    Wavelength dependent ac-Stark shift of the 1S0 - 3P1 transition at 657 nm in Ca

    We have measured the ac-Stark shift of the 4s2 1S0 - 4s4p 3P1 line in 40Ca for perturbing laser wavelengths between 780 nm and 1064 nm with a time domain Ramsey-Borde atom interferometer. We found a zero crossing of the shift for the mS = 0 - mP = 0 transition and \sigma polarized perturbation at 800.8(22) nm. The data was analyzed by a model deriving the energy shift from known transition wavelengths and strengths. To fit our data, we adjusted the Einstein A coefficients of the 4s3d 3D - 4s4p 3P and 4s5s 3S - 4s4p 3P fine structure multiplets. With these we can predict vanishing ac-Stark shifts for the 1S0 m = 0 - 3P1 m = 1 transition and \sigma- light at 983(12) nm and at 735.5(20) nm for the transition to the 3P0 level.Comment: 8 pages, 5 figures, 2 table

    Long range transport of ultra cold atoms in a far-detuned 1D optical lattice

    We present a novel method to transport ultra cold atoms in a focused optical lattice over macroscopic distances of many Rayleigh ranges. With this method ultra cold atoms were transported over 5 cm in 250 ms without significant atom loss or heating. By translating the interference pattern together with the beam geometry the trap parameters are maintained over the full transport range. Thus, the presented method is well suited for tightly focused optical lattices that have sufficient trap depth only close to the focus. Tight focusing is usually required for far-detuned optical traps or traps that require high laser intensity for other reasons. The transport time is short and thus compatible with the operation of an optical lattice clock in which atoms are probed in a well designed environment spatially separated from the preparation and detection region.Comment: 14 pages, 6 figure

    Insulin inhibits cardiac contractility by inducing a Gi-biased β2-adrenergic signaling in hearts.

    Insulin and adrenergic stimulation are two divergent regulatory systems that may interact under certain pathophysiological circumstances. Here, we characterized a complex consisting of insulin receptor (IR) and β2-adrenergic receptor (β2AR) in the heart. The IR/β2AR complex undergoes dynamic dissociation under diverse conditions such as Langendorff perfusions of hearts with insulin or after euglycemic-hyperinsulinemic clamps in vivo. Activation of IR with insulin induces protein kinase A (PKA) and G-protein receptor kinase 2 (GRK2) phosphorylation of the β2AR, which promotes β2AR coupling to the inhibitory G-protein, Gi. The insulin-induced phosphorylation of β2AR is dependent on IRS1 and IRS2. After insulin pretreatment, the activated β2AR-Gi signaling effectively attenuates cAMP/PKA activity after β-adrenergic stimulation in cardiomyocytes and consequently inhibits PKA phosphorylation of phospholamban and contractile responses in myocytes in vitro and in Langendorff perfused hearts. These data indicate that increased IR signaling, as occurs in hyperinsulinemic states, may directly impair βAR-regulated cardiac contractility. This β2AR-dependent IR and βAR signaling cross-talk offers a molecular basis for the broad interaction between these signaling cascades in the heart and other tissues or organs that may contribute to the pathophysiology of metabolic and cardiovascular dysfunction in insulin-resistant states

    Absolute frequency measurement of the magnesium intercombination transition 1S03P1^1S_0 \to ^3P_1

    We report on a frequency measurement of the (3s2)1S0(3s3p)3P1(3s^2)^1S_0\to(3s3p)^3P_1 clock transition of 24^{24}Mg on a thermal atomic beam. The intercombination transition has been referenced to a portable primary Cs frequency standard with the help of a femtosecond fiber laser frequency comb. The achieved uncertainty is 2.5×10122.5\times10^{-12} which corresponds to an increase in accuracy of six orders of magnitude compared to previous results. The measured frequency value permits the calculation of several other optical transitions from 1S0^1S_0 to the 3PJ^3P_J-level system for 24^{24}Mg, 25^{25}Mg and 26^{26}Mg. We describe in detail the components of our optical frequency standard like the stabilized spectroscopy laser, the atomic beam apparatus used for Ramsey-Bord\'e interferometry and the frequency comb generator and discuss the uncertainty contributions to our measurement including the first and second order Doppler effect. An upper limit of 3×10133\times10^{-13} in one second for the short term instability of our optical frequency standard was determined by comparison with a GPS disciplined quartz oscillator.Comment: 8 pages, 8 figure

    Exercise training improves vascular mitochondrial function

    Exercise training is recognized to improve cardiac and skeletal muscle mitochondrial respiratory capacity; however, the impact of chronic exercise on vascular mitochondrial respiratory function is unknown. We hypothesized that exercise training concomitantly increases both vascular mitochondrial respiratory capacity and vascular function. Arteries from both sedentary (SED) and swim-trained (EX, 5 wk) mice were compared in terms of mitochondrial respiratory function, mitochondrial content, markers of mitochondrial biogenesis, redox balance, nitric oxide (NO) signaling, and vessel function. Mitochondrial complex I and complex I + II state 3 respiration and the respiratory control ratio (complex I + II state 3 respiration/complex I state 2 respiration) were greater in vessels from EX relative to SED mice, despite similar levels of arterial citrate synthase activity and mitochondrial DNA content. Furthermore, compared with the SED mice, arteries from EX mice displayed elevated transcript levels of peroxisome proliferative activated receptor-γ coactivator-1α and the downstream targets cytochrome c oxidase subunit IV isoform 1, isocitrate dehydrogenase (Idh) 2, and Idh3a, increased manganese superoxide dismutase protein expression, increased endothelial NO synthase phosphorylation (Ser1177), and suppressed reactive oxygen species generation (all P \u3c 0.05). Although there were no differences in EX and SED mice concerning endothelium-dependent and endothelium-independent vasorelaxation, phenylephrine-induced vasocontraction was blunted in vessels from EX compared with SED mice, and this effect was normalized by NOS inhibition. These training-induced increases in vascular mitochondrial respiratory capacity and evidence of improved redox balance, which may, at least in part, be attributable to elevated NO bioavailability, have the potential to protect against age- and disease-related challenges to arterial function

    Recent archaeological investigations at Ascoli Satriano have shed new light on the pre-Roman occupation of northern Apulia (ancient Daunia). The findings document a long-term and locally consistent attachment to the core site of Giarnera Piccola from the 8th to the late 4th century BCE. A close relationship is discerned, both in time and space, of architectural structures and burials. The new local discoveries testify to the existence of this situation from the earliest hut structures onwards and show its persistence until the place’s abandonment. While the detection of new fragments of two Daunian stelae in a shaft/well points to early burial activities, the character of the entangled relationship still has to be scrutinized. Archaeometric and scientific analyses furthermore attest to activities in agriculture and in pottery production

    Photoassociation spectroscopy of cold calcium atoms

    Photoassociation spectroscopy experiments on 40Ca atoms close to the dissociation limit 4s4s 1S0 - 4s4p 1P1 are presented. The vibronic spectrum was measured for detunings of the photoassociation laser ranging from 0.6 GHz to 68 GHz with respect to the atomic resonance. In contrast to previous measurements the rotational splitting of the vibrational lines was fully resolved. Full quantum mechanical numerical simulations of the photoassociation spectrum were performed which allowed us to put constraints on the possible range of the calcium scattering length to between 50 a_0 and 300 a_0

    Fine Pathogen Discrimination within the APL1 Gene Family Protects Anopheles gambiae against Human and Rodent Malaria Species

    Genetically controlled resistance of Anopheles gambiae mosquitoes to Plasmodium falciparum is a common trait in the natural population, and a cluster of natural resistance loci were mapped to the Plasmodium-Resistance Island (PRI) of the A. gambiae genome. The APL1 family of leucine-rich repeat (LRR) proteins was highlighted by candidate gene studies in the PRI, and is comprised of paralogs APL1A, APL1B and APL1C that share ≥50% amino acid identity. Here, we present a functional analysis of the joint response of APL1 family members during mosquito infection with human and rodent Plasmodium species. Only paralog APL1A protected A. gambiae against infection with the human malaria parasite P. falciparum from both the field population and in vitro culture. In contrast, only paralog APL1C protected against the rodent malaria parasites P. berghei and P. yoelii. We show that anti-P. falciparum protection is mediated by the Imd/Rel2 pathway, while protection against P. berghei infection was shown to require Toll/Rel1 signaling. Further, only the short Rel2-S isoform and not the long Rel2-F isoform of Rel2 confers protection against P. falciparum. Protection correlates with the transcriptional regulation of APL1A by Rel2-S but not Rel2-F, suggesting that the Rel2-S anti-parasite phenotype results at least in part from its transcriptional control over APL1A. These results indicate that distinct members of the APL1 gene family display a mutually exclusive protective effect against different classes of Plasmodium parasites. It appears that a gene-for-pathogen-class system orients the appropriate host defenses against distinct categories of similar pathogens. It is known that insect innate immune pathways can distinguish between grossly different microbes such as Gram-positive bacteria, Gram-negative bacteria, or fungi, but the function of the APL1 paralogs reveals that mosquito innate immunity possesses a more fine-grained capacity to distinguish between classes of closely related eukaryotic pathogens than has been previously recognized
