    Influence of wet distillers grains diets on beef cattle fecal bacterial community structure

    <p>Abstract</p> <p>Background</p> <p>The high demand for ethanol in the U.S. has generated large stocks of wet distillers grains (DG), a byproduct from the manufacture of ethanol from corn and sorghum grains. Little is known, however, about the potential influence of dietary DG on fecal microbial community structure. A better understanding of the microbial population in beef cattle feces could be an important monitoring tool to facilitate goals of improving nutrient management, increasing animal growth performance and decreasing odors and/or shedding of pathogens. Five diets consisting of a traditional diet fed to finishing beef cattle in the Southern High Plains of Texas-CON (steam-flaked corn control with 0% DG), and four concentrations of DG in the dietary dry matter; 10 C (10% corn-based DG), 5S (5% sorghum-based DG), 10S (10% sorghum DG), and 15S (15% sorghum DG) were fed to steers at the Texas Tech University Burnett Animal Center. Diets were essentially isonitrogenous with a formulated crude protein value of 13.5%.</p> <p>Results</p> <p>Fecal grab samples were obtained from 20 steers (n = 4 per diet) and the barcoded DNA pyrosequencing method was used to generate 127,530 16S operational taxonomic units (OTUs). A total of 24 phyla were observed, distributed amongst all beef cattle on all diets, revealing considerable animal to animal variation, however only six phyla (core set) were observed in all animals regardless of dietary treatment. The average abundance and range of abundance, respectively of the core phyla were as follows: Firmicutes (61%, 19 to 83%), Bacteroidetes (28%, 11 to 63%), Proteobacteria (3%, 0.34 to 17.5%), Tenericutes (0.15%, 0.0 to 0.35%), Nitrospirae (0.11%, 0.03 to 0.22%), and Fusobacteria (0.086%, 0.017 to 0.38%). Feeding DG-based diets resulted in significant shifts in the fecal microbial community structure compared with the traditional CON. Four low abundance phyla significantly responded to dietary treatments: Synergistetes (<it>p </it>= 0.01), WS3 (<it>p </it>= 0.054), Actinobacteria (<it>p </it>= 0.06), and Spirochaetes (<it>p </it>= 0.06).</p> <p>Conclusions</p> <p>This is, to our knowledge, the first study using this method to survey the fecal microbiome of beef cattle fed various concentrations of wet DG. Comparison of our results with other cattle DNA sequencing studies of beef and dairy cattle feces from a variety of geographical locations and different management practices identifies a core set of three phyla shared across all cattle. These three phyla, in order of relative abundance are; Firmicutes, Bacteroidetes, and Proteobacteria. The presence of large animal-to-animal variation in cattle microbiome was noted in our study as well as by others.</p

    We show that asymptotic lifetime-based fluorescence tomography can localize multiple-lifetime targets separated well below the diffuse point spread function of a turbid medium. This is made possible due to a complete diagonalization of the time domain forward problem in the asymptotic limit. We also show that continuous wave or direct time gate approaches to fluorescence tomography are unable to achieve this separation, Assuming that the fluorophores under study have lifetimes Ļ„ n that are longer than the intrinsic diffusion time scales (a condition widely satisfied for biological tissue where j indicates the measurement index (r s , r d pair). Thus, the ATD results in a complete diagonalization of the diffuse fluorescence forward problem with respect to the yield distributions Ī· n r. This should be contrasted with CW, FD, and DTD approaches, where each measurement is related to all the Ī· n values (1) offers a powerful approach to obtain separate reconstructions for each fluorophore in a mixture based on their distinct lifetime

    Mapping spatial dimensions of Wilderness recreation outcomes: a study of overnight users

    Grand Teton National Park (GRTE) is a popular mountain recreation destination which, like many National Park Service (NPS) units, has experienced a significant increase in visitation in recent years, with total visits increasing by 27% between 2014 and 2017 (NPS 2020). Particularly popular within GRTE is the String and Leigh Lakes (SLL) area, which is a favoured alpine destination for numerous day-use recreation activities and also an important starting point for backcountry and overnight recreational users within GRTEā€™s Recommended Wilderness. To better understand the visitor experience of overnight backcountry recreationists in the SLL area, data were collected using novel public participatory geographic information systems (PPGIS) during the summer of 2018. PPGIS data were used to identify the locations in which overnight recreationists experienced positive and negative recreation outcomes. Results indicate that they experience more positive outcomes within the Recommended Wilderness, away from high-density, trailhead-proximate areas outside the Recommended Wilderness. Findings also indicate that overnight users experience crowding and conflict more outside of the Recommended Wilderness than elsewhere on their backcountry trip. While this may seem intuitive, these are some of the first empirical results spatially contextualizing backcountry visitor outcomes in a popular national park. The findings thus provide managers with a visitor experience baseline that can be monitored and adaptively managed in the future

    A theoretical investigation of the low lying electronic structure of poly(p-phenylene vinylene)

    The two-state molecular orbital model of the one-dimensional phenyl-based semiconductors is applied to poly(p-phenylene vinylene). The energies of the low-lying excited states are calculated using the density matrix renormalization group method. Calculations of both the exciton size and the charge gap show that there are both Bu and Ag excitonic levels below the band threshold. The energy of the 1Bu exciton extrapolates to 2.60 eV in the limit of infinite polymers, while the energy of the 2Ag exciton extrapolates to 2.94 eV. The calculated binding energy of the 1Bu exciton is 0.9 eV for a 13 phenylene unit chain and 0.6 eV for an infinite polymer. This is expected to decrease due to solvation effects. The lowest triplet state is calculated to be at ca. 1.6 eV, with the triplet-triplet gap being ca. 1.6 eV. A comparison between theory, and two-photon absorption and electroabsorption is made, leading to a consistent picture of the essential states responsible for most of the third-order nonlinear optical properties. An interpretation of the experimental nonlinear optical spectroscopies suggests an energy difference of ca. 0.4 eV between the vertical energy and ca. 0.8 eV between the relaxed energy, of the 1Bu exciton and the band gap, respectively.Comment: LaTeX, 19 pages, 7 eps figures included using epsf.
