Personal dataThis thesis is a contribution to the environmental risk assessment of
explosives for the soil fauna. As typical representatives the collembola F.
candida and the enchytraeid E. crypticus were chosen. For both test organisms
standardised tests have been established. With the mortality, the reproduction
and the choice test the following issues have been investigated:
-Toxicity in the standard soil Lufa 2.2 of the explosives TNT, Hexyl, Hexogen (RDX) and Octogen (HMX) as well as of Triaminotoluene (TAT), the end product of reductive microbial degradation of TNT
-Influence of the soil type on the toxicity of TNT in four uncontaminated soil materials with different contents of clay and organic carbon
-Influence of ageing on TNT with the collembola-biotest in the standard soil Lufa 2.2
-Toxicity of contaminated soils on both test organisms
-Success of different remediation processes.
The toxicity of the pure substances decreases from TNT to Hexyl for both test
organisms. TAT is toxic for E. crypticus, but has no effect on F. candida.
Hexogen and Octogen are not toxic. The reproduction test is generally more
sensitive than the mortality test. In the choice test only the enchytraeid
reacts at even lower concentrations than in the reproduction test.
The reference soils differ in their content of clay and organic carbon from
the standard soil Lufa 2.2. The toxicity of TNT in the soil materials is
correlated with their contents of clay and organic matter. Both species show
soil specific reproduction rates in the uncontaminated reference soils.
The investigation of the ageing process reveals a time dependent decrease of
the toxicity of TNT. The decrease is greater at higher temperatures than at
lower. This is probably the result of the temperature dependence of the
microbial activity.
The toxicity of TNT in contaminated soil materials was investigated for six
soils from four different sites. It varies with the content of clay and
organic matter of the soil materials.
With the biotests it could be shown that remediation processes can improve the
habitat function of soil materials. Processes which use autochthon
microorganisms, improve it. However, it was observed that the the habitat
function became worse with white rot fungi, even though TNT was completely
transformed.Diese Arbeit untersucht die ökotoxikologischen Auswirkungen von
sprengstofftypischen Verbindungen auf die Lebensraumfunktion des Bodens. Als
typische Vertreter fĂŒr die Bodenfauna wurden die Collembole F. candida und die
Enchytraee E. crypticus ausgewĂ€hlt, fĂŒr die standardisierte Testverfahren
entwickelt sind. Mit Hilfe von MortalitÀts-, Reproduktions- und
Verhaltenstests wurden die folgenden Fragestellungen untersucht:
-ToxizitÀt der Sprengstoffe TNT, Hexyl, Hexogen (RDX) und Octogen (HMX), sowie von Triaminotoluol (TAT), dem Endprodukt des reduktiven mikrobiellen Abbaus von TNT, im Standardboden Lufa 2.2
-Einfluà der Bodenart auf die ToxizitÀt von TNT mit vier unbelasteten Referenzböden unterschiedlichen Ton- und organischen Kohlenstoffgehalts
-EinfluĂ der Alterung von TNT im Standardboden Lufa 2.2 im Collembolen-Test
-ToxizitÀt von nutzungsbedingt belasteten Böden auf beide Testorganismen
-Erfolg verschiedener Sanierungsverfahren.
Von den Reinsubstanzen nimmt die ToxizitĂ€t fĂŒr beide Testorganismen in der
Reihenfolge TNT zu Hexyl ab. TAT ist fĂŒr E. crypticus noch toxisch, wirkt
dagegen auf F. candida nicht. Hexogen und Octogen sind untoxisch. Der
Reproduktionstest ist generell sensitiver als der MortalitÀtstest. Im
Verhaltenstest reagiert nur die Enchytraee und zwar schon bei niederen
Konzentrationen als im Reproduktionstest.
Die Referenzböden unterscheiden sich in ihren Gehalten an Ton und organischen
Kohlenstoff von dem Standardboden Lufa 2.2. Die ToxizitĂ€t fĂŒr TNT in den
Bodenmaterialen korreliert mit ihrem Gehalt an Ton und organischer Materie.
Beide Arten zeigen in den unbelasteten Referenzböden bodenspezifische
Reproduktionsraten.
In den Versuchen zur Alterung wurde eine zeitabhÀngige Abnahme der ToxizitÀt
von TNT nachgewiesen. Die ToxizitÀtsabnahme ist bei höheren Temperaturen
gröĂer als bei niedrigen. Dies ist vermutlich auf die TemperaturabhĂ€ngigkeit
der mikrobiellen AktivitĂ€t zurĂŒckzufĂŒhren.
Die ToxizitĂ€t von TNT in nutzungsbedingt belasteten Böden wurde fĂŒr sechs
Böden von vier verschiedenen Standorten untersucht. Sie variiert mit dem
Gehalt der Böden an Ton und organischer Materie.
Mit diesen Biotests konnte gezeigt werden, daĂ Sanierungsverfahren die
HabitatqualitÀt des Bodenmaterials verbessern. Verfahren, die auf autochthone
Mikroorganismen zurĂŒckgreifen, verbessern sie nachhaltig. Bei einem Verfahren
mit WeiĂfĂ€ulnispilzen, daĂ zu einer vollstĂ€ndigen Transformierung von TNT
fĂŒhrte, wurde sie jedoch verschlechtert. Dagegen reduzierte eine BodenwĂ€sche
sowohl die ToxizitÀt der Sand- als auch der Feinkornfraktion
The b alleles of U. maydis, whose combinations program pathogenic development, code for polypeptides containing a homeodomain-related motif
AbstractU. maydis is a fungal pathogen of corn with two forms: one is yeast-like and nonpathogenic; the other is filamentous and pathogenic. The b locus, with 25 different alleles, regulates this dimorphism: any combination of two different alleles triggers pathogenic development, whereas the presence of identical alleles results in the yeast-like form. We have cloned four b alleles (b1, b2, b3, and b4) and show that the b locus contains a single open reading frame (ORF) of 410 amino acids with a variable N-terminal region and a highly conserved C-terminal region (60% and 93% identity, respectively). Mutational analysis confirms that this ORF is responsible for b activity. The b polypeptides appear to be DNA binding proteins because they contain a motif related to the homeodomain in their constant region. We propose that combinatorial interactions between b polypeptides generate regulatory proteins that determine the developmental program of the fungus
A novel multiplex detection array revealed systemic complement activation in oral squamous cell carcinoma
Oral squamous cell carcinoma (OSCC) is one of the most common tumors within the oral cavity. Early diagnosis and prognosis tools are urgently needed. This study aimed to investigate the activation of the complement system in OSCC patients as potential biomarker. Therefore, an innovative complement activation array was developed. Characterized antibodies detecting the complement activation specific epitopes C3a, C5a and sC5b-9 along with control antibodies were implemented into a suspension bead array. Human serum from a healthy (n = 46) and OSCC patient (n = 57) cohort were used to investigate the role of complement activation in oral tumor progression. The novel multiplex assay detected C3a, C5a and sC5b-9 from a minimal sample volume of human tears, aqueous humor and blood samples. Limits of detection were 0.04 ng/mL for C3a, 0.03 ng/mL for C5a and 18.9 ng/mL for sC5b-9, respectively. Biological cut-off levels guaranteed specific detections from serum. The mean serum concentration of a healthy control cohort was 680 ng/mL C3a, 70 ng/mL C5a and 2247 ng/mL sC5b-9, respectively. The assay showed an intra-assay precision of 2.9-6.4% and an inter-assay precision of 9.2-18.2%. Increased systemic C5a (p < 0.0001) and sC5b-9 (p = 0.01) concentrations in OSCC patients were determined using the validated multiplex complement assay. Higher C5a concentrations correlated with tumor differentiation and OSCC extension state. Systemic sC5b-9 determination provided a novel biomarker for infiltrating tumor growth and C3a levels were associated with local tumor spreading. Our study suggests that systemic complement activation levels in OSCC patients may be useful to assess disease progression
Massenspektrometrische und gelelektrophoretische Analyse altersabhÀngiger VerÀnderungen des mitochondrialen Proteoms von Modellorganismen
Der Prozess des Alterns betrifft alle Menschen, jedoch sind bisher nur seine Auswirkungen gut erforscht wĂ€hrend ĂŒber die eigentlichen Ursachen des Alterns bisher noch sehr wenig bekannt ist. Gesichert ist lediglich, dass es sich um einen sehr komplexen Vorgang handelt, bei dem genetische, biochemische und zellphysiologische Defekte und VerĂ€nderungen ebenso eine Rolle spielen wie natĂŒrliche Abnutzungserscheinungen und Ă€uĂere EinflĂŒsse. Altern beobachtet man nicht nur beim Menschen, sondern auch bei Tieren, Pflanzen und bestimmten Pilzarten. Einige Mechanismen des Alterns spielen sowohl beim Menschen als auch bei bestimmten Modellorganismen eine Rolle, so dass anhand letzterer gewonnene Ergebnisse gegebenenfalls auf den Alternsprozess des Menschen ĂŒbertragbar sind. Im Rahmen der vorliegenden Arbeit wurde der Alternsprozess anhand des filamentösen Pilzes Podospora anserina, dem extrem kurzlebigen TĂŒrkisen PrachtgrundkĂ€rpfling Nothobranchius furzeri sowie der SchwarzbĂ€uchigen Taufliege Drosophila melanogaster und dem Fadenwurm Caenorhabditis elegans untersucht. Augenmerk lag dabei auf VerĂ€nderungen des mitochondrialen Proteoms, insbesondere der Menge und Zusammensetzung von Komplexen und Superkomplexen der oxidativen Phosphorylierung (OxPhos) sowie altersassoziiert auftretenden posttranslationalen Modifikationen.
FĂŒr Podospora anserina wurde fĂŒr vier verschiedene Altersstufen eine detaillierte Quantifizierung der Komplexe und Superkomplexe der OxPhos durchgefĂŒhrt wobei auch das Monomer-Dimer-VerhĂ€ltnis der ATP-Synthase, die Menge an freiem F1-Teil der ATP-Synthase sowie die Menge der verschiedenen Superkomplexe und die prozentuale Verteilung der einzelnen Komplexe auf ihre individuellen Formen sowie die Superkomplexe bestimmt wurden. Dies erfolgte durch die Kombination von 2-dimensionaler (2D) Gelelektrophorese, FluoreszenzfĂ€rbung und softwarebasierter Quantifizierung. Da es sich bei den Komplexen der OxPhos um Membranproteine handelt wurde fĂŒr die Auftrennung in der ersten Dimension die blau-native Gelelektrophorese (BN-PAGE) verwendet. Weiterhin kamen verschiedene massenspektrometrische Methoden zur Analyse altersabhĂ€ngiger posttranslationaler Modifikationen der ATP-Synthase aus Podospora anserina zum Einsatz. Bei Caenorhabditis elegans wurde das mitochondriale Proteom des Wildtyps sowie der daf-2(e1370)-Mutante und bei Drosophila melanogaster das Proteom des Wildtyps und einer Hsp22-Ăberexpressionsmutante mittels gelelektrophoretischer Methoden untersucht. Die Analysen des extrem kurzlebigen TĂŒrkisen PrachtgrundkĂ€rpflings Nothobranchius furzeri beziehen sich auf eine mögliche Verlangsamung des Alternsprozesses. Hier wurden Tiere mit unterschiedlichem ErnĂ€hrungsstatus (ad libitum gefĂŒtterte Tiere sowie Fische mit kurzzeitiger oder lebenslanger Kalorienrestriktion (CR)) sowie Fische denen das natĂŒrliche Antioxidans Resveratrol verabreicht worden war mittels gelelektrophoretischer Methoden analysiert
