    Evolutionary Expansion of Nematode-Specific Glycine-Rich Secreted Peptides

    A genome‐wide survey across 10 species from algae Guillardia theta to mammals revealed that Caenorhabditis elegans and Caenorhabditis briggsae acquired a large number of glycine‐rich secreted peptides (GRSPs, 110 GRSPs in C. elegans and 93 in C. briggsae) during evolution in this study. Chromosomal mapping indicated that most GRSPs were clustered on their genomes [103 (93.64%) in C. elegans and 82 (88.17%) in C. briggsae]. Totally, there are 18 GRSPs cluster units in C. elegans and 13 in C. briggsae. Except for four C. elegans where GRSP clusters lacking matching clusters in C. briggsae, all other GRSP clusters had its corresponding orthologous clusters between the two nematodes. Using eight transcriptomic datasets of Affmyetrix microarray, genome‐wide association studies identified many co‐expressed GRSPs clusters after C. elegans infections. Highly homologous coding sequences and conserved exon‐intron organizations indicated that GRSP tight clusters might have originated from local DNA duplications. The conserved synteny blocks of GRSP clusters between their genomes, the co‐expressed GRSPs clusters after C. elegans infections, and a strong purifying selection of protein‐coding sequences suggested evolutionary constraint acting on C. elegans to ensure that C. elegans could rapidly launch and fulfill systematic responses against infections by co‐expression, co‐regulation, and co‐functionality of GRSP clusters

    Rapid Invasion of Spartina alterniflora in the Coastal Zone of Mainland China: New Observations from Landsat OLI Images

    Plant invasion imposes significant threats to biodiversity and ecosystem function. Thus, monitoring the spatial pattern of invasive plants is vital for effective ecosystem management. Spartina alterniflora (S. alterniflora) has been one of the most prevalent invasive plants along the China coast, and its spread has had severe ecological consequences. Here, we provide new observation from Landsat operational land imager (OLI) images. Specifically, 43 Landsat-8 OLI images from 2014 to 2016, a combination of object-based image analysis (OBIA) and support vector machine (SVM) methods, and field surveys covering the whole coast were used to construct an up-to-date dataset for 2015 and investigate the spatial variability of S. alterniflora in the coastal zone of mainland China. The classification results achieved good estimation, with a kappa coefficient of 0.86 and 96% overall accuracy. Our results revealed that there was approximately 545.80 km2 of S. alterniflora distributed in the coastal zone of mainland China in 2015, from Hebei to Guangxi provinces. Nearly 92% of the total area of S. alterniflora was distributed within four provinces: Jiangsu, Shanghai, Zhejiang, and Fujian. Seven national nature reserves invaded by S. alterniflora encompassed approximately one-third (174.35 km2) of the total area of S. alterniflora over mainland China. The Yancheng National Nature Reserve exhibited the largest area of S. alterniflora (115.62 km2) among the reserves. Given the rapid and extensive expansion of S. alterniflora in the 40 years since its introduction and its various ecological effects, geospatially varied responding decisions are needed to promote sustainable coastal ecosystems

    Spatial Expansion and Soil Organic Carbon Storage Changes of Croplands in the Sanjiang Plain, China

    Soil is the largest pool of terrestrial organic carbon in the biosphere and interacts strongly with the atmosphere, climate and land cover. Remote sensing (RS) and geographic information systems (GIS) were used to study the spatio-temporal dynamics of croplands and soil organic carbon density (SOCD) in the Sanjiang Plain, to estimate soil organic carbon (SOC) storage. Results show that croplands increased with 10,600.68 km2 from 1992 to 2012 in the Sanjiang Plain. Area of 13,959.43 km2 of dry farmlands were converted into paddy fields. Cropland SOC storage is estimated to be 1.29 ± 0.27 Pg C (1 Pg = 103 Tg = 1015 g) in 2012. Although the mean value of SOCD for croplands decreased from 1992 to 2012, the SOC storage of croplands in the top 1 m in the Sanjiang Plain increased by 70 Tg C (1220 to 1290). This is attributed to the area increases of cropland. The SOCD of paddy fields was higher and decreased more slowly than that of dry farmlands from 1992 to 2012. Conversion between dry farmlands and paddy fields and the agricultural reclamation from natural land-use types significantly affect the spatio-temporal patterns of cropland SOCD in the Sanjiang Plain. Regions with higher and lower SOCD values move northeast and westward, respectively, which is almost consistent with the movement direction of centroids for paddy fields and dry farmlands in the study area. Therefore, these results were verified. SOC storages in dry farmlands decreased by 17.5 Tg·year−1 from 1992 to 2012, whilst paddy fields increased by 21.0 Tg·C·year−1

    AST1306, A Novel Irreversible Inhibitor of the Epidermal Growth Factor Receptor 1 and 2, Exhibits Antitumor Activity Both In Vitro and In Vivo

    Despite the initial response to the reversible, ATP-competitive quinazoline inhibitors that target ErbB-family, such a subset of cancer patients almost invariably develop resistance. Recent studies have provided compelling evidence that irreversible ErbB inhibitors have the potential to override this resistance. Here, we found that AST1306, a novel anilino-quinazoline compound, inhibited the enzymatic activities of wild-type epidermal growth factor receptor (EGFR) and ErbB2 as well as EGFR resistant mutant in both cell-free and cell-based systems. Importantly, AST1306 functions as an irreversible inhibitor, most likely through covalent interaction with Cys797 and Cys805 in the catalytic domains of EGFR and ErbB2, respectively. Further studies showed that AST1306 inactivated pathways downstream of these receptors and thereby inhibited the proliferation of a panel of cancer cell lines. Although the activities of EGFR and ErbB2 were similarly sensitive to AST1306, ErbB2-overexpressing cell lines consistently exhibited more sensitivity to AST1306 antiproliferative effects. Consistent with this, knockdown of ErbB2, but not EGFR, decreased the sensitivity of SK-OV-3 cells to AST1306. In vivo, AST1306 potently suppressed tumor growth in ErbB2-overexpressing adenocarcinoma xenograft and FVB-2/Nneu transgenic breast cancer mouse models, but weakly inhibited the growth of EGFR-overexpressing tumor xenografts. Tumor growth inhibition induced by a single dose of AST1306 in the SK-OV-3 xenograft model was accompanied by a rapid (within 2 h) and sustained (≥24 h) inhibition of both EGFR and ErbB2, consistent with an irreversible inhibition mechanism. Taken together, these results establish AST1306 as a selective, irreversible ErbB2 and EGFR inhibitor whose growth-inhibitory effects are more potent in ErbB2-overexpressing cells

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

In view of problem that maintenance mode of current coal cleaning and processing equipment cannot accurately and effectively identify early fault and potential threat, a predictive maintenance system of coal cleaning and processing equipments was designed. The system is mainly composed of function module of basic information management, function module of real-time monitoring of equipment condition, function module of fault simulation, function module of predictive maintenance and decision, four modules are complement with each other and coordinates effect, which can evaluate running status of current equipment, and forecast development trend of fault, so as to realize the predictive maintenance and health diagnosis of the equipments

    Research on Direct Drive Technology of the Permanent Magnet Synchronous Motor for Urban Rail Vehicles

    Compared with the traditional gear reduction mechanism of the asynchronous motor, the permanent magnet synchronous motor (PMSM) direct drive technology has obvious advantages and has attracted increasing attention. We analyzed the advantages of the PMSM direct drive technology and put forward the hollow shaft structure of the coupling to reduce the impact force of the wheel and rail, additional noise, vibration, and loss. We deeply studied the topology and mathematical model of the PMSM. To keep the maximum running speed unchanged, it is necessary to conduct a flux weakening control above the rated speed to increase the speed regulation range of the PMSM. To prevent frequent switching jitter at the transition point and smooth the transition between different weak magnetic fields, we designed a new flux-weakening controller to control the current. Simulation results have been presented to verify the proposed approach efficiency. Taking the PMSM system train on Suzhou Metro line 3 as an example, the on-site energy consumption test was carried out. The energy consumption of the PMSM direct drive and asynchronous motor gear mechanism was compared. The energy consumption of the PMSM direct drive is increased by 16.50%. The flexible plate coupling can alleviate the adverse effects of vibration and improve the reliability of train operation. Finally, we put forward the technical difficulties and challenges of the PMSM direct drive technology. We need to optimize the mass distribution of the direct drive bogie to improve the dynamic performance of the bogie, the heat dissipation conditions, and the accurate control of the PMSM. It will have reference value for the development of the PMSM direct drive in the future

    Rostral-Caudal Effect of Cervical Magnetic Stimulation on the Diaphragm Motor Evoked Potential after Cervical Spinal Cord Contusion in the Rat

    International audienceThe present study was designed to investigate the rostral-caudal effect of spinal magnetic stimulation on diaphragmatic motor-evoked potentials after cervical spinal cord injury. The diaphragm electromyogram was recorded in rats that received a laminectomy or a left midcervical contusion at the acute (1 day), subchronic (2 weeks), or chronic (8 weeks) injury stages. The center of a figure-eight coil was placed at 30 mm lateral to bregma on the left side, and the effect of magnetic stimulation was evaluated by stimulating the rostral, middle, and caudal cervical regions in spontaneously breathing rats. The results demonstrated that cervical magnetic stimulation induced intensity-dependent motor-evoked potentials in the bilateral diaphragm in both uninjured and contused rats; however, the left diaphragm exhibited a higher amplitude and earlier onset than the right diaphragm. Moreover, the intensity-response curve was shifted upward in the rostral-To-caudal direction of magnetic stimulation, suggesting that caudal cervical magnetic stimulation produced more robust diaphragmatic motor-evoked potentials compared with rostral cervical magnetic stimulation. Interestingly, the diaphragmatic motor-evoked potentials were similar between uninjured and contused rats during cervical magnetic stimulation despite weaker inspiratory diaphragmatic activity in contused rats. In addition, in contused animals but not uninjured animals, diaphragmatic motor-evoked potential amplitudes were greater at the chronic stage than during earlier injury stages. These results demonstrated that cervical magnetic stimulation can excite the residual phrenic motor circuit to activate the diaphragm in the presence of a significant lesion in the cervical spinal cord. These findings indicate that this non-invasive approach is effective for modulating diaphragmatic excitability after cervical spinal cord injury

    Gluing Techniques on Bond Performance and Mechanical Properties of Cross-Laminated Timber (CLT) Made from Larix kaempferi

    Previous studies have proved that Larix kaempferi is a good material for preparing cross-laminated timber (CLT), but under bending shear stress, CLT made by Larix kaempferi is prone to the phenomenon of bonding face cracking, which seriously affects the shear performance of CLT. To solve this problem, this paper took Larix kaempferi as raw material, conducted experiments on the surface sanding conditions, gluing pressure and adhesive types of sawing timber, and explored the influence of these three factors on the bonding quality of CLT. The microscopic characteristics of the bonding layer were further studied. The results showed that for Larix kaempferi with a density of 0.68 g/cm3 used in this experiment, a high bonding pressure is required. Among the three cold curing adhesives selected in the experiment, emulsion polymer isocyanate (EPI) adhesive needs 1.5 MPa bonding pressure to ensure the bonding quality, while for polyurethane (PUR) and phenol resorcinol formaldehyde (PRF), 1.2 MPa can meet the need of adhesive pressure. This is concerned with the permeability of different adhesives under different pressures. The microscopic results of the bonding layer show that EPI adhesives have poor permeability, so it requires high bonding pressure. The influence of sanding surface of different sand-belt on block shear strength (BSS) and wood failure percentage (WFP) is not obvious, while the durability of bonding layer is better when sanding mesh number is 100. Hence, a high pressure should be used for CLT industrial production when the laminate density is higher, especially when the adhesive has poor permeability. Reasonable sanding surface treatment can be used in laminate surface treatment to improve the durability of CLT
