    A Rapid and Selective LC-MS/MS Method for Quantification of Quetiapine in Human Plasma and its Application to Pharmacokinetic Study on Indian Schizophrenia Patients

    A rapid, robust and selective high pressure liquid chromatography–positive electrospray ionization tandem mass spectrometry method has been developed and validated for the quantification of quetiapine (QUE) in human plasma with K2EDTA using oxcarbazepine (IS) as an internal standard. Analyte and internal standard were extracted from human plasma by solid-phase extraction using acetonitrile. The eluted samples were chromatographed on a C18 column by using a 10:75:15v/v mixture of ammonium formate buffer (5 mM, pH 4.50) and acetonitrile and methanol as an isocratic mobile phase at a flow rate of 0.4 mL/min and analyzed by mass spectrometry in the multiple reaction monitoring mode using the respective [M+H]+ ions, m/z 384.3/253.2 for Quetiapine and m/z 253.1/208.1 for the internal standard. The assay exhibited a linear dynamic range of 5.01 - 2501.04 ng/mL for quetiapine in human plasma. Acceptable precision and accuracy were obtained for concentrations over the standard curve range. A run time of 2.5 min for each sample made it possible to analyze 300 patient plasma samples per day. The validated method has been successfully used for the estimation of quetiapine in real time schizophrenia patient’s plasma samples for pharmacokinetic study

    Gene effects for grain iron and zinc in sorghum (sorghum bicolor L. Moench)

    Sorghum bicolor L. Moench is a major food crop in African and Asian countries. Eradication of hidden hunger through fortification of food consumed is an easy way. Iron and Zinc deficiencies are widespread across the world.Thus the present investigation was carried out in sorghum to determine the gene action involved in the inheritance of grain iron and zinc concentrations and other related traits using generation mean analysis. Two crosses viz., ICSB 52 × IS 13211, ICSB 52 × SPV 1359, were made using diverse parents with varied levels of grain iron and zinc concentrations. Six generations viz., P1, P2, F1, F2, B1 and B2 were developed for the above crosses and were evaluated during post-rainy season, 2012- 13 at ICRISAT, Patancheru. The results of generation mean analysis revealed the predominant role of additive gene action and additive × additive component of epistasis was found responsible in the crosses ICSB 52 × IS 13211 and ICSB 52 × SPV 1359 in governing grain iron and zinc concentrations and days to 50 % flowering. For remaining traits viz., plant height, panicle length, panicle width, grain yield plant -1, 100- grain weight had dominance component and found responsible than additive. Among the interactions, dominance × dominance component was of higher value than the remaining in governing the traits under concern. For all the traits, the magnitude of interactions was of higher value than their respective direct effects

    Inheritance studies on grain iron and zinc concentration and agronomic traits in sorghum [Sorghum bicolor (L.) Moench]

    Sorghum is a major staple globally and biofortifying sorghum with increased grain iron and zinc complements ongoing efforts to combat micronutrient malnutrition. Limited information available on the nature and magnitude of gene effects for grain iron and zinc. So generation mean analysis was done using four crosses, ICSB 52 × IS 13211, ICSB 52 × SPV 1359, IS 20843 × IS 2248 and IS 2248 × IS 20843. Six generations, P1, P2, F1, F2, B1 and B2, were generated for each of these crosses which were evaluated during the postrainy season 2012–13. The mean sum of squares due to generations were significant for days to flowering, plant height, grain yield/plant, 100-grain weight, and grain Fe and Zn. Generation mean studies revealed the presence of both additive and dominant gene interactions in inheritance of agronomic traits. Duplicative epistasis was observed for days to flowering, plant height, grain yield/plant, 100-grain weight, and grain Zn with a predominance of dominant gene action in inheritance of these traits. Panicle length, panicle width, and grain Fe showed both additive and dominance gene effects, with higher magnitude of dominance. This information can be utilized in developing sorghum lines with high grain Fe and Zn

    Contact heat evoked potentials using simultaneous EEG and fMRI and their correlation with evoked pain

    BACKGROUND: The Contact Heat Evoked Potential Stimulator (CHEPS) utilises rapidly delivered heat pulses with adjustable peak temperatures to stimulate the differential warm/heat thresholds of receptors expressed by Adelta and C fibres. The resulting evoked potentials can be recorded and measured, providing a useful clinical tool for the study of thermal and nociceptive pathways. Concurrent recording of contact heat evoked potentials using electroencephalogram (EEG) and functional magnetic resonance imaging (fMRI) has not previously been reported with CHEPS. Developing simultaneous EEG and fMRI with CHEPS is highly desirable, as it provides an opportunity to exploit the high temporal resolution of EEG and the high spatial resolution of fMRI to study the reaction of the human brain to thermal and nociceptive stimuli. METHODS: In this study we have recorded evoked potentials stimulated by 51° C contact heat pulses from CHEPS using EEG, under normal conditions (baseline), and during continuous and simultaneous acquisition of fMRI images in ten healthy volunteers, during two sessions. The pain evoked by CHEPS was recorded on a Visual Analogue Scale (VAS). RESULTS: Analysis of EEG data revealed that the latencies and amplitudes of evoked potentials recorded during continuous fMRI did not differ significantly from baseline recordings. fMRI results were consistent with previous thermal pain studies, and showed Blood Oxygen Level Dependent (BOLD) changes in the insula, post-central gyrus, supplementary motor area (SMA), middle cingulate cortex and pre-central gyrus. There was a significant positive correlation between the evoked potential amplitude (EEG) and the psychophysical perception of pain on the VAS. CONCLUSION: The results of this study demonstrate the feasibility of recording contact heat evoked potentials with EEG during continuous and simultaneous fMRI. The combined use of the two methods can lead to identification of distinct patterns of brain activity indicative of pain and pro-nociceptive sensitisation in healthy subjects and chronic pain patients. Further studies are required for the technique to progress as a useful tool in clinical trials of novel analgesics

    The Changing Landscape for Stroke\ua0Prevention in AF: Findings From the GLORIA-AF Registry Phase 2

    Background GLORIA-AF (Global Registry on Long-Term Oral Antithrombotic Treatment in Patients with Atrial Fibrillation) is a prospective, global registry program describing antithrombotic treatment patterns in patients with newly diagnosed nonvalvular atrial fibrillation at risk of stroke. Phase 2 began when dabigatran, the first non\u2013vitamin K antagonist oral anticoagulant (NOAC), became available. Objectives This study sought to describe phase 2 baseline data and compare these with the pre-NOAC era collected during phase 1. Methods During phase 2, 15,641 consenting patients were enrolled (November 2011 to December 2014); 15,092 were eligible. This pre-specified cross-sectional analysis describes eligible patients\u2019 baseline characteristics. Atrial fibrillation disease characteristics, medical outcomes, and concomitant diseases and medications were collected. Data were analyzed using descriptive statistics. Results Of the total patients, 45.5% were female; median age was 71 (interquartile range: 64, 78) years. Patients were from Europe (47.1%), North America (22.5%), Asia (20.3%), Latin America (6.0%), and the Middle East/Africa (4.0%). Most had high stroke risk (CHA2DS2-VASc [Congestive heart failure, Hypertension, Age  6575 years, Diabetes mellitus, previous Stroke, Vascular disease, Age 65 to 74 years, Sex category] score  652; 86.1%); 13.9% had moderate risk (CHA2DS2-VASc = 1). Overall, 79.9% received oral anticoagulants, of whom 47.6% received NOAC and 32.3% vitamin K antagonists (VKA); 12.1% received antiplatelet agents; 7.8% received no antithrombotic treatment. For comparison, the proportion of phase 1 patients (of N = 1,063 all eligible) prescribed VKA was 32.8%, acetylsalicylic acid 41.7%, and no therapy 20.2%. In Europe in phase 2, treatment with NOAC was more common than VKA (52.3% and 37.8%, respectively); 6.0% of patients received antiplatelet treatment; and 3.8% received no antithrombotic treatment. In North America, 52.1%, 26.2%, and 14.0% of patients received NOAC, VKA, and antiplatelet drugs, respectively; 7.5% received no antithrombotic treatment. NOAC use was less common in Asia (27.7%), where 27.5% of patients received VKA, 25.0% antiplatelet drugs, and 19.8% no antithrombotic treatment. Conclusions The baseline data from GLORIA-AF phase 2 demonstrate that in newly diagnosed nonvalvular atrial fibrillation patients, NOAC have been highly adopted into practice, becoming more frequently prescribed than VKA in Europe and North America. Worldwide, however, a large proportion of patients remain undertreated, particularly in Asia and North America.     Contribution of PEPFAR-Supported HIV and TB Molecular Diagnostic Networks to COVID-19 Testing Preparedness in 16 Countries.

    The US President's Emergency Plan for AIDS Relief (PEPFAR) supports molecular HIV and tuberculosis diagnostic networks and information management systems in low- and middle-income countries. We describe how national programs leveraged these PEPFAR-supported laboratory resources for SARS-CoV-2 testing during the COVID-19 pandemic. We sent a spreadsheet template consisting of 46 indicators for assessing the use of PEPFAR-supported diagnostic networks for COVID-19 pandemic response activities during April 1, 2020, to March 31, 2021, to 27 PEPFAR-supported countries or regions. A total of 109 PEPFAR-supported centralized HIV viral load and early infant diagnosis laboratories and 138 decentralized HIV and TB sites reported performing SARS-CoV-2 testing in 16 countries. Together, these sites contributed to >3.4 million SARS-CoV-2 tests during the 1-year period. Our findings illustrate that PEPFAR-supported diagnostic networks provided a wide range of resources to respond to emergency COVID-19 diagnostic testing in 16 low- and middle-income countries

    Retrospective evaluation of whole exome and genome mutation calls in 746 cancer samples

    Funder: NCI U24CA211006Abstract: The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC) curated consensus somatic mutation calls using whole exome sequencing (WES) and whole genome sequencing (WGS), respectively. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2,658 cancers across 38 tumour types, we compare WES and WGS side-by-side from 746 TCGA samples, finding that ~80% of mutations overlap in covered exonic regions. We estimate that low variant allele fraction (VAF < 15%) and clonal heterogeneity contribute up to 68% of private WGS mutations and 71% of private WES mutations. We observe that ~30% of private WGS mutations trace to mutations identified by a single variant caller in WES consensus efforts. WGS captures both ~50% more variation in exonic regions and un-observed mutations in loci with variable GC-content. Together, our analysis highlights technological divergences between two reproducible somatic variant detection efforts

    Rice is sensitive to salinity at seedling and reproductive stages. A wide genetic variation was reported in rice for salinity tolerance, a pre-requisite for any plant breeding programme. In the present study, we have characterized a set of 192 rice genotypes for their seedling stage salinity tolerance by combining morpho-physiological and molecular markers. The experiment was conducted under control (EC ∼ 1.2 dS m−1) and saline stress conditions (EC ∼ 12 dS m−1) using hydroponics. There was a significant difference among the genotypes for the ten characters studied. Under saline conditions, the chlorophyll concentration, root length, shoot length, and K+ concentration decreased as compared to control condition and the opposite was true for Na+ concentration. Na+ concentration showed strong negative correlation with chlorophyll concentration, K+ concentration, root length and shoot length. Thirteen SSR markers associated with Saltol region on chromosome 1 were screened across the 192 rice genotypes, of which ten gave scoreable results. Polymorphism Information Content (PIC) and genetic diversity indices showed that the markers RM 493 and RM 10793 were highly useful for distinguishing genotypes. Hierarchical cluster analysis revealed eleven clusters for phenotypic and genotypic data, but very low correspondence was observed between two dendograms. The haplotypes of genotypes for Saltol associated markers, when compared with FL 478, revealed that the genotypes CST 7-1 and Arvattelu could be good candidates for novel genomic regions governing salinity tolerance in rice
