Considering documents in lifelog information retrieval
Lifelogging is a research topic that is receiving increasing attention and although lifelog research has progressed in recent years, the concept of what represents a document in lifelog retrieval has not yet been sufficiently explored. Hence, the generation of multimodal lifelog documents is a fundamental concept that must be addressed. In this paper, I introduce my general perspective on generating documents in lifelogging and reflect on learnings from collecting multimodal lifelog data from a number of participants in a study on lifelog data organization. In addition, the main motivation be- hind document generation is proposed and the challenges faced while collecting data and generating documents are discussed in detail. Finally, a process for organizing the documents in lifelog data retrieval is proposed, which I intend to follow in my PhD research
Love Experiences of Older African Americans: A Qualitative Study
The purpose of this study was to explore the romantic love experiences of African American older adults. Popular views on love among older African Americans remain highly subjective, open to the imaginations of other individuals and groups. This study attempts to challenge the popular descriptions of the nature of love among African American elderly by gathering narratives on various aspects of love as told by a sample of African-American elderly. A qualitative design is used to sample, gather and analyze data on love among African-American elderly. Our findings reveal that African American older adults value emotional intimacy. Cultural and historical factors appear to influence the choice of partners in later life. Implications for social intervention are discussed
Ekspresija i pročišćavanje glavnoga proteina (OmpH) bakterije Pasteurella multocida P52 proizvedenoga u bakteriji Escherichia coli.
Porin H (OmpH) is the major outer membrane protein in the envelope of Pasteurella multocida. The gene ompH, encoding major outer membrane protein was amplified by PCR excluding the region coding for signal peptide and cloned in the pQE32 prokaryotic expression vector. The recombinant OmpH was expressed as a fusion protein with 6-His tag at N-terminal in E. coli M15 cells transformed with recombinant plasmid pQE32-ompH. The expressed protein was purified from E. coli and characterized by SDS-PAGE and western blot analysis. The fusion recombinant protein eluted had a molecular mass of about 37 kDa. The expressed recombinant protein was confirmed with western blot analysis using RGS-His antibody and anti-P. multocida serum raised against whole cell lysate.Porin H (OmpH) je glavni protein stanične stijenke bakterije Pasteurella multocida. Rekombinantni OmpH bio je izražen kao fuzijski protein sa 6-His tag na N-kraju u stanicama E. coli M15 transformiranima rekombinantnim plazmidom pQE32- ompH. Proizveden protein bio je pročišćen iz E. coli i identificiran SDS-PAGE-om i western blotom. Izdvojeni fuzijski rekombinantni protein imao je molekularnu masu oko 37 kDa. Transcriptional landscape of epithelial and immune cell populations revealed through FACS-seq of healthy human skin.
Human skin consists of multiple cell types, including epithelial, immune, and stromal cells. Transcriptomic analyses have previously been performed from bulk skin samples or from epithelial and immune cells expanded in cell culture. However, transcriptomic analysis of bulk skin tends to drown out expression signals from relatively rare cells while cell culture methods may significantly alter cellular phenotypes and gene expression profiles. To identify distinct transcriptomic profiles of multiple cell populations without substantially altering cell phenotypes, we employed a fluorescence activated cell sorting method to isolate keratinocytes, dendritic cells, CD4+ T effector cells, and CD8+ T effector cells from healthy skin samples, followed by RNA-seq of each cell population. Principal components analysis revealed distinct clustering of cell types across samples, while differential expression and coexpression network analyses revealed transcriptional profiles of individual cell populations distinct from bulk skin, most strikingly in the least abundant CD8+ T effector population. Our work provides a high resolution view of cutaneous cellular gene expression and suggests that transcriptomic profiling of bulk skin may inadequately capture the contribution of less abundant cell types
