    Genotype and Genotype X Environment Interaction Effects on Forage Yield and Quality of Crested Wheatgrasses

    Genotype, environment, and genotype X environment interaction effects for forage yield and quality of the crested wheatgrasses (Agropyron cristatum) and (A. desertorum) were studied using an array of 42 crested wheatgrass introductions, experimental lines, and released cultivars that were selected to be representative of the mix of germplasm available to a breeder. They were grown in two environments, Lincoln and Alliance, Nebr., that differ markedly in climate. Forage yield and forage quality as measured by in vitro dry matter digestibility (IVDMD) and percent protein were determined in both locations in 1980 and 1981. In the combined analyses over years and locations, there were significant differences among strains or genotypes and locations for first-harvest forage yield, IVDMD, and protein. Strain X location interaction effects were significant only for first-harvest forage yield and for this trait its variance component was only 0.3 as large as the variance component for strains. Strain X year interaction effects were not significant. Spearmen correlations, used to show relative ranking of the strains at the two locations, were moderate and positive for all first-cut traits. The results indicate that existing genetic variability in forage yield and quality should permit the development of improved crested wheatgrasses and that at least in the initial stages of a breeding program, selection for these traits could be done in a single location and year

    Evaluation of Crested Wheatgrass Introductions for Forage Yield and Quality

    Crested wheatgrass, Agropyron cristatum and A. desertorum, are among the most important cool-season forage grasses in the United States and Canada, particularly for reseeding arid range sites. Further improvement in this grass by breeding depends on identifying sources of genetic variability for forage yield and quality. Foreign introductions are an obvious source of genetic variation since crested wheatgrasses are introduced species. In this study 38 accessions (PI lines) and 8 Nebraska experimental lines were evaluated for forage quality as measured by in vitro dry matter digestibility (IVDMD) and protein content and for forage yield. The cultivars \u27Ruff\u27 and \u27Nordan\u27 and two clonal lines were included as checks. The strains were evaluated at Lincoln and Alliance, NE., which differ markedly in climate. There were large differences among strains evaluated for all traits including first- and second-cut forage yield, IVDMD, protein content, heading date, height and first year basal spread. Utilization of PI 369167, PI 369170, Ruff, and Nordan in a breeding program could result in crested wheatgrasses with improved forage yield and quality

    Dynamical instabilities of Bose-Einstein condensates at the band-edge in one-dimensional optical lattices

    We report on experiments that demonstrate dynamical instability in a Bose-Einstein condensate at the band-edge of a one-dimensional optical lattice. The instability manifests as rapid depletion of the condensate and conversion to a thermal cloud. We consider the collisional processes that can occur in such a system, and perform numerical modeling of the experiments using both a mean-field and beyond mean-field approach. We compare our numerical results to the experimental data, and find that the Gross-Pitaevskii equation is not able to describe this experiment. Our beyond mean-field approach, known as the truncated Wigner method, allows us to make quantitative predictions for the processes of parametric growth and thermalization that are observed in the laboratory, and we find good agreement with the experimental results.Comment: v2: Added several reference

    Social parasitism by honeybee workers (Apis mellifera capensis Escholtz): host finding and resistance of hybrid host colonies

    We studied possible host finding and resistance mechanisms of host colonies in the context of social parasitism by Cape honeybee (Apis mellifera capensis) workers. Workers often join neighboring colonies by drifting, but long-range drifting (dispersal) to colonies far away from the maternal nests also rarely occurs. We tested the impact of queenstate and taxon of mother and host colonies on drifting and dispersing of workers and on the hosting of these workers in A. m. capensis, A. m. scutellata, and their natural hybrids. Workers were paint-marked according to colony and reintroduced into their queenright or queenless mother colonies. After 10 days, 579 out of 12,034 labeled workers were recaptured in foreign colonies. We found that drifting and dispersing represent different behaviors, which were differently affected by taxon and queenstate of both mother and host colonies. Hybrid workers drifted more often than A. m. capensis and A. m. scutellata. However, A. m. capensis workers dispersed more often than A. m. scutellata and the hybrids combined, and A. m. scutellata workers also dispersed more frequently than the hybrids. Dispersers from queenright A. m. capensis colonies were more often found in queenless host colonies and vice versa, indicating active host searching and/or a queenstate-discriminating guarding mechanism. Our data show that A. m. capensis workers disperse significantly more often than other races of A. mellifera, suggesting that dispersing represents a host finding mechanism. The lack of dispersal in hybrids and different hosting mechanisms of foreign workers by hybrid colonies may also be responsible for the stability of the natural hybrid zone between A. m. capensis and A. m. scutellata

    The importance of high-throughput cell separation technologies for genomics/proteomics-based clinical diagnostics

    Gene expression microarray analyses of mixtures of cells approximate a weighted average of the gene expression profiles (GEPs) of each cell type according to its relative abundance in the overall cell sample being analyzed. If the targeted subpopulation of cells is in the minority, or the expected perturbations are marginal, then such changes will be masked by the GEP of the normal/unaffected cells. We show that the GEP of a minor cell subpopulation is often lost when that cell subpopulation is of a frequency less than 30 percent. The GEP is almost always masked by the other cell subpopulations when that frequency drops to 10 percent or less. On the basis of these results one should always assume that the GEP of a given cell subpopulation is probably seriously affected by, the presence of significant numbers of other "contaminating" cell types. Several methodologies can be employed to enrich the target cells submitted for microarray analyses. These include magnetic sorting and laser capture microdissection. If a cell subpopulation of interest is small, very high-throughput cell separation technologies are needed to separate enough cells for conventional microarrays. However, high-throughput flow cytometry/cell sorting overcomes many restrictions of experimental enrichment conditions. This technology can also be used to sort smaller numbers of cells of specific cell subpopulations and subsequently amplify their mRNAs before microarray analyses. When purification techniques are applied to unfixed samples, the potential for changes in gene levels during the process of collection is an additional concern. Since RNA rapidly degrades, and specific mRNAs turn over in minutes or hours, the cell separation process must be very rapid. Hence, high-throughput cell separation (HTS) technologies are needed that can process the necessary number of cells expeditiously in order to avoid such uncontrolled changes in the target cells GEP. In cases where even the use of HTS yields only a small number of cells, the mRNAs (after reverse transcription to cDNA's) must be amplified to yield enough material for conventional microarray analyses. However, the problem of using "microamplification" PCR methods to expand the amount of cDNAs (from mRNAs) is that it is very difficult to amplify equally all of the mRNAs. Unequal amplification leads to a distorted gene expression profile on the microarray. Linear amplifications is difficult to achieve. Unfortunately, present-day gene-chips need to be about 100 times more sensitive than they are now to be able to do many biologically and biomedically meaningful experiments and clinical tests

    Sensitivity of wide band detectors to quintessential gravitons

    There are no reasons why the energy spectra of the relic gravitons, amplified by the pumping action of the background geometry, should not increase at high frequencies. A typical example of this behavior are quintessential inflationary models where the slopes of the energy spectra can be either blue or mildly violet. In comparing the predictions of scenarios leading to blue and violet graviton spectra we face the problem of correctly deriving the sensitivities of the interferometric detectors. Indeed, the expression of the signal-to-noise ratio not only depends upon the noise power spectra of the detectors but also upon the spectral form of the signal and, therefore, one can reasonably expect that models with different spectral behaviors will produce different signal-to-noise ratios. By assuming monotonic (blue) spectra of relic gravitons we will give general expressions for the signal-to-noise ratio in this class of models. As an example we studied the case of quintessential gravitons. The minimum achievable sensitivity to h02ΩGWh^2_{0} \Omega_{GW} of different pairs of detectors is computed, and compared with the theoretical expectations.Comment: 10 pages in Revtex style, 3 figure

    Virulence, drug sensitivity and transmission success in the rodent malaria, Plasmodium chabaudi.

    Here, we test the hypothesis that virulent malaria parasites are less susceptible to drug treatment than less virulent parasites. If true, drug treatment might promote the evolution of more virulent parasites (defined here as those doing more harm to hosts). Drug-resistance mechanisms that protect parasites through interactions with drug molecules at the sub-cellular level are well known. However, parasite phenotypes associated with virulence might also help parasites survive in the presence of drugs. For example, rapidly replicating parasites might be better able to recover in the host if drug treatment fails to eliminate parasites. We quantified the effects of drug treatment on the in-host survival and between-host transmission of rodent malaria (Plasmodium chabaudi) parasites which differed in virulence and had never been previously exposed to drugs. In all our treatment regimens and in single- and mixed-genotype infections, virulent parasites were less sensitive to pyrimethamine and artemisinin, the two antimalarial drugs we tested. Virulent parasites also achieved disproportionately greater transmission when exposed to pyrimethamine. Overall, our data suggest that drug treatment can select for more virulent parasites. Drugs targeting transmission stages (such as artemisinin) may minimize the evolutionary advantage of virulence in drug-treated infections

    Searching for the light dark gauge boson in GeV-scale experiments

    We study current constraints and search prospects for a GeV scale vector boson at a range of low energy experiments. It couples to the Standard Model charged particles with a strength <= 10^-3 to 10^-4 of that of the photon. The possibility of such a particle mediating dark matter self-interactions has received much attention recently. We consider searches at low energy high luminosity colliders, meson decays, and fixed target experiments. Based on available data, searches both at colliders and in meson decays can discover or exclude such a scenario if the coupling strength is on the larger side. We emphasize that a dedicated fixed target experiment has a much better potential in searching for such a gauge boson, and outline the desired properties of such an experiment. Two different optimal designs should be implemented to cover the range of coupling strength 10^-3 to 10^-5, and < 10^-5 of the photon, respectively.
