    Coexpression, copurification, crystallization and preliminary X-ray analysis of a complex of ARL2-GTP and PDE delta

    The small GTPase ARL2 (from Mus musculus) and an effector protein, the δ subunit of human cGMP phosphodiesterase (hPDE δ), were coexpressed and copurified from Escherichia coli as a stable complex. Coexpression significantly increased the otherwise low yield of PDE δ production in E. coli. The complex, which contains ARL2 in the activated GTP-bound form, was crystallized in two forms. The first belongs to the monoclinic space group P21, with unit-cell parameters a = 48.1, b = 45.7, c = 74.7 Å, β = 94.0° and one complex (39 kDa) in the asymmetric unit. Cryocooled crystals diffract to 2.3 Å using synchrotron radiation. The micro-focused X-­ray beam at beamline ID13 (ESRF) allowed the use of very small crystals, which helped to overcome twinning and enabled the identification of a molecular-replacement solution. The second form recrystallized from the first one after several months. These crystals belong to the orthorhombic space group P212121, with unit-cell parameters a = 44.5, b = 65.4, c = 104.4 Å and one complex in the asymmetric unit. They diffracted to 1.8 Å using synchrotron radiation

    Design and optimization of electrochemical microreactors for continuous electrosynthesis

    The study focuses on the design and construction, as well as the theoretical and experimental optimization of electrochemical filter press microreactors for the electrosynthesis of molecules with a high added value. The main characteristics of these devices are firstly a high-specific electrochemical area to increase conversion and selectivity, and secondly the shape and size of themicrochannels designed for a uniform residence time distribution of the fluid. A heat exchanger is integrated into the microstructured electrode to rapidly remove (or supply) the heat required in exo- or endothermic reactions. The microreactors designed are used to perform-specific electrosynthesis reactions such as thermodynamically unfavorable reactions (continuous NADH regeneration), or reactions with high enthalpy changes

    Games on graphs with a public signal monitoring

    We study pure Nash equilibria in games on graphs with an imperfect monitoring based on a public signal. In such games, deviations and players responsible for those deviations can be hard to detect and track. We propose a generic epistemic game abstraction, which conveniently allows to represent the knowledge of the players about these deviations, and give a characterization of Nash equilibria in terms of winning strategies in the abstraction. We then use the abstraction to develop algorithms for some payoff functions.Comment: 28 page

    An insight into polarization states of solid-state organic lasers

    The polarization states of lasers are crucial issues both for practical applications and fundamental research. In general, they depend in a combined manner on the properties of the gain material and on the structure of the electromagnetic modes. In this paper, we address this issue in the case of solid-state organic lasers, a technology which enables to vary independently gain and mode properties. Different kinds of resonators are investigated: in-plane micro-resonators with Fabry-Perot, square, pentagon, stadium, disk, and kite shapes, and external vertical resonators. The degree of polarization P is measured in each case. It is shown that although TE modes prevail generally (P>0), kite-shaped micro-laser generates negative values for P, i.e. a flip of the dominant polarization which becomes mostly TM polarized. We at last investigated two degrees of freedom that are available to tailor the polarization of organic lasers, in addition to the pump polarization and the resonator geometry: upon using resonant energy transfer (RET) or upon pumping the laser dye to an higher excited state. We then demonstrate that significantly lower P factors can be obtained.Comment: 12 pages, 12 figure

    Constrained Dynamics of Universally Coupled Massive Spin 2-spin 0 Gravities

    The 2-parameter family of massive variants of Einstein's gravity (on a Minkowski background) found by Ogievetsky and Polubarinov by excluding lower spins can also be derived using universal coupling. A Dirac-Bergmann constrained dynamics analysis seems not to have been presented for these theories, the Freund-Maheshwari-Schonberg special case, or any other massive gravity beyond the linear level treated by Marzban, Whiting and van Dam. Here the Dirac-Bergmann apparatus is applied to these theories. A few remarks are made on the question of positive energy. Being bimetric, massive gravities have a causality puzzle, but it appears soluble by the introduction and judicious use of gauge freedom.Comment: 6 pages; Talk given at QG05, Cala Gonone (Italy), September 200

    Type I D-branes in an H-flux and twisted KO-theory

    Witten has argued that charges of Type I D-branes in the presence of an H-flux, take values in twisted KO-theory. We begin with the study of real bundle gerbes and their holonomy. We then introduce the notion of real bundle gerbe KO-theory which we establish is a geometric realization of twisted KO-theory.
