Fast electrophoresis in conventional capillaries by employing a rapid injection device and contactless conductivity detection
A purpose-made set-up featuring an automated fast injector allowed the easy optimization of the injected amount and the adjustment of the separation length of conventional capillaries from a minimum of 5cm upward. It was found that a compromise in capillary length for separation efficiency and analysis time also has to take into account the injected amount, which in turn affects the sensitivity and hence the detection limit. The versatility of the system was demonstrated by the analysis of the major cations and anions in natural water samples in less than 1min, the concurrent determination of a mixture of amino acids and carbohydrates in 160s, and of three active ingredients in a pharmaceutical preparation in 40s. Beiträge zur Miniaturisierung der Fliessinjektionsanalyse und der Kapillarelektrophorese
Die vorliegende Arbeit beschreibt die Entwicklung von drei Projekten. Das erste
Projekt betrachtete die Automatisierung des Injektionssystems eines
Fliessinjektionsanalysesystems im Mikromassstab (ÎĽFIA), welches ohne die teueren
lithographischen Prozesse hergestellt wurde. Um die Reproduzierbarkeit zu verbessern
wurden alle Teile der Injektion durch einen Computer gesteuert. Die spektroskopische
Bestimmung von Analyten wurde durch den Ersatz der gewöhnlichen Teile des UV-Vis
Detektors, durch eine Leuchtdiode, optische Fasern und Photodioden vereinfacht.
Im zweiten Projekt wurde ein selbstgebautes tragbares
Kapillarelektrophoresegerät mit einer einfachen Absorptionszelle ohne fokussierende
optische Teile getestet. Dazu wurden zusätzlich die erste UV-Leuchtdiode und die erste
batteriebetriebene UV-Lichtquelle verwendet, die neu auf den Markt gekommen sind.
Das dritte Projekt bestand aus einer billigen und flexibleren Alternative zu dem
elektrophoretischen Chip mit der Benutzung einer konventionellen kurzen Kapillare. Es
wurde ein völlig automatisches Injektionssystem unten Verwendung von einem
Kreuzteil, zwei Spritzenpumpen und einem Ventil entwickelt. Zur Charakterisierung
des neuen Injektors wurden verschiedene Grundexperimente mit einem kommerziellen
UV-Detektor durchgefĂĽhrt. Verschiedene CE Parameter wurden im Vergleich zu der
Bodenzahl gemessen. Ein „in house“ gebauter kontaktloser Leitfähigkeitsdetektor
wurde dazu mit dem neuen Injektionssystem fĂĽr eine schnelle Trennung getestet. Am
Ende wurden Alkali- und Erdalkalimetalle, anorganische Anionen, Kohlenhydrate,
Kohlenhydrate und Aminosäure sowie Arzneimittel mit dem kontaktlosen
Leitfähigkeitsdetektor gemessen
