    Effect on Constant Versus Fluctuating Temperature Regimes on \u3ci\u3eBathyplectes Curculionis\u3c/i\u3e (Hymenoptera: Ichneumonidae) Activity

    Individual female Bathyplectes curculionis parasites were exposed to either a series of constant or fluctuating temperature regimes and supplied with a new group of host Hypera postica larvae every day. The fluctuating temperatures were calculated from an average of weekly air temperatures during a period of actual field oviposition by the parasite. The rearing of adult parasites under a constant versus fluctuating temperature regime resulted in no significant difference in parasitism, longevity, or fecundity. The threshold for B. curculionis activity was estimated to be 6-8°C

    Dynamic Properties of Fine-Grained Coal Refuse

    A simplified method of slope stability analysis is presented for upstream-constructed coal tailings dams subjected to earthquake shaking. The method employs a conventional method of slices approach, in which dynamic loads are represented as psuedostatic forces applied to each slice. Excess pore water pressures are estimated from cyclic triaxial tests performed on specimens of fine coal refuse. Cyclic triaxial test results are presented for fine coal refuse materials from six sites in the western Appalachian region. Measured excess pore water pressure values appear to be influenced by fine coal processing procedures, although material plasticity and grain characteristics are also important.     Desmosomes are prominent adhesive junctions present between many epithelial cells as well as cardiomyocytes. The mechanisms controlling desmosome assembly and remodeling in epithelial and cardiac tissue are poorly understood. We recently identified protein palmitoylation as a mechanism regulating desmosome dynamics. In this study, we have focused on the palmitoylation of the desmosomal cadherin desmoglein-2 (Dsg2) and characterized the role that palmitoylation of Dsg2 plays in its localization and stability in cultured cells. We identified two cysteine residues in the juxtamembrane (intracellular anchor) domain of Dsg2 that, when mutated, eliminate its palmitoylation. These cysteine residues are conserved in all four desmoglein family members. Although mutant Dsg2 localizes to endogenous desmosomes, there is a significant delay in its incorporation into junctions, and the mutant is also present in a cytoplasmic pool. Triton X-100 solubility assays demonstrate that mutant Dsg2 is more soluble than wild-type protein. Interestingly, trafficking of the mutant Dsg2 to the cell surface was delayed, and a pool of the non-palmitoylated Dsg2 co-localized with lysosomal markers. Taken together, these data suggest that palmitoylation of Dsg2 regulates protein transport to the plasma membrane.     Social media use, connectedness, and depression in graduate students

    This study will examine the relationship between social media use, sense of connectedness, and depression among graduate students. Graduate students are often disconnected from their social supports due to their program\u27s demands; additionally, the risk of depression in this population has been established in the literature. The impact of social media on depression has been investigated, and the results are discrepant. The relationship between a sense of connection, the use of social media, and depression in a graduate student population has not yet been explored. Graduate students at The Philadelphia College of Osteopathic Medicine (PCOM) and other universities studying psychology, medicine, biomedical science, or physician assistant studies will be included. Participants will be recruited through email and social media platforms. A control group of participants that do not use social media will be included. The following measures will be used: The Social Connectedness Scale-Revised (SCS-R), The Generalized Problematic Internet Use Scale 2 (GPIUS2), and The Patient Health Questionnaire-9 (PHQ-9). An inverse relationship is predicted between connectedness and depression. Participants with high scores on problematic internet use are anticipated to have elevated levels of depression and a low sense of connection compared to participants with low scores on problematic use. The subscales from the GPIUS2 are hypothesized to be predictive of participants\u27 sense of connection

    Development of tools for in situ hybridization in human embryonic stem cells

    Abstract only availableOur laboratory uses human embryonic stem cells (hESC) to understand how embryonic cells differentiate into the cells of the placenta. The long term goal of my research is to develop in situ hybridization methods and use them to determine where genes are being expressed within differentiating hESC colonies. I focused my research on two genes that code for transcription factors, OCT 4 and GATA 2. OCT 4 is required to maintain stem cells in an undifferentiated state. GATA 2 is thought to be one of the key transcription factors that promote stem cell differentiation into syncytiotrophoblast, a placental cell type. The first step in in situ hybridization is the development of a labeled RNA probe. I designed primers to amplify a 214bp portion of the GATA2 gene sequence by PCR, and then cloned the PCR product into E. coli bacteria. I confirmed the presence and orientation of the insert by using restriction digestion and DNA sequencing. I used bacteria already transformed with a 182bp OCT4 probe sequence to create a riboprobe. I grew up the bacteria overnight, lysed them, and purified the plasmid DNA. I linearized the plasmid with a restriction enzyme, purified it, and transcribed the DNA to RNA, incorporating digoxygenin (DIG) labeled nucleotides.     21-cm synthesis observations of VIRGOHI 21 - a possible dark galaxy in the Virgo Cluster

    Many observations indicate that dark matter dominates the extra-galactic Universe, yet no totally dark structure of galactic proportions has ever been convincingly identified. Previously we have suggested that VIRGOHI 21, a 21-cm source we found in the Virgo Cluster using Jodrell Bank, was a possible dark galaxy because of its broad line-width (~200 km/s) unaccompanied by any visible gravitational source to account for it. We have now imaged VIRGOHI 21 in the neutral-hydrogen line and find what could be a dark, edge-on, spinning disk with the mass and diameter of a typical spiral galaxy. Moreover, VIRGOHI 21 has unquestionably been involved in an interaction with NGC 4254, a luminous spiral with an odd one-armed morphology, but lacking the massive interactor normally linked with such a feature. Numerical models of NGC 4254 call for a close interaction ~10^8 years ago with a perturber of ~10^11 solar masses. This we take as additional evidence for the massive nature of VIRGOHI 21 as there does not appear to be any other viable candidate. We have also used the Hubble Space Telescope to search for stars associated with the HI and find none down to an I band surface brightness limit of 31.1 +/- 0.2 mag/sq. arcsec.Comment: 8 pages, accepted to ApJ, uses emulateapj.cls.
