    Modulatory and Toxicological Perspectives on the Effects of the Small Molecule Kinetin

    Plant hormones are small regulatory molecules that exert pharmacological actions in mammalian cells such as anti-oxidative and pro-metabolic effects. Kinetin belongs to the group of plant hormones cytokinin and has been associated with modulatory functions in mammalian cells. The mammalian adenosine receptor (A2a-R) is known to modulate multiple physiological responses in animal cells. Here, we describe that kinetin binds to the adenosine receptor (A2a-R) through the Asn253 residue in an adenosine dependent manner. To harness the beneficial effects of kinetin for future human use, we assess its acute toxicity by analyzing different biochemical and histological markers in rats. Kinetin at a dose below 1 mg/kg had no adverse effects on the serum level of glucose or on the activity of serum alanine transaminase (ALT) or aspartate aminotransferase (AST) enzymes in the kinetin treated rats. Whereas, creatinine levels increased after a kinetin treatment at a dose of 0.5 mg/kg. Furthermore, 5 mg/kg treated kinetin rats showed normal renal corpuscles, but a mild degeneration was observed in the renal glomeruli and renal tubules, as well as few degenerated hepatocytes were also observed in the liver. Kinetin doses below 5 mg/kg did not show any localized toxicity in the liver and kidney tissues. In addition to unraveling the binding interaction between kinetin and A2a-R, our findings suggest safe dose limits for the future use of kinetin as a therapeutic and modulatory agent against various pathophysiological conditions

    ISOLATION, OPTIMIZATION, AND ANTITUMOR ACTIVITY OF L-ASPARAGINASE EXTRACTED FROM PECTOBACTERIUM CAROTOVORUM AND SERRATIA MARCESCENS ON HUMAN BREAST ADENOCARCINOMA AND HUMAN HEPATOCELLULAR CARCINOMA CANCER CELL LINES

    Objectives: The objective of this research was to obtain isolates capable of producing a high yield of L-asparaginase enzyme and to evaluate the antitumor activity of the purified enzyme against different cancer and normal cell lines. Methods: Isolation of bacteria was performed by the serial dilution technique of soil samples collected from Cairo, Egypt, using modified M9 agar plates. Culture filtrates of selected isolates were quantitatively screened for L-asparaginase production using well-diffusion and direct nesslerization techniques. Factors influencing L-asparaginase activity were optimized by studying the effect of physical and nutritional conditions on the enzyme activity. The purification of L-asparaginase extracted from both the isolates was achieved using chilled acetone (−20°C), followed by gel filtration on Sephadex G-100. The anticancer activity of the purified enzyme against human breast adenocarcinoma (MCF-7), human hepatocellular carcinoma (HepGII) and homo sapiens human (WISH) cell line was examined by 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyl tetrazolium bromide assay. Results: Two L-asparaginase producers were identified by Biolog identification system as Pectobacterium carotovorum and Serratia marcescens. Optimization increased the production of L-asparaginase to 4.835 and 5.221 U/ml for P. carotovorum and S. marcescens, respectively. L-asparaginase was extracted, purified, and tested in vitro for cytotoxic activity using 3-(4,5-Dimethylthiazol-2-yl)-2,5 diphenyl tetrazolium bromide (MTT assay) against MCF-7, HepGII, and WISH cell line. L-asparaginase from P. carotovorum and S. marcescens was neutral to normal epithelial WISH cells. On the other hand, L-asparaginase from both isolates was cytotoxic to MCF-7 and HepGII cancer cell lines with an half maximal inhibitory concentration of 15 μg/ml and 26 μg/ml and 26 μg/ml and 25 μg/ml, respectively. Conclusion: L-asparaginase extracted from P. carotovorum and S. marcescens showed remarkable anticancer activity. Further studies on hypersensitivity action need to be carried out to recommend the use of L-asparaginase as an alternative to commercially available preparations

    Evaluation of IR Spectral Analysis and Dyeing Parameters for Plasma and /or Nano-Silver Treatments of Polyester and Nylon Fabrics

    In our work of this paper, we study the effect of surface modification of polyester and nylon fabrics induced by DC plasma discharge and/ or nano-siliver treatments .DC plasma discharge was employed at first, as a function of plasma device parameters including different time, different current and different hydrostatic pressure using chemically inert working gas: argon or nitrogen. Optimization of the performance of the applied DC plasma discharge with various applied conditions were performed using Fourier Transform Infra-Red (FTIR) Spectroscopy spectral analysis, by following up the changes in the peak intensity values of the characteristic functional groups that characterize polyester fabric. Then the dyeing properties of different pretreated fabrics with plasma by the best conditions are subjected to nano-silver treatment by concentration 50 ppm under the effect of different dye concentrations, different dyeing temperature and different dyeing time. Finally, the fastness properties to light and washing for the treated samples were studied. The results obtained showed that both of the dyeing parameters and fastness properties were highly improved by the treatment of fabrics by either individual plasma treatment or combined DC cold plasma and nano-silver treatments

    Evaluation of the Detectability of Electromechanical Faults in Induction Motors Via Transient Analysis of the Stray Flux

    © 1972-2012 IEEE. The stray flux that is present in the vicinity of an induction motor is a very interesting information source to detect several types of failures in these machines. The analysis of this quantity can be employed, in some cases, as a supportive tool to complement the diagnosis provided by other quantities. In other cases, when no other motor quantities are available, stray flux analysis can become one of the few alternatives to evaluate the motor condition. Its noninvasive nature, low cost, and easy implementation makes it a very interesting option that requires further investigation. The aim of this work is to evaluate the suitability of the stray flux analysis under the starting transient as a way to detect certain faults in induction motors (broken rotor bars and misalignments), even when these types of faults coexist in the motor. To this end, advanced signal processing tools will be applied. Several positions of the flux sensors are considered in this study. Also, for the first time, a fault indicator based on the stray flux analysis under the starting is introduced and its sensitivity is compared versus other indicators relying on other quantities.     SULPHATED POLYSACCHARIDES (SPS) FROM THE GREEN ALGA ULVA FASCIATA EXTRACT MODULATES LIVER AND KIDNEY FUNCTION IN HIGH FAT DIET-INDUCED HYPERCHOLESTEROLEMIC RATS

    Objective: Hypercholesterolemia (HC) was frequently associated with oxidative stress, and release of inflammatory cytokines is to determine the hypolipidemic effects of sulphated polysaccharides from seaweed Ulva fasciata algal extracts through measuring the activities of some parameters related to liver and kidney functions in the serum of hypercholesterolemic rats as compared to normal one.Methods: Different groups of rats were administered a high cholesterol diet. Liver and kidney functions, inflammatory cytokines (TNF-α, CRP, MPO and IL-10), oxidative stress (GSH, MDA and NO), in addition to cell adhesion molecules (ICAM-1 and VCAM-1) were assessed before and after treatment with the algal polysaccharides. In addition, histological examination of liver and kidney were performed to confirm the biochemical findings.Results: The obtained results showed that oxidative stress and inflammatory markers associated with hypercholesterolemia were significantly increased in HC-rats. The histopathological examination of liver and kidney demonstrated severe degeneration with diffuse vacuolar degeneration, necrosis and the presence of fatty droplets. In addition; nephron-histological examination revealed, mild glomerular injury with mild vascular and inflammatory changes. Treatment with the algal sulphated polysaccharides effectively improved these disorders and diminished the formation of fatty liver, as well as renal dysfunction more than the reference drug; fluvastatin. Conclusion: It could be concluded that the consumption of UFP (Ulva fasciata polysaccharides), may be associated with attenuation of inflammatory markers, amelioration of fatty liver and improvement of renal dysfunction, that in turn lead to counteract hypercholesterolemia and its related disorders; such as obesity, and heart disease.Keywords: Non-alcoholic fatty liver disease, Seaweed, Ulva fasciata, Hypercholesterolemia, Hypolipidemic activity, Sulphated polysaccharides (SPs

    Tetracycline resistance phenotypes and genotypes of coagulase-negative staphylococcal isolates from bubaline mastitis in Egypt

    Aim: This study was devoted to elucidate the tetracycline resistance of coagulase-negative staphylococci (CNS) derived from normal and subclinical mastitic (SCM) buffaloes' milk in Egypt. Materials and Methods: A total of 81 milk samples from 46 normal buffalo milk samples and 35 SCM buffalo milk samples at private dairy farms of Egypt were used in this study. CNS were identified using phenotypic and molecular methods (polymerase chain reaction [PCR]). CNS isolates were tested for tetracycline resistance using routine methods and multiplex PCR targeting tetracycline (tet) resistance genes followed by sequencing of positive PCR products and phylogenetic analysis. Results: Isolation and identification of 28 (34.5%) CNS from normal and SCM buffaloes' milk, namely, Staphylococcus intermedius (39.2%), Staphylococcus xylosus (25.0%), Staphylococcus epidermidis (10.7%), Staphylococcus hominis (10.7%), and 3.5% to each of Staphylococcus sciuri, Staphylococcus hyicus, Staphylococcus lugdunensis, and Staphylococcus simulans. Using nested PCR, all the 28 CNS isolates revealed positive for 16srRNA gene specific for genus staphylococci and negative for thermonuclease (nuc) gene specific for Staphylococcus aureus species. The presence of tetracycline resistance-encoding genes (tetK, tetL, tetM, and tetO) was detected by multiplex PCR. All isolates were negative for tetL, M, and O genes while 14 (50%) CNS isolates were positive for tetK gene, namely, S. lugdunensis (100%), S. hominis (100%), S. epidermidis (66.6%), S. intermedius (45.4%), and S. xylosus (42.8%). Nucleotide sequencing of tetK gene followed by phylogenetic analysis showed the high homology between our CNS isolates genes of tetracycline resistance with S. aureus isolates including Egyptian ones. This proves the transfer of the tetracycline resistance encoding genes between coagulase-negative and coagulase-positive Staphylococcus spp. Conclusion: CNS isolates have distinguishingly high resistance to tetracycline. Abundant tetracycline usage for mastitis treatment leads to the spread of genetic resistance mechanisms inside CNS strains and among all Staphylococcus spp. Consequently, tetracycline is not effective anymore

    Chemical Profile of Cyperus laevigatus and Its Protective Effects against Thioacetamide-Induced Hepatorenal Toxicity in Rats

    Cyperus species represent a group of cosmopolitan plants used in folk medicine to treat several diseases. In the current study, the phytochemical profile of Cyperus laevigatus ethanolic extract (CLEE) was assessed using UPLC-QTOF–MS/MS. The protective effect of CLEE at 50 and 100 mg /kg body weight (b.w.) was evaluated on hepatorenal injuries induced by thioacetamide (100 mg/kg) via investigation of the extract’s effects on oxidative stress, inflammatory markers and histopathological changes in the liver and kidney. UPLC-QTOF–MS/MS analysis of CLEE resulted in the identification of 94 compounds, including organic and phenolic acids, flavones, aurones, and fatty acids. CLEE improved the antioxidant status in the liver and kidney, as manifested by enhancement of reduced glutathione (GSH) and coenzyme Q10 (CoQ10), in addition to the reduction in malondialdehyde (MDA), nitric oxide (NO), and 8-hydroxy-2′-deoxyguanosine (8OHdG). Moreover, CLEE positively affected oxidative stress parameters in plasma and thwarted the depletion of hepatorenal ATP content by thioacetamide (TAA). Furthermore, treatment of rats with CLEE alleviated the significant increase in plasma liver enzymes, kidney function parameters, and inflammatory markers. The protective effect of CLEE was confirmed by a histopathological study of the liver and kidney. Our results proposed that CLEE may reduce TAA-hepatorenal toxicity via its antioxidant and anti-inflammatory properties suppressing oxidative stress
