    Practical and Theoretical Underpinnings of INFFER (Investment Framework For Environmental Resources)

    INFFER (Investment Framework for Environmental Resources) was developed to help investors of public funds to improve the delivery of outcomes from environmental programs. It assists environmental managers to design projects, to select delivery mechanisms, and to rank competing projects on the basis of benefits and costs. The design of INFFER and the activities of the INFFER projects are based on extensive experience of working with environmental managers and policy makers. This experience has highlighted a number of important practical lessons, that have strongly influenced the design and implementation of INFFER. These lessons include the need for simplicity, training and support of users, trusting relationships with users, transparency, flexibility, compatibility with the needs and contexts of users, and supportive institutional arrangements. In additions, the developers have paid close attention to the need for processes that are theoretically rigorous, resulting in a tool that deals appropriately and consistently with projects for different assets types, of different scales and durations, consistent with Benefit: Cost Analysis. The paper outlines theoretical considerations underpinning the way that INFFER deals with asset valuation, time lags, uncertainty, and the design of the metric used to rank projects.Environmental Economics and Policy,

    Lessons from implementing INFFER with regional catchment management organisations

    Investment in natural resource management (NRM) by regional organisations in Australia has been widely criticised for failing to achieve substantial environmental outcomes. The Investment Framework for Environmental Resources (INFFER) is a tool for developing and prioritising projects to address environmental issues such as water quality and biodiversity decline, environmental pest impacts and land degradation. It aims to achieve the most valuable environmental outcomes with the available resources. During 2008 and 2009 INFFER has been implemented with a number of catchment management organisations (CMOs) throughout Australia. In this paper, we report on lessons from and implications of this experience. Data on implementation were collected in formal and informal ways from staff of organisations that were using INFFER and state agencies, including: an on-line survey, benchmarking questions at training workshops, a formal on-going monitoring and evaluation process tracking the use of INFFER by CMOs, and comments made in correspondence and informal feedback to the INFFER team. In this paper we describe issues that arise when implementing INFFER with regions and organisations, and how the INFFER team has attempted to address these. Key issues include a desire to consider the community as an asset and emphasise capacity building, a rejection of the need for targeted investment, and various difficulties associated with specific aspects of the Framework. Existing institutional arrangements, and the legacy of past institutional arrangements, remain serious barriers to the adoption of methods to improve environmental outcomes from NRM investment. A lack of rigour in investment planning has become accepted as the norm, and resistance to processes to improve rigour is common.     Changing the direction of environmental investment in Australia: Learnings from implementing INFFER

    Investment in natural resource management (NRM) by regional organisations in Australia has been widely criticised for failing to achieve substantial environmental outcomes. The Investment Framework for Environmental Resources (INFFER) is a tool for developing and prioritising projects to address environmental issues such as water quality, biodiversity decline, environmental pest impacts and land degradation. INFFER is an asset-based, targeted, and outcome-focussed approach to environmental investment, and as such is a very different and more rigorous approach to prioritising possible environmental projects than used previously by most catchment management organisations (CMOs) in Australia. From 2008 to 2010 INFFER has been trialled with CMOs. Evaluation and benchmarking data obtained at 2-day INFFER training sessions with seven CMOs in three eastern Australia states are reported. Before commencing to use INFFER, CMO staff are generally confident about the current decision-making processes for environmental investment used within their organisation. In some cases, this initial perception challenges their acceptance of a new approach to investment decisionmaking. Key issues when implementing INFFER include concerns about changing the direction of CMO investment, concerns about compatibility with funder requirements, and various issues associated with specific aspects of the Framework. Perceived complexity of INFFER, existing institutional arrangements, and the legacy of past institutional arrangements remain serious barriers to the adoption of methods to improve environmental outcomes from NRM investment. Despite these difficulties INFFER is being used by a number of CMOs.     Mass spectrometric characterisation of the circulating peptidome following oral glucose ingestion in control and gastrectomised patients.

    RATIONALE: Meal ingestion triggers secretion of a variety of gut and endocrine peptides important in diabetes research which are routinely measured by immunoassays. However, similarities between some peptides (glucagon, oxyntomodulin and glicentin) can cause specificity issues with immunoassays. We used a liquid chromatography/tandem mass spectrometry (LC/MS/MS) methodology to unambiguously monitor multiple gut peptides in human plasma. METHODS: A simple acetonitrile-based protein precipitation step, followed by evaporation and solid-phase extraction, removed high-abundance proteins from samples prior to nano-LC/MS/MS analysis on an Orbitrap Q-Exactive Plus mass spectrometer using a data-dependent methodology. Database searching using PEAKS identified multiple gut-derived peptides, including peptides in the mid-pg/mL range. The relative levels of these and previously characterised peptides were assessed in plasma samples from gastrectomised and control subjects during an oral glucose tolerance test. RESULTS: Analysis of plasma extracts revealed significantly elevated levels of a number of peptides following glucose ingestion in subjects who had undergone gastrectomy compared with controls. These included GLP-1(7-36), GLP-1(9-36), glicentin, oxyntomodulin, GIP(1-42), GIP(3-42), PYY(1-36), PYY(3-36), neurotensin, insulin and C-peptide. Motilin levels decreased following glucose ingestion. Results showed good correlation with immunoassay-derived concentrations of some peptides in the same samples. The gastrectomy group also had higher, but non-glucose-dependent, circulating levels of peptides from PIGR and DMBT1. CONCLUSIONS: Overall, the approach showed that a fast, generic and reproducible LC/MS/MS methodology requiring only a small volume of plasma was capable of the multiplexed detection of a variety of diabetes-related peptides.Wellcome Trust and MR

    Terrane correlation between Antarctica, Mozambique and Sri Lanka; comparisons of geochronology, lithology, structure and metamorphism and possible implications for the geology of southern Africa and Antarctica

    Analysis of new lithological, structural, metamorphic and geochronological data from extensive mapping in Mozambique permits recognition of two distinct crustal blocks separated by the Lurio Belt shear zone. Extrapolation of the Mozambique data to adjacent areas in Sri Lanka and Dronning Maud Land, Antarctica permits the recognition of similar crustal blocks and allows the interpretation that the various blocks in Mozambique, Sri Lanka and Antarctica were once part of a mega-nappe, forming part of northern Gondwana, which was thrust-faulted c. 600 km over southern Gondwana during amalgamation of Gondwana at c. 590-550 Ma. The data suggest a deeper level of erosion in southern Africa compared with Antarctica. It is possible that this thrust domain extends, through the Zambezi Belt or Valley, as far west as the Damara Orogen in Namibia with the Naukluft nappes in Namibia, the Makuti Group, the Masoso Suite in the Rushinga area and the Urungwe klippen in northern Zimbabwe, fitting the mega-nappe pattern. Erosional products of the mountain belt are now represented by 700-400 Ma age detrital zircons present in the various sandstone formations of the Transantarctic Mountains, their correlatives in Australia, as well as the Urfjell Group (western Dronning Maud Land) and probably the Natal Group in South Africa

    Mathematics education is rarely out of the policy spotlight in England. Over the last ten years, considerable attention has been given to improving 14-19 mathematics curriculum pathways. In this paper we consider some of the challenges of enacting curriculum change by drawing upon evidence from our evaluation of the Mathematics Pathways Project. From 2004-10 this project, which was directed by England’s Qualifications and Curriculum Authority, aimed to improve the engagement, attainment and participation rates of 14-19 year old learners of mathematics. Our particular focus is upon the temporal problems of piloting new curriculum and assessment and we draw on Lemke’s discussion of time-scales, heterochrony and the adiabatic principle to consider the interlocking and interference of various change processes

    Differences in iNOS and Arginase Expression and Activity in the Macrophages of Rats Are Responsible for the Resistance against T. gondii Infection

    Toxoplasma gondii infects humans and warm blooded animals causing devastating disease worldwide. It has long been a mystery as to why the peritoneal macrophages of rats are naturally resistant to T. gondii infection while those of mice are not. Here, we report that high expression levels and activity of inducible nitric oxide synthase (iNOS) and low levels of arginase-1 (Arg 1) activity in the peritoneal macrophages of rats are responsible for their resistance against T. gondii infection, due to high nitric oxide and low polyamines within these cells. The opposite situation was observed in the peritoneal macrophages of mice. This discovery of the opposing functions of iNOS and Arg 1 in rodent peritoneal macrophages may lead to a better understanding of the resistance mechanisms of mammals, particularly humans and livestock, against T. gondii and other intracellular pathogens
