Cancer Cachexia: Traditional Therapies and Novel Molecular Mechanism-Based Approaches to Treatment
The complex syndrome of cancer cachexia (CC) that occurs in 50% to 80% cancer patients has been identified as an independent predictor of shorter survival and increased risk of treatment failure and toxicity, contributing to the mortality and morbidity in this population. CC is a pathological state including a symptom cluster of loss of muscle (skeletal and visceral) and fat, manifested in the cardinal feature of emaciation, weakness affecting functional status, impaired immune system, and metabolic dysfunction. The most prominent feature of CC is its non-responsiveness to traditional treatment approaches; randomized clinical trials with appetite stimulants, 5-HT3 antagonists, nutrient supplementation, and Cox-2 inhibitors all have failed to demonstrate success in reversing the metabolic abnormalities seen in CC. Interventions based on a clear understanding of the mechanism of CC, using validated markers relevant to the underlying metabolic abnormalities implicated in CC are much needed. Although the etiopathogenesis of CC is poorly understood, studies have proposed that NFkB is upregulated in CC, modulating immune and inflammatory responses induce the cellular breakdown of muscle, resulting in sarcopenia. Several recent laboratory studies have shown that n-3 fatty acid may attenuate protein degradation, potentially by preventing NFkB accumulation in the nucleus, preventing the degradation of muscle proteins. However, clinical trials to date have produced mixed results potentially attributed to timing of interventions (end stage) and utilizing outcome markers such as weight which is confounded by hydration, cytotoxic therapies, and serum cytokines. We propose that selective targeting of proteasome activity with a standardized dose of omega-3-acid ethyl esters, administered to cancer patients diagnosed with early stage CC, in addition to a standard intervention with nutritionally adequate diet and appetite stimulants, will alter metabolic abnormalities by downregulating NFkB, preventing the breakdown of myofibrillar proteins and resulting in increasing serum protein markers, lean body mass, and functional status
QuickNGS elevates Next-Generation Sequencing data analysis to a new level of automation
BACKGROUND: Next-Generation Sequencing (NGS) has emerged as a widely used tool in molecular biology. While time and cost for the sequencing itself are decreasing, the analysis of the massive amounts of data remains challenging. Since multiple algorithmic approaches for the basic data analysis have been developed, there is now an increasing need to efficiently use these tools to obtain results in reasonable time. RESULTS: We have developed QuickNGS, a new workflow system for laboratories with the need to analyze data from multiple NGS projects at a time. QuickNGS takes advantage of parallel computing resources, a comprehensive back-end database, and a careful selection of previously published algorithmic approaches to build fully automated data analysis workflows. We demonstrate the efficiency of our new software by a comprehensive analysis of 10 RNA-Seq samples which we can finish in only a few minutes of hands-on time. The approach we have taken is suitable to process even much larger numbers of samples and multiple projects at a time. CONCLUSION: Our approach considerably reduces the barriers that still limit the usability of the powerful NGS technology and finally decreases the time to be spent before proceeding to further downstream analysis and interpretation of the data. What are the competences in information system required by managers? Curriculum development for management and public administration degrees
[EN] This paper analyzes the competences required by executives to manage information
system, and consequently, the competences that must define the information system
subjects in non-technical degrees, degrees, such as Public Administration or Business
Management. This work reviews the literature about business managers competences
on Information Technologies (IT) and compares the theory with the traditional body
of knowledge about information systems taught at business schools. By analyzing the
executives function, their role in the information system management, and, above,
all the importance of their decisions in the effective integration of IT in business

Identification of lung cancer with high sensitivity and specificity by blood testing
<p>Abstract</p> <p>Background</p> <p>Lung cancer is a very frequent and lethal tumor with an identifiable risk population. Cytological analysis and chest X-ray failed to reduce mortality, and CT screenings are still controversially discussed. Recent studies provided first evidence for the potential usefulness of autoantigens as markers for lung cancer.</p> <p>Methods</p> <p>We used extended panels of arrayed antigens and determined autoantibody signatures of sera from patients with different kinds of lung cancer, different common non-tumor lung pathologies, and controls without any lung disease by a newly developed computer aided image analysis procedure. The resulting signatures were classified using linear kernel Support Vector Machines and 10-fold cross-validation.</p> <p>Results</p> <p>The novel approach allowed for discriminating lung cancer patients from controls without any lung disease with a specificity of 97.0%, a sensitivity of 97.9%, and an accuracy of 97.6%. The classification of stage IA/IB tumors and controls yielded a specificity of 97.6%, a sensitivity of 75.9%, and an accuracy of 92.9%. The discrimination of lung cancer patients from patients with non-tumor lung pathologies reached an accuracy of 88.5%.</p> <p>Conclusion</p> <p>We were able to separate lung cancer patients from subjects without any lung disease with high accuracy. Furthermore, lung cancer patients could be seprated from patients with other non-tumor lung diseases. These results provide clear evidence that blood-based tests open new avenues for the early diagnosis of lung cancer.</p
Covariation in Plant Functional Traits and Soil Fertility within Two Species-Rich Forests
The distribution of plant species along environmental gradients is expected to be predictable based on organismal function. Plant functional trait research has shown that trait values generally vary predictably along broad-scale climatic and soil gradients. This work has also demonstrated that at any one point along these gradients there is a large amount of interspecific trait variation. The present research proposes that this variation may be explained by the local-scale sorting of traits along soil fertility and acidity axes. Specifically, we predicted that trait values associated with high resource acquisition and growth rates would be found on soils that are more fertile and less acidic. We tested the expected relationships at the species-level and quadrat-level (20×20 m) using two large forest plots in Panama and China that contain over 450 species combined. Predicted relationships between leaf area and wood density and soil fertility were supported in some instances, but the majority of the predicted relationships were rejected. Alternative resource axes, such as light gradients, therefore likely play a larger role in determining the interspecific variability in plant functional traits in the two forests studied
The Supermassive Black Hole at the Galactic Center
The inner few parsecs at the Galactic Center have come under intense scrutiny
in recent years, in part due to the exciting broad-band observations of this
region, but also because of the growing interest from theorists motivated to
study the physics of black hole accretion, magnetized gas dynamics and unusual
star formation. The Galactic Center is now known to contain arguably the most
compelling supermassive black hole candidate, weighing in at a little over 2.6
million suns. Its interaction with the nearby environment, comprised of
clusters of evolved and young stars, a molecular dusty ring, ionized gas
streamers, diffuse hot gas, and a hypernova remnant, is providing a wealth of
accretion phenomenology and high-energy processes for detailed modeling. In
this review, we summarize the latest observational results, and focus on the
physical interpretation of the most intriguing object in this region---the
compact radio source Sgr A*, thought to be the radiative manifestation of the

Combining functional weed ecology and crop stable isotope ratios to identify cultivation intensity: a comparison of cereal production regimes in Haute Provence, France and Asturias, Spain
This investigation combines two independent
methods of identifying crop growing conditions and husbandry
practices—functional weed ecology and crop stable
carbon and nitrogen isotope analysis—in order to assess
their potential for inferring the intensity of past cereal
production systems using archaeobotanical assemblages.
Present-day organic cereal farming in Haute Provence,
France features crop varieties adapted to low-nutrient soils
managed through crop rotation, with little to no manuring.
Weed quadrat survey of 60 crop field transects in this region
revealed that floristic variation primarily reflects
geographical differences. Functional ecological weed data
clearly distinguish the Provence fields from those surveyed
in a previous study of intensively managed spelt wheat in
Asturias, north-western Spain: as expected, weed ecological
data reflect higher soil fertility and disturbance in
Asturias. Similarly, crop stable nitrogen isotope values
distinguish between intensive manuring in Asturias and
long-term cultivation with minimal manuring in Haute
Provence. The new model of cereal cultivation intensity
based on weed ecology and crop isotope values in Haute
Provence and Asturias was tested through application to
two other present-day regimes, successfully identifying a
high-intensity regime in the Sighisoara region, Romania,
and low-intensity production in Kastamonu, Turkey. Application
of this new model to Neolithic archaeobotanical
assemblages in central Europe suggests that early farming
tended to be intensive, and likely incorporated manuring,
but also exhibited considerable variation, providing a finer
grained understanding of cultivation intensity than previously
available
Brief review on systematic hypothermia for the protection of central nervous system during aortic arch surgery: a double-sword tool?
Antegrade selective cerebral perfusion in conjunction with hypothermia attenuate postoperative neurological injury, which in turn still remains the main cause of mortality and morbidity following aortic arch surgery. Hypothermic circulatory arrest however could be a useful tool during arch surgery, surgery for chronic thromboembolic disease, air on the arterial line during CPB, during cavotomy for extraction of renal cell carcinoma with level IV extension, or when dealing with difficult trauma to the SVC or IVC. Cerebral protective effects with hypothermic procedures including inhibition of neuron excitation, and discharge of excitable amino acids, and thereby, prevention of an increase in intercellular calcium ions, hyperoxidation of lipids in cell membranes, and free radical production
