tato bakalářská práce se zabývá 5 cestopisy Karla Čapka ze zahraničních cest. Cestopisy jsou tematicky interpretovány a vzájemně srovnávány. Jsou zasazeny do kontextu publicistiky 20. a 30. let minulého století a do autorovy novinářské tvorby. Práce se také zabývá otázkou určení žánru a charakteristikou jednotlivých žánrů, zejména cestopisu a fejetonu. Nakonec je doplněna dobová recepce cestopisů jednotlivých literárních vědců nebo kritiků.This bachelor thesis deals with five travelogues of Karel Čapek from foreign trips. Travelogues are thematically interpreted and compared with each other. They are set in the context of journalism in the 1920's and 1930's and in the context of author's journalistic work. Partnerships and sexuality of people with mild and moderate mental disabilities.

The topic of this bachelor thesis is the partnership and sexual relations of people with mild and moderate mental disabilities. The bachelor thesis deals with the investigation of partner and sexual life of people with mild and moderate mental disabilities. The aim of this work is to present data obtained during interviews and to map problematic aspects in relationships of persons with mild and moderate mental disabilities who live in institutions and families. The thesis is divided into theoretical and empirical part. The theoretical part explains terms such as: mental disability, partnership, sexuality, partner and sexual life in institutions. In the empirical part is the analysis and interpretation of data, which is based on research conducted in the form of interviews, whose respondents are persons with mild and moderate mental disabilities. Respondents are users of social services such as sheltered housing, day care centre, home for people with disabilities and two respondents are students of special secondary school and live in a family.Tématem bakalářské práce jsou partnerské a sexuální vztahy osob s lehkým a středním mentálním postižením. Bakalářská práce se zabývá zkoumáním partnerského a sexuálního života osob s lehkým a středním mentálním postižením. Cílem práce je prezentace dat zjištěných během rozhovorů a zmapování problematických aspektů ve vztazích osob s lehkým a středním mentálním postižením žijících v institucích a v rodinách. Práce je rozdělena na teoretickou a empirickou část. V teoretické části jsou vysvětleny pojmy jako: mentální postižení, partnerství, sexualita, partnerský a sexuální život v zařízeních. V empirické části je analýza a interpretace dat, která vychází z výzkumu realizovaného formou rozhovorů, jehož respondenty jsou osoby s lehkým a středním mentálním postižením. Strategic plan of association Vraťme dětem pohyb z.s.

NÁZEV DIPLOMOVÉ PRÁCE: Strategický plán spolku Vraťme dětem pohyb, z. s. CÍLE: Diplomová práce se zabývá managementem neziskové sportovní organizace typu spolek, respektive jeho dlouhodobým plánováním. Cílem práce je charakterizovat organizaci Vraťme dětem pohyb, z. s., a její produkty. Dále provést strategickou situační analýzu prostředí předmětné organizace. Na základě zjištěných informací určit prioritní strategické cíle a oblasti, které budou následně řešeny v návrhu strategického plánu spolku, a to včetně doporučení na jeho implementaci. METODY: Práce je řešena jako aplikovaný výzkum. Pro účely práce bude využita kombinace kvantitativních a kvalitativních metod. Konkrétními metodami tohoto smíšeného výzkumu jsou pozorování, interview, dotazování, analýza textů a dokumentů, SWOT analýza, analýza PORTER a PEST a další dílčí analýzy. Design výzkumu včetně výzkumných otázek, které dávají práci rámec, jsou uvedeny v příslušné kapitole. VÝSLEDKY: V rámci strategické situační analýzy bylo analyzováno vnitřního i vnějšího prostředí předmětné organizace a vypracovaná souhrnná SWOT analýza. The Process of Employment in Public Service (The District Court of Mladá Boleslav)

katedra: KPE; přílohy: 2 CD; rozsah: 78 s., 6 s. příloh.The theme of this diploma work is the process of employment in public service which is particularly concentrated on the recruitment and selection of suitable workers at the district court of Mladá Boleslav. The theme was chosen on the basis of the personal work experience in this organization and possibility to obtain interesting and detailed information related to human resource. The diploma work is divided into two main chapters. The first chapter deals with theoretical aspects. According to these theoretical aspects, the human resource management is firstly put into context with other activities of the organization. Furthermore, this chapter describes the possibility to obtain workers in connection with the labour market, the process of recruitment of workers, the selection of job candidates and the completion of an employment contract and the worker´s introduction into the organization. The second chapter of diploma work deals with a brief description of the district court of Mladá Boleslav, a character and basic personnel information of the organization. Another part of this chapter includes a description of the process of employment at the district court of Mladá Boleslav. The chapter deals also with a problem of employment of persons with changed ability to work and suggestions of solving this problem. In conclusion of this chapter the process of employment at the organization is summarized.Tématem této práce je proces zaměstnávání ve státní správě, čili získávání a výběr vhodných pracovníků na Okresním soudě v Mladé Boleslavi. Téma práce bylo zvoleno na základě osobních pracovních zkušeností v této organizaci a možnosti získání zajímavých a podrobných informací týkajících se lidských zdrojů. Diplomová práce je rozdělena do dvou hlavních kapitol, kde první kapitola se zabývá teoretickou stránkou, ve které je nejprve zařazeno řízení lidských zdrojů mezi ostatní činnosti probíhající v organizaci. Dále se tato kapitola zabývá možnostmi získávání pracovníků v souvislosti s trhem práce a v neposlední řadě procesem získávání pracovníků jako takovým, výběrem uchazečů počínaje a uzavřením pracovní smlouvy a uvedením nového zaměstnance do organizace konče. Druhá kapitola se věnuje ve stručnosti popisu dané organizace, tedy Okresního soudu v Mladé Boleslavi, jejímu charakteru a základním personálním údajům. Další část kapitoly obsahuje popis samotného procesu zaměstnávání, tak jak probíhá na Okresním soudě v Mladé Boleslavi. Kapitola se také zabývá problémem zaměstnávání osob se změněnou pracovní schopností a návrhy řešení tohoto problému. Na závěr této kapitoly je shrnut celý proces zaměstnávání v této organizaci

Payroll system and employee benefits in a selected company

This bachelor thesis focuses on the payroll system and employee benefits in the selected accounting unit. The main aim of the thesis is to analyze the wage and employee benefits system and to propose measures that could optimize wage costs and improve employee satisfaction. The thesis firstly focuses on explaining the basic concepts related to this topic from a theoretical point of view and then these theoretical findings are applied in the practical part of the thesis to the selected accounting unit. Based on the analysis of the payroll system, the employee benefits system and other analyses in the accounting unit, and especially from the questionnaire survey conducted, certain measures for improving the current situation of the accounting unit are proposed. These suggestions are calculated in the work, and the effect of adding various benefits on the employee's wage and on the employer's wage costs is examined. By using employee benefits, the tax liability and therefore the wage cost on the employer's side can be optimized and the tax impact on the employee can be reduced. The different proposals are compared and finally a new system of employee benefits is proposed which is more beneficial for both the employee and the employer. The results of the work can help the selected accounting unit to improve its remuneration system and increase employee satisfaction
