    Selection of reference genes for gene expression studies in ultraviolet B-irradiated human skin fibroblasts using quantitative real-time PCR

    <p>Abstract</p> <p>Background</p> <p>Reference genes are frequently used to normalise mRNA levels between different samples. The expression level of these genes, however, may vary between tissues or cells and may change under certain circumstances. Cytoskeleton genes have served as multifunctional tools for experimental studies as reference genes. Our previous studies have demonstrated that the expression of vimentin, one cytoskeletal protein, was increased in ultraviolet B (UVB)-irradiated fibroblasts. Thus, we examined the expression of other cytoskeleton protein genes, <it>ACTB </it>(<it>actin, beta</it>), <it>TUBA1A </it>(<it>tubulin, alpha 1a</it>), and <it>TUBB1 </it>(<it>tubulin, beta 1</it>), in human dermal fibroblasts irradiated by UVB to determine which of these candidates were the most appropriate reference genes.</p> <p>Results</p> <p>Quantitative real-time PCR followed by analysis with the NormFinder and geNorm software programmes was performed. The initial screening of the expression patterns demonstrated that the expression of <it>VIM </it>was suppressed after UVB irradiation at doses ≥25 mJ/cm<sup>2 </sup>and that the expression of <it>TUBA1A </it>was significantly reduced by UVB doses ≥75 mJ/cm<sup>2 </sup>in cultured human dermal fibroblasts. The analysis of the experimental data revealed <it>ACTB </it>to be the most stably expressed gene, followed by <it>GAPDH </it>(<it>aglyceraldehyde-3-phosphate dehydrogenase</it>), under these experimental conditions. By contrast, <it>VIM </it>was found to be the least stable gene. The combination of <it>ACTB </it>and <it>TUBB1 </it>was revealed to be the gene pair that introduced the least systematic error into the data normalisation.</p> <p>Conclusion</p> <p>The data herein provide evidence that <it>ACTB </it>and <it>TUBB1 </it>are suitable reference genes in human skin fibroblasts irradiated by UVB, whereas <it>VIM </it>and <it>TUBA1A </it>are not and should therefore be excluded as reference genes in any gene expression studies involving UVB-irradiated human skin fibroblasts.</p

    X-ray spectral components observed in the afterglow of GRB 130925A

    We have identified spectral features in the late-time X-ray afterglow of the unusually long, slow-decaying GRB 130925A using NuSTAR, Swift/X-Ray Telescope, and Chandra. A spectral component in addition to an absorbed power law is required at >4σ significance, and its spectral shape varies between two observation epochs at 2 × 105 and 106 s after the burst. Several models can fit this additional component, each with very different physical implications. A broad, resolved Gaussian absorption feature of several keV width improves the fit, but it is poorly constrained in the second epoch. An additive blackbody or second power-law component provide better fits. Both are challenging to interpret: the blackbody radius is near the scale of a compact remnant (108 cm), while the second power-law component requires an unobserved high-energy cutoff in order to be consistent with the non-detection by Fermi/Large Area Telescope. © 2014.     Vegan diets : practical advice for athletes and exercisers.

    With the growth of social media as a platform to share information, veganism is becoming more visible, and could be becoming more accepted in sports and in the health and fitness industry. However, to date, there appears to be a lack of literature that discusses how to manage vegan diets for athletic purposes. This article attempted to review literature in order to provide recommendations for how to construct a vegan diet for athletes and exercisers. While little data could be found in the sports nutrition literature specifically, it was revealed elsewhere that veganism creates challenges that need to be accounted for when designing a nutritious diet. This included the sufficiency of energy and protein; the adequacy of vitamin B12, iron, zinc, calcium, iodine and vitamin D; and the lack of the long-chain n-3 fatty acids EPA and DHA in most plant-based sources. However, via the strategic management of food and appropriate supplementation, it is the contention of this article that a nutritive vegan diet can be designed to achieve the dietary needs of most athletes satisfactorily. Further, it was suggested here that creatine and β-alanine supplementation might be of particular use to vegan athletes, owing to vegetarian diets promoting lower muscle creatine and lower muscle carnosine levels in consumers. Empirical research is needed to examine the effects of vegan diets in athletic populations however, especially if this movement grows in popularity, to ensure that the health and performance of athletic vegans is optimised in accordance with developments in sports nutrition knowledge

    V1432 Aql: A polar or an intermediate polar?

    We present evidence for a spin period of 4070 s in a magnetic cataclysmic variable V1432 Aql, classified as a polar. The evidence is based on the analysis of archival X-ray data obtained from a 25 ks long and uninterrupted observations with XMM-Newton. We also detect modulation of the X-ray flux with the orbital period, various harmonics of the spin period, and side-bands due to beating of the spin period with the orbital period. The energy-dependent light curves and hardness ratios show a very complex behaviour with both the spin phase and the orbital phase, indicating the presence of multiple absorbers of different densities. The observed characteristics suggest the presence of a disk in the system and accretion via a disk-overflow mechanism as is commonly observed in intermediate polars

    We present the analysis of archival Chandra high resolution X-ray spectra of AM Her. Emission lines from several hydrogen-like ions, helium-like ions, Fe-L shell transitions and Fe-K fluorescent are identified. Using the resonance, intercombination and forbidden lines of the few prominent helium-like ions, we infer a density greater than 2 x 10(12) cm(-3) and a temperature of similar to 2 MK for the oxygen and neon line emitting regions in the accretion column of AM Her

    Periodicities In The X-Ray Intensity Variations of TV Columbae: An Intermediate Polar

    We present results from a temporal analysis of the longest and the most sensitive X-ray observations of TV Columbae--an intermediate polar. The observations were carried out with the RXTE PCA, ROSAT PSPC, and ASCA. Data were analyzed using a 1-dimensional CLEAN and Bayesian algorithms. The presence of a nearly sinusoidal modulation due to the spin of the white dwarf is seen clearly in all the data, confirming the previous reports based on the EXOSAT data. An improved period of 1909.7+/-2.5s is derived for the spin from the RXTE data.The binary period of 5.5hr is detected unambiguously in X-rays for the first time. Several side-bands due to the interaction of these periods are observed in the power spectra, thereby suggesting contributions from both the disk-fed and the stream-fed accretion for TV Col. The accretion disk could perhaps be precessing as side-bands due to the influence of 4 day period on the orbital period are seen. The presence of a significant power at certain side-bands of the spin frequency indicates that the emission poles are asymmetrically located. The strong power at the orbital side-bands seen in both the RXTE and ROSAT data gives an indication for an absorption site fixed in the orbital frame. Both the spin and the binary modulation are found to be energy-dependent. Increased hardness ratio during a broad dip in the intensity at binary phase of 0.75--1.0 confirms the presence of a strong attenuation due to additional absorbers probably from an impact site of the accretion stream with the disk or magnetosphere. Hardness ratio variations and the energy dependent modulation depth during the spin modulation can be explained by partially covered absorbers in the path of X-ray emission region in the accretion stream

    Australia provides abundant examples of continental-scale evolutionary radiations. The collision of two continental shelves around 30 Ma facilitated an influx of squamates and the subsequent squamate radiations resulted in high taxonomic diversity. The morphological disparity seen in these major squamate groups, however, remains underexplored. Here, we examine the major cranial proportions of over 1,000 specimens using 2D linear measurements to explicitly quantify the morphological disparity of Australian agamid lizards (Amphibolurinae) and compare it to that of agamid, acrodont, and iguanian clades from other parts of the world. Our results indicate the Australian Amphibolurinae have exceptionally high cranial disparity, and we suggest that this is linked to the relaxed selective environment that greeted the founders of Amphibolurinae when they first arrived in Australia.
