    SIRT1 negatively regulates the expression of Prl2C3,a senescence-associated protein

    SIRT1 is a mammalian homologue of yeast longevity protein Sir2. SIRT1 deacetylates transcription factors, cofactors, and histones in an NAD+-dependent manner, and promotes cell survival, anti-oxidative function, and DNA repair. Although some studies have indicated that SIRT1 is involved in longevity, the function of SIRT1 for preventing aging and senescence is still unclear. In mouse embryonic fibroblasts (MEFs), we found that SIRT1 expression decreased by aging and IRT1 reciprocally regulated the expression level of Prl2C3, one of the prolactin-like peptides. In young MEFs, purified Prl2C3 inhibited the growth and increased the number of senescence-associated β galactosidase-positive cells with enlarged and flattened shapes. Moreover, immunostaining of human skin sections showed the expression of Prl2C3 in the basal cells of the epidermis. These results indicate that SIRT1 negatively regulates a senescence-associated protein rl2C3

    Multiplex Genome Editing in Tomato

    Several expression systems for multiple guide RNA (gRNAs) have been developed in the CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR associated protein 9) system to induce multiple-gene modifications in plants. Here, we evaluated mutation efficiencies in the tomato genome using multiplex CRISPR/Cas9 vectors consisting of various Cas9 expression promoters with multiple gRNA expression combinations. In transgenic tomato calli induced with these vectors, mutation patterns varied depending on the promoters used to express Cas9. By using the tomato ELONGATION FACTOR-1α (SlEF1α) promoter to drive Cas9, occurrence of various types of mutations with high efficiency was detected in the tomato genome. Furthermore, sequence analysis showed that the majority of mutations using the multiplex system with the SlEF1α promoter corresponded to specific mutation pattern of deletions produced by self-ligation at two target sites of CRISPR/Cas9 with low mosaic mutations. These results suggest that optimizing the Cas9 expression promoter used in CRISPR/Cas9-mediated mutation improves multiplex genome editing, and could be used effectively to disrupt functional domains precisely in the tomato genome

    Relationship between mind wandering and autobiographical reasoning

    Mind wandering (MW) is the shift in the contents of thought away from an ongoing task to self-generated thoughts which is unrelated to the task. MW is thought to have the role of integrating one’s experiences into a meaningful context through autobiographical reasoning. However, the relationship between MW and autobiographical reasoning is only a theoretical suggestion and no previous studies have directly examined this relationship. In this study, we performed a questionnaire survey to clarify the role of MW that integrates experiences by examining whether there is a relationship between MW and autobiographical reasoning. As a result, a positive correlation was found between the Mind-Wandering Questionnaire and the Thinking About Life Experiences (TALE) scale. There was also a positive correlation between the Daydream Frequency and TALE scales. This result indicates that MW is related to autobiographical reasoning.本研究は,JST,COI,JPMJCE1311の支援を受けたものです

    SlIAA9 Controls Tomato Elongation

    Tomato INDOLE-3-ACETIC ACID9 (SlIAA9) is a transcriptional repressor in auxin signal transduction, and SlIAA9 knockout tomato plants develop parthenocarpic fruits without fertilization. We generated sliaa9 mutants with parthenocarpy in several commercial tomato cultivars (Moneymaker, Rio Grande, and Ailsa Craig) using CRISPR-Cas9, and null-segregant lines in the T1 generation were isolated by self-pollination, which was confirmed by PCR and Southern blot analysis. We then estimated shoot growth phenotypes of the mutant plants under different light (low and normal) conditions. The shoot length of sliaa9 plants in Moneymaker and Rio Grande was smaller than those of wild-type cultivars in low light conditions, whereas there was not clear difference between the mutant of Ailsa Craig and the wild-type under both light conditions. Furthermore, young seedlings in Rio Grande exhibited shade avoidance response in hypocotyl growth, in which the hypocotyl lengths were increased in low light conditions, and sliaa9 mutant seedlings of Ailsa Craig exhibited enhanced responses in this phenotype. Fruit production and growth rates were similar among the sliaa9 mutant tomato cultivars. These results suggest that control mechanisms involved in the interaction of AUX/IAA9 and lights condition in elongation growth differ among commercial tomato cultivars

    Vegetable juice preload ameliorates postprandial blood glucose concentration in healthy women : A randomized cross-over trial

    Background and Objectives: The aim of this study was to evaluate the acute effect of drinking vegetable juice 20 min before carbohydrate on postprandial blood glucose concentrations in young healthy women. Method: In this randomized controlled cross-over study, 24 women (age 21.3 ±0.6 years, HbA1c 5.4 ±0.2 %, mean ± SD) consumed either 200 g of vegetable juice, vegetable (150 g of tomato and 40 g of broccoli), or water at 20 min before consuming 200 g of boiled white rice for 3 separate days. The blood glucose concentrations were measured by self-monitoring blood glucose pre- and post-breakfast at -20, 0, 15, 30, 45, 60, 120, and 180 min. The glycemic parameters were compared among 3 days. Results: The incremental glucose peak at 45 min (vegetable juice 48.3 ± 4.1, vegetable 47.4 ± 3.3 vs. water 66.8 ± 4.3 mg/dl, respectively, both p < 0.01, mean ± SEM) and large amplitude of glycemic excursion (LAGE; vegetable juice 57.1 ± 3.1, vegetable 58.3 ± 3.6 vs. water 78.3 ± 4.3 mg/dl, respectively, both p < 0.05) in consuming vegetable juice and vegetable at 20 min before carbohydrate intake were all significantly lower than those of water. There was no significant difference between glycemic parameters of vegetable juice and vegetable. Conclusions: Drinking vegetable juice 20 min before carbohydrate ameliorates the postprandial blood glucose concentrations as well as vegetable preload, despite total amounts of energy and carbohydrate of vegetable juice or vegetable are higher than those of water

    Genome editing in plants using CRISPR type I-D nuclease

    Genome editing in plants has advanced greatly by applying the clustered regularly interspaced short palindromic repeats (CRISPRs)-Cas system, especially CRISPR-Cas9. However, CRISPR type I—the most abundant CRISPR system in bacteria—has not been exploited for plant genome modification. In type I CRISPR-Cas systems, e.g., type I-E, Cas3 nucleases degrade the target DNA in mammals. Here, we present a type I-D (TiD) CRISPR-Cas genome editing system in plants. TiD lacks the Cas3 nuclease domain; instead, Cas10d is the functional nuclease in vivo. TiD was active in targeted mutagenesis of tomato genomic DNA. The mutations generated by TiD differed from those of CRISPR/Cas9; both bi-directional long-range deletions and short indels mutations were detected in tomato cells. Furthermore, TiD can be used to efficiently generate bi-allelic mutant plants in the first generation. These findings indicate that TiD is a unique CRISPR system that can be used for genome engineering in plants

    Relationship between Changes in Physical Activity and Changes in Health-related Quality of Life in Patients on Chronic Hemodialysis with 1-Year Follow-up

    In a longitudinal study, we examined the link between changes in physical activity and changes in health-related quality of life (HRQOL) in patients on chronic hemodialysis. Seventy-one patients (43 males, 28 females; aged 70.9±10.6 years) on chronic hemodialysis in September 2013 were enrolled. The data of the 43 patients whose complete measurements were taken again in September 2014 were used for the longitudinal analysis. Clinical parameters including age, height, dry weight, duration of hemodialysis, blood pressure (BP), blood triglyceride and HDL cholesterol levels, physical activity, and HRQOL were evaluated. Physical activity was measured by a tri-accelerometer, and HRQOL was evaluated by the EuroQol questionnaire (EQ-5D). In the first cross-sectional analysis, EQ-5D scores were significantly correlated with daily step counts (steps per day) on all days and non-hemodialysis days. In the second longitudinal analysis, in the women, changes in EQ-5D scores were positively correlated with changes in daily step counts on all days. In all patients, changes in EQ-5D were weakly and negatively correlated with changes in physical activity (1-3 METs: min per day) on hemodialysis days. Promoting daily physical activity may improve the HRQOL in patients on chronic hemodialysis, especially in women

    Wash or wipe? A comparative study of skin physiological changes between water washing and wiping after skin cleaning

    Background/purpose: Presently, skin-cleaning agents that claim to be removed by water or wiping alone are commercially available and have been used for the purpose of bed baths. However, there is a lack of knowledge on how water washing and wiping differently affect skin physiological functions or ceramide content. The aim of this study was to compare the effects of water washing and wiping on skin physiological functions and ceramide content. Methods: Three kinds of the cleaning agents with different removal techniques (ie, water washing and wiping) were used in this study. Skin physiological functions (ie, transepidermal water loss, skin hydration, and skin pH) and skin ceramide content were measured before and after seven consecutive days of the application of each cleaning agent. Results: No significant differences in skin physiological functions or ceramide content were observed between water washing and wiping. Conclusion: Cleaning agents that claim to be removed by water washing or wiping do not affect skin physiological functions or ceramide content by either removal method. © 2017 John Wiley & Sons A/S.Embargo Period 12 month
