    Mapping the dielectric properties of unknown targets by using a network of microwave sensors: A proof-of-concept

    The subject of this paper is the possible use of a network of microwave sensors to achieve a map of the electromagnetic properties of unknown targets. The basic idea is to use a set of microwave sensors to illuminate a region of interest and to measure the resulting axial component of the electric field. Measurements are then processed by means of a technique based on inverse-scattering, which provides an estimate map of the dielectric values of the area under examination, allowing to discriminate among possible targets. In order to initially evaluate the feasibility of the proposed approach, numerical results in a simulated environment are preliminarily considered and discussed. Furthermore, an initial test on experimental data in a simplified configuration is also presented

    FoodOmics as a new frontier to reveal microbial community and metabolic processes occurring on table olives fermentation

    Table olives are considered the most widespread fermented food in the Mediterranean area and their consumption is expanding all over the world. This fermented vegetable can be considered as a natural functional food thanks to their high nutritional value and high content of bioactive compounds that contribute to the health and well-being of consumers. The presence of bioactive compounds is strongly influenced by a complex microbial consortium, traditionally exploited through culture-dependent approaches. Recently, the rapid spread of omics technologies has represented an important challenge to better understand the function, the adaptation and the exploitation of microbial diversity in different complex ecosystems, such as table olives. This review provides an overview of the potentiality of omics technologies to in depth investigate the microbial composition and the metabolic processes that drive the table olives fermentation, affecting both sensorial profile and safety properties of the final product. Finally, the review points out the role of omics approaches to raise at higher sophisticated level the investigations on microbial, gene, protein, and metabolite, with huge potential for the integration of table olives composition with functional assessments

    Metagenetic and Volatilomic Approaches to Elucidate the Effect of Lactiplantibacillus plantarum Starter Cultures on Sicilian Table Olives

    The present study aimed to evaluate the effect of selected Lactiplantibacillus plantarum strains on both microbiota composition and volatile organic compound profile of Sicilian table olives. Two mixed cultures, named O1 and O2, were set up for pilot-plan scale fermentations at 5% of NaCl. Uninoculated table olives at 5 and 8% (C5 and C8) of salt were used as control. The fermentation process was monitored until 80 days through a dual approach, which included both classical microbiological and 16S amplicon-based sequencing and volatilomics analyses. Compared with control samples (C5 and C8), experimental samples, inoculated with starter cultures (O1 and O2), exhibited a faster acidification with a more pronounced drop in pH. Metagenetics data revealed significant differences of microbiota composition among samples, highlighting the dominance of lactobacilli in both experimental samples; a high occurrence of Enterobacter genus only in control samples with 5% of NaCl; and the presence of Bacteroides, Faecalibacterium, Klebsiella, and Raoultella genera only in control samples with 8% of NaCl. Furthermore, microbiota composition dynamics, through the fermentation process, significantly affected the volatile organic compounds of the final products, whereas no compounds involved in off-odors metabolites were detected in all samples investigated. In conclusion, the addition of the proposed starter cultures and the use of low concentrations of sodium chloride positively affected the microbiota and volatile organic compounds, ensuring the microbiological safety and the pleasant flavors of the final product

    Initial Experimental Tests of an ANN-Based Microwave Imaging Technique for Neck Diagnostics

    In this letter, a microwave imaging strategy based on an artificial neural network (ANN) is applied, for the first time, to experimental data gathered from simplified neck phantoms. The ANN is used for solving the underlying inverse scattering problem, with the aim of retrieving the dielectric properties of the neck for monitoring and diagnostic purposes. The ANN is trained using simulated phantoms, to overcome the limited availability of experimental data. First, a simple configuration with a liquid-filled glass beaker is tested. Then, simplified 3-D-printed models of the human neck are considered. The preliminary findings indicate the possibility of training the network with numerical simulations and testing it against experimental measurements

    A clinical pilot study on the effect of the probiotic Lacticaseibacillus rhamnosus TOM 22.8 strain in women with vaginal dysbiosis

    Lactobacilli with probiotic features play an essential role in maintaining a balanced vaginal microbiota and their administration has been suggested for the treatment and prevention of vaginal dysbiosis. The present study was aimed to in vitro and in vivo investigate the probiotic potential of the Lacticaseibacillus rhamnosus TOM 22.8 strain, isolated from the vaginal ecosystem of a healthy woman. For this purpose, safety and functional properties were in depth evaluated. The strain exhibited a broad spectrum of antagonistic activity against vaginal pathogens; adhesion capacity to both the vaginal VK2/E6E7 and the intestinal Caco-2 cells; anti-inflammatory and antioxidant activities, suggesting its promising probiotic features. In addition, an in vivo pilot-study was planned. Based on both clinical and microbiological parameters, the oral or vaginal strain administration, determined a significant pathogens reduction after 10 days of administration and a maintenance of eubiosis up to 30 days after the end of the treatment. Therefore, the L. rhamnosus TOM 22.8 strain can be proposed as valuable oral and/or vaginal treatment for vaginal dysbiosis

    SummarySome HGPRT spontaneous revertants were isolated from a mutant line (E2) of V79 Chinese hamster cells and phenotypically characterized. Dot–Blot hybridization with a32P-Iabelled HGPRT probe revealed an increase in the number of HGPRT sequences in some of these revertants, suggesting the occurrence of gene amplification. Cytogenetic analysis performed in three of these revertants showed a characteristic abnormally banding region (ABR) on the elongated p arm of theXchromosome.In Situhybridization in one revertant (RHE2) showed that the amplified sequences reside on the p+arm of theXchromsome in two different localizations. Because of the very probable clonal origin of the revertant, these features indicate that the amplified sequences might rearrange after their integration into the chromosome

    Demonstrating a smart controller in a hospital integrated energy system

    Integrated energy systems have recently gained primary importance in clean energy transition. The combination of the electricity, heating and gas sectors can improve the overall system efficiency and integration of renewables by exploiting the synergies among the energy vectors. In particular, real-time optimization tools based on Model Predictive Control (MPC) can considerably improve the performance of systems with several conversion units and distribution networks by automatically coordinating all interacting technologies. Despite the relevance of several simulation studies on the topic, however, it is significantly harder to have an experimental demonstration of this improvement. This work presents a methodology for the real-world implementation of a novel smart control strategy for integrated energy systems, based on two coordinated MPC levels, which optimize the operation of all conversion units and all energy vectors in the short- and long-term, respectively, to account also for economic incentives on critical units. The strategy that was previously developed and evaluated in a simulation environment has now been implemented, as a supervisory controller, in the integrated energy system of a hospital in Italy. The optimal control logic is easily actuated by dynamically communicating the optimal set-points to the existing Building Management System, without having to alter the system configuration. Field data collected over a two-year period, firstly when it was business as usual and when the new operation was introduced, show that the MPC increased the economic margin and revenues from yearly incentives and lowered the amount of electricity purchased, reducing dependency on the power grid

    AbstractHerein we report the quantification of purine lesions arising from gamma-radiation sourced hydroxyl radicals (HO•) on tertiary dsDNA helical forms of supercoiled (SC), open circular (OC), and linear (L) conformation, along with single-stranded folded and non-folded sequences of guanine-rich DNA in selected G-quadruplex structures. We identify that DNA helical topology and folding plays major, and unexpected, roles in the formation of 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxo-dG) and 8-oxo-7,8-dihydro-2'-deoxyadenosine (8-oxo-dA), along with tandem-type purine lesions 5′,8-cyclo-2′-deoxyguanosine (5′,8-cdG) and 5′,8-cyclo-2′-deoxyadenosine (5′,8-cdA). SC, OC, and L dsDNA conformers together with folded and non-folded G-quadruplexes d[TGGGGT]4 (TG4T), d[AGGG(TTAGGG)3] (Tel22), and the mutated tel24 d[TTGGG(TTAGGG)3A] (mutTel24) were exposed to HO• radicals and purine lesions were then quantified via stable isotope dilution LC-MS/MS analysis.
