The species has been observed in Pisa (Pisa, Tuscany) and Piano di Vezzano (La Spezia, Liguria) on bay laurel plants. S. lauri has been firstly described in Greece and it was recorded in Cote d’Azur (France) in 2017

    BACKGROUND: The control of viral infections in the brain involves the activation of microglial cells, the macrophages of the brain that are constantly surveying the central nervous system, and the production of amyloid-beta (Aβ) as an anti-microbial molecule. Recent findings suggest a possible implication of HHV-6A in AD. We evaluated the effect of HHV-6A infection on microglial cell expression Aβ and the activation status, determined by TREM2, ApoE, cytokines, and tau expression. METHODS: We have infected microglial cells (HMC3, ATCC®CRL-3304), in monolayer and human peripheral blood monocyte-derived microglia (PBM-microglia) spheroid 3D model, with HHV-6A (strain U1102) cell-free virus inocula with 100 genome equivalents per 1 cell. We collected the cells 1, 3, 7, and 14 days post-infection (d.p.i.) and analyzed them for viral DNA and RNA, ApoE, Aβ (1-40, 1-42), tau, and phospho-tau (Threonine 181) by real-time immunofluorescence and cytokines by immunoenzymatic assay. RESULTS: We observed a productive infection by HHV-6A. The expression of Aβ 1-42 increased from 3 d.p.i., while no significant induction was observed for Aβ 1-40. The HHV-6A infection induced the activation (TREM2, IL-1beta, ApoE) and migration of microglial cells. The secretion of tau started from 7 d.p.i., with an increasing percentage of the phosphorylated form. CONCLUSIONS: In conclusion, microglial cells are permissive to HHV-6A infection that induces the expression of Aβ and an activation status. Meanwhile, we hypothesize a paracrine effect of HHV-6A infection that activates and induces microglia migration to the site of infection

    Promjenjljivost elektroforetskoga proteinskog profila u “Quarter” konja tijekom natjecanja u obuzdavanju.

    The aim of this study was to assess the effect of a reining session on serum total protein and their fractions. In the present study, eight Quarter Horses aged 5-15 years were subjected to reining pattern 5 of the National Reining Horse Association (NRHA). Blood samples were collected from each animal before (T0), immediately after the pattern (T1), after 1h (T2), 2h (T3) and 24h (T4), during the recovery period. On all samples serum total proteins (TP), albumin (Alb), α1-globulins, α2-globulins, β1-globulins, β2-globulins and γ-globulins concentrations, and albumin/globulin ratio (A/G) were assessed. One way repeated measure analysis of variance (ANOVA) showed the statistical significant effect of time on TP and Alb. The application of Bonferroni’s post-hoc comparison showed a statistical significant increase in TP values at T1 compared to T0. Alb concentration increased at T1 compared to T0, T2, T3 and T4. The present study shows how reining exercise affects electrophoretic parameters by increasing TP and Alb levels immediately after exercise. These results provide insight into the reining horse’s physiological response to exercise, allowing better evaluation of the athletic performance of this sport horse.Cilj ovog istraživanja bio je procijeniti učinak natjecanja u obuzdavanju konja na ukupne serumske proteine i na njihove pojedine frakcije. U istraživanju je osam konja Quarter pasmine u dobi od 5 do 15 godina bilo podvrgnuto vježbi 5. stupnja obuzdavanja prema Nacionalnoj udruzi za obuzdavanja konja. Uzorci krvi bili su uzeti od svakog konja prije vježbe (T0), neposredno nakon vježbe (T1), zatim nakon jednog sata (T2), dva sata (T3) i 24 sata (T4) odmora. U svim uzorcima seruma određena je koncentracija ukupnih proteina, albumina (Alb), α1-globulina, α2-globulina, β1-globulina, β2-globulina i γ-globulina te je određen omjer između albumina i globulina (A/G). Jednosmjerna analiza varijance (ANOVA) pokazala je statistički značajan učinak vremena na ukupne proteine i albumin. Primjena Bonferronijeve post-hoc usporedbe pokazala je statistički značajno povećanje vrijednosti ukupnih proteina neposredno nakon vježbe u odnosu na one prije vježbe. Koncentracija albumina bila je povećana neposredno nakon vježbe u usporedbi s koncentracijom prije vježbe te onom jednog sata, dva sata i 24 sata nakon vježbe. Istraživanje pokazuje kako natjecateljsko obuzdavanje konja utječe na elektroforetske pokazatelje povećanjem razine ukupnih proteina i albumina neposredno nakon vježbe. Rezultati pružaju uvid u fiziološki odgovor na vježbu što omogućuje bolju prosudbu atletskih sposobnosti ove pasmine športskog konja

    Adaptation of some energetic parameters during transition period in dairy cows

    The aim of this study was to evaluate the effect of peripartum period and lactation class on serum β-hydroxybutyrate, non-esterified fatty acids (NEFA), insulin and glucose concentration in dairy cows. Thirty-five multiparous Holstein cows were selected from a high-producing dairy farm. The animals were divided according to their lactation class: second lactation group (L2, n = 16), third lactation group (L3, n = 10) and fourth lactation group (L4, n = 9). Blood sampling were performed 7 ± 5 days before calving (Pre/C) and 7 ± 5 days after calving (Post/C). Two-way analysis of variance was applied to determine significant effects of lactation class (L2, L3 and L4) and period (Pre/C and Post/C) on studied parameters. Increased NEFA values and lower insulin levels were found in Post/C respect to Pre/C in L2 and L4. These changes highlight the difficulty of dairy cows to cope with the energy demand characterizing the transition period. Improving in knowledge of energetic metabolism changes may help to supply a new strategy of farm management and reproductive performance in dairy cows during the transition period

    Mineralogy and heavy metal assessment of the Pietra del Pertusillo reservoir sediments (Southern Italy)

    The Pietra del Pertusillo freshwater reservoir is a major artificial lake of environmental, biological, and ecological importance located in the Basilicata region, southern Italy. The reservoir arch-gravity dam was completed in 1963 for producing hydroelectric energy and providing water for human use, and nearby there are potential sources of anthropogenic pollution such as urban and industrial activities. For the first time, the minero-chemistry of the lake and fluvio-lacustrine sediments of the reservoir have been evaluated to assess the environmental quality. Moreover, the composition of fluvial sediments derived from the peri-lacual zone of the reservoir and of local outcropping bedrock were also studied to understand the factors affecting the behavior of elements in the freshwater reservoir, with particular attention paid to heavy metals. In Italy, specific regulatory values concerning the element threshold concentration for lake and river sediments do not exist, and for this reason, soil threshold values are considered the standard for sediments of internal waters. The evaluation of the environmental quality of reservoir sediments has been performed using enrichment factors obtained with respect to the average composition of a reconstructed local upper continental crust. We suggest this method as an innovative standard in similar conditions worldwide. In the studied reservoir sediments, the trace elements that may be of some environmental concern are Cr, Cu, Zn, As, and Pb although, at this stage, the distribution of these elements appears to be mostly driven by geogenic processes. However, within the frame of the assessment and the preservation of the quality of aquatic environments, particular attention has to be paid to As (which shows median value of 10 ppm, reaching a maximum value of 26 ppm in Quaternary sediments), constantly enriched in the lacustrine samples and especially in the fine-grained fraction (median = 8.5 ppm)

    Transforming activities of Chlamydia pneumoniae in human mesothelial cells

    Knowledge in viral oncology has made considerable progress in the field of cancer fight. However, the role of bacteria as mediators of oncogenesis has not yet been elucidated. As cancer still is the leading cause of death in developed countries, understanding the long-term effects of bacteria has become of great importance as a possible means of cancer prevention. This study reports that Chlamydia pneumoniae infection induce transformation of human mesothelial cells. Mes1 cells infected with C. pneumoniae at a multiplicity of infection of 4 inclusion-forming units/cell showed many intracellular inclusion bodies. After a 7-day infection an increased proliferative activity was also observed. Real-time PCR analysis revealed a strong induction of calretinin, Wilms’ tumour gene 1, osteopontin, matrix metalloproteinases-2, and  membrane-type 1 metalloproteinases gene expression in Mes1 cell, infected for a longer period (14 days). The results were confirmed by western blot analysis. Zymography analysis showed that C. pneumoniae modulated the in-vitro secretion of MMP-2 in Mes1 cells both at 7 and 14 days. Cell invasion, as measured by matrigel-coated filter, increased after 7 and 14 days infection with C. pneumoniae, compared with uninfected Mes1 cells. The results of this study suggest that C. pneumoniae infection might support cellular transformation, thus increasing lung cancer risk.     The Rapid Identification of Anoplophora chinensis (Coleoptera: Cerambycidae) From Adult, Larval, and Frass Samples Using TaqMan Probe Assay

    A molecular diagnostic method using TaqMan probe qPCR is presented for the identification of Anoplophora chinensis (Förster) (Coleoptera: Cerambycidae) from whole body insects (adults and larvae) and frass samples stored under different conditions. The results showed a perfect amplification of DNA from all samples; the repeatability and reproducibility of the protocol were very good, with standard deviations of inter-run and intrarun variability less than or equal to 0.5. The assay allowed to discern all A. chinensis samples from those of the other non-target wood-borer species, with 100% correspondence to the homologous sequences. No amplification or cross reactions were observed with A. glabripennis (Motschulsky) (Coleoptera: Cerambycidae), which is the most related species among those tested. The protocol was validated by an internal blind panel test which showed a good correspondence between the results obtained by different operators in the same lab. The analytical sensitivity for the lab frass with the Probe qPCR, namely the lowest amount of A. chinensis DNA that can be detected (LoD), was 0.64 pg/μl with a Cq of 34.87. The use of indirect evidence for the identification of a pest is an important feature of the method, which could be crucial to detect the presence of wood-boring insects. This diagnostic tool can help prevent the introduction of A. chinensis into new environments or delimit existing outbreak areas thanks to indirect frass diagnosis

    Association between the uremic toxins indoxyl-sulfate and p-cresyl-sulfate with sarcopenia and malnutrition in elderly patients with advanced chronic kidney disease

    Abstract Background in patients with chronic kidney disease (CKD) indoxyl sulfate (IS) and p-cresyl sulfate (PCs) may induce sarcopenia either directly or via systemic inflammation. We evaluated whether IS and PCs were associated with: sarcopenia, systemic inflammation and nutritional status. Methods: we examined cross sectionally 93 patients with advanced CKD. Sarcopenia was identified according to EWGSOP2 definition. Malnutrition was assessed by Malnutrition Inflammation Score (MIS) and Protein Energy Wasting syndrome (PEW). Inflammatory status was assessed by dosing: CRP, IL6, TNFα, MCP1, IL10, IL17, IL12p70. Results: we did not find any association of sarcopenia with IS and PCs. IS was associated with LogTNFα and LogMCP-1 in the overall cohort (r = 0.30, p = 0.0043; r = 0.22 p = 0.047) and in not sarcopenic patients (r = 0.32, p = 0.0077; r = 0.25, p = 0.041). PCs was associated with LogIL10 and LogIL12p70 in sarcopenic patients (r = 0.58, p = 0.0042; r = 0.52, p = 0.013). IS was higher in patients without PEW (p = 0.029), while PCs was higher in patients with PEW (p = 0.0040). IS and PCs were not different in patients with normal or increased MIS. Conclusions: IS and PCs were not associated with sarcopenia, although they were both associated with some inflammatory pathways. Notably, we found a positive association of PCs with PEW syndrome

    First record of Aleurocanthus camelliae Kanmiya & Kasai, 2011 (Hemiptera, Aleyrodidae) from Italy, on ornamental Camellia spp. plants

    This paper provides a first report of Aleurocanthus camelliae, the Camellia spiny whitefly, from Italy. The pest was found on plants of Camellia spp. grown in the nursery. Brief morphological and biological information is provided on this whitefly, as well as some considerations on the phytosanitary measures to be adopted to reduce the potential risk of its spread on ornamental plants in Europe and the EPPO region
