    Oxygen photolysis in the Mauritanian upwelling: Implications for net community production

    We carried out 16 photochemical experiments of filtered surface water in a custom-built solar simulator and concomitant measurements of in vitro gross primary production (GPP) and respiration (R) in the Mauritanian upwelling during a Lagrangian study following three sulfur hexafluoride–labeled patches of upwelled water (P1 to P3). Oxygen photolysis rates were correlated with the absorbance of chromophoric dissolved organic matter (CDOM) at 300 nm, suggesting first-order kinetics with respect to CDOM. An exponential fit was used to calculate the apparent quantum yield (AQY) for oxygen photolysis, giving an average AQY of 0.00053 mmol O2 (mole photons m22 s21)21 at 280 nm and slope of 0.0012 nm21. Modeled photochemical oxygen demand (POD) at the surface (3–16 mmol m23 d21) occasionally exceeded R and was dominated by ultraviolet radiation (71– 79%). Euphotic-layer integrated GPP decreased with time during both P-1 and P-3, whereas R remained relatively constant and POD increased during P-1 and decreased during P-3. On Day 4 of P-3, GPP and POD maxima coincided with high CDOM absorbance, suggesting ‘‘new’’ CDOM production. Omitting POD may lead to an underestimation of net community production (NCP), both through in vitro and geochemical methods (here by 2–22%). We propose that oxygen-based NCP estimates should be revised upward. For the Mauritanian upwelling, the POD-corrected NCP was strongly correlated with standard NCP with a slope of 1.0066 6 0.0244 and intercept of 46.51 6 13.15 mmol m22 d21

    Horticultural land within the periurban fringe of NZ towns and cities increasingly is being developed for residential subdivision. Recent surveys have shown that concentrations of As, Cd, Cu, Pb, and ΣDDT (sum of DDT and its degradation products DDE and DDD) in such soils can exceed criteria protective of human health.¹ Soil ingestion is a key exposure pathway for non-volatile contaminants in soil. Currently in NZ, site-specific risk assessments and the derivation of soil guidelines protective of human health assume that all of the contaminant present in the soil is available for uptake and absorption by the human gastrointestinal tract. This assumption can overestimate health risks and has implications for the remediation of contaminated sites.² In comparison, the bioavailability of contaminants is considered when estimating exposure via dermal absorption and by ingestion of home-grown produce.³ Dermal absorption factors and plant uptake factors are included in the calculations for estimating exposures via these routes

The proposal builds upon EU-level initiatives to foster the voluntary cooperation of service providers in stopping the dissemination of terrorist content online, and echoes ongoing national developments which go a step further in imposing obligations – underpinned by considerable fines – on service providers. This article describes the main features of the proposal and highlights some of the policy challenges, legal questions and technological concerns it is likely to face on the road to adoption

    Characteristics of Adults with Anxiety or Depression Treated at an Internet Clinic: Comparison with a National Survey and an Outpatient Clinic

    Background There is concern that people seeking treatment over the Internet for anxiety or depressive disorders may not resemble the general population or have less severe disorders than patients attending outpatient clinics or cases identified in community surveys. Thus the response to treatment in Internet based trials might not generalize. Methodology We reviewed the characteristics of applicants to an Australian Internet-based treatment clinic for anxiety and depression, and compared this sample with people from a national epidemiological survey and a sample of patients at a specialist outpatient anxiety and depression clinic. Participants included 774 volunteers to an Internet clinic, 454 patients at a specialist anxiety disorders outpatient clinic, and 627 cases identified in a national epidemiological survey. Main measures included demographic characteristics, and severity of symptoms as measured by the Kessler 10-Item scale (K-10), the 12-item World Health Organisation Disability Assessment Schedule second edition (WHODAS-II), the Penn State Worry Questionnaire (PSWQ), the Body Sensations Questionnaire (BSQ), the Automatic Cognitions Questionnaire (ACQ), the Social Interaction Anxiety Scale (SIAS) and the Social Phobia Scale (SPS). Principal Findings The severity of symptoms of participants attending the two clinics was similar, and both clinic samples were more severe than cases in the epidemiological survey. The Internet clinic and national samples were older and comprised more females than those attending the outpatient clinic. The Internet clinic sample were more likely to be married than the other samples. The Internet clinic and outpatient clinic samples had higher levels of educational qualifications than the national sample, but employment status was similar across groups. Conclusions The Internet clinic sample have disorders as severe as those attending an outpatient clinic, but with demographic characteristics more consistent with the national sample. These data indicate that the benefits of Internet treatment could apply to the wider population.5 page(s

    Tracking TCRß sequence clonotype expansions during antiviral therapy using high-throughput sequencing of the hypervariable region

    To maintain a persistent infection viruses such as hepatitis C virus (HCV) employ a range of mechanisms that subvert protective T cell responses. The suppression of antigen-specific T cell responses by HCV hinders efforts to profile T cell responses during chronic infection and antiviral therapy. Conventional methods of detecting antigen-specific T cells utilize either antigen stimulation (e.g., ELISpot, proliferation assays, cytokine production) or antigen-loaded tetramer staining. This limits the ability to profile T cell responses during chronic infection due to suppressed effector function and the requirement for prior knowledge of antigenic viral peptide sequences. Recently, high-throughput sequencing (HTS) technologies have been developed for the analysis of T cell repertoires. In the present study, we have assessed the feasibility of HTS of the TCRβ complementarity determining region (CDR)3 to track T cell expansions in an antigen-independent manner. Using sequential blood samples from HCV-infected individuals undergoing antiviral therapy, we were able to measure the population frequencies of >35,000 TCRβ sequence clonotypes in each individual over the course of 12 weeks. TRBV/TRBJ gene segment usage varied markedly between individuals but remained relatively constant within individuals across the course of therapy. Despite this stable TRBV/TRBJ gene segment usage, a number of TCRβ sequence clonotypes showed dramatic changes in read frequency. These changes could not be linked to therapy outcomes in the present study; however, the TCRβ CDR3 sequences with the largest fold changes did include sequences with identical TRBV/TRBJ gene segment usage and high junction region homology to previously published CDR3 sequences from HCV-specific T cells targeting the HLA-B*0801-restricted 1395HSKKKCDEL1403 and HLA-A*0101-restricted 1435ATDALMTGY1443 epitopes. The pipeline developed in this proof of concept study provides a platform for the design of future experiments to accurately address the question of whether T cell responses contribute to SVR upon antiviral therapy. This pipeline represents a novel technique to analyze T cell dynamics in situations where conventional antigen-dependent methods are limited due to suppression of T cell functions and highly diverse antigenic sequences

    A Brief Wellbeing Training Session Delivered by a Humanoid Social Robot: A Pilot Randomized Controlled Trial

    Mental health and psychological distress are rising in adults, showing the importance of wellbeing promotion, support, and technique practice that is effective and accessible. Interactive social robots have been tested to deliver health programs but have not been explored to deliver wellbeing technique training in detail. A pilot randomised controlled trial was conducted to explore the feasibility of an autonomous humanoid social robot to deliver a brief mindful breathing technique to promote information around wellbeing. It contained two conditions: brief technique training (Technique) and control designed to represent a simple wait-list activity to represent a relationship-building discussion (Simple Rapport). This trial also explored willingness to discuss health-related topics with a robot. Recruitment uptake rate through convenience sampling was high (53%). A total of 230 participants took part (mean age = 29 years) with 71% being higher education students. There were moderate ratings of technique enjoyment, perceived usefulness, and likelihood to repeat the technique again. Interaction effects were found across measures with scores varying across gender and distress levels. Males with high distress and females with low distress who received the simple rapport activity reported greater comfort to discuss non-health topics than males with low distress and females with high distress. This trial marks a notable step towards the design and deployment of an autonomous wellbeing intervention to investigate the impact of a brief robot-delivered mindfulness training program for a sub-clinical population
