    Presence of glucose, xylose, and glycerol fermenting bacteria in the deep biosphere of the former Homestake gold mine, South Dakota

    Eight fermentative bacterial strains were isolated from mixed enrichment cultures of a composite soil sample collected at 1.34 km depth from the former Homestake gold mine in Lead, SD, USA. Phylogenetic analysis of their 16S rRNA gene sequences revealed that these isolates were affiliated with the phylum Firmicutes belonging to genera Bacillus and Clostridium. Batch fermentation studies demonstrated that isolates had the ability to ferment glucose, xylose, or glycerol to industrially valuable products such as ethanol and 1,3-propanediol (PDO). Ethanol was detected as the major fermentation end product in glucose-fermenting cultures at pH 10 with yields of 0.205–0.304 g of ethanol/g of glucose. While a xylose-fermenting strain yielded 0.189 g of ethanol/g of xylose and 0.585 g of acetic acid/g of xylose at the end of fermentation. At pH 7, glycerol-fermenting isolates produced PDO (0.323–0.458 g of PDO/g of glycerol) and ethanol (0.284–0.350 g of ethanol/g of glycerol) as major end products while acetic acid and succinic acid were identified as minor by-products in fermentation broths. These results suggest that the deep biosphere of the former Homestake gold mine harbors bacterial strains which could be used in bio-based production of ethanol and PDO

    UNC-104/KIF1A is a Kinesin-3 motor that transports synaptic vesicles from the cell body towards the synapse by binding to PI(4,5)P(2) through its PH domain. The fate of the motor upon reaching the synapse is not known. We found that wild-type UNC-104 is degraded at synaptic regions through the ubiquitin pathway and is not retrogradely transported back to the cell body. As a possible means to regulate the motor, we tested the effect of cargo binding on UNC-104 levels. The unc-104(e1265) allele carries a point mutation (D1497N) in the PI(4,5)P(2) binding pocket of the PH domain, resulting in greatly reduced preferential binding to PI(4,5)P(2)in vitro and presence of very few motors on pre-synaptic vesicles in vivo. unc-104(e1265) animals have poor locomotion irrespective of in vivo PI(4,5)P(2) levels due to reduced anterograde transport. Moreover, they show highly reduced levels of UNC-104 in vivo. To confirm that loss of cargo binding specificity reduces motor levels, we isolated two intragenic suppressors with compensatory mutations within the PH domain. These show partial restoration of in vitro preferential PI(4,5)P(2) binding and presence of more motors on pre-synaptic vesicles in vivo. These animals show improved locomotion dependent on in vivo PI(4,5)P(2) levels, increased anterograde transport, and partial restoration of UNC-104 protein levels in vivo. For further proof, we mutated a conserved residue in one suppressor background. The PH domain in this triple mutant lacked in vitro PI(4,5)P(2) binding specificity, and the animals again showed locomotory defects and reduced motor levels. All allelic variants show increased UNC-104 levels upon blocking the ubiquitin pathway. These data show that inability to bind cargo can target motors for degradation. In view of the observed degradation of the motor in synaptic regions, this further suggests that UNC-104 may get degraded at synapses upon release of cargo

    Effect of nano based seed treatment insecticides on seed quality in Pigeonpea

    A laboratory experiment was conducted to know the effect seed treatment with nano insecticides on seed quality of pigeonpea (Cajanus cajan (L.) Millsp.) cv. TS3R. This study was conducted to evaluate the effect of macro and nano insecticides on seed germination and vigour of Pigeonpea. Different recommended seed treatment insecticides viz, malathion, fenvalerate, emamectine benzoate, thiodicarb, sweet flag and neem seed kernel powder insecticides were synthesized to nano form using high energy planetary ball mill. The Pigeonpea seed were treated with different nano insecticides i.e., 10-90 per cent reduction in actual dosage. Among the different treatments studied, seed treated with nano malathion 50 per cent lesser than normal dosage, fenvalerate 60 per cent lesser, thiodicarb 10 per cent lesser, emamectine benzoate 30 per cent lesser, sweetflag 70 per cent lesser, neem seed kernel powder 40 per cent lesser than actual recommended dosage gave significantly higher seed germination (98.0, 98.67, 98.67, 97.0, 99.0 and 98.67 percent) ,less number of abnormal seedlings (1.0, 0.33, 1.0, 1.0, 1.0 and 0.33 per cent) , shoot length (10.13, 9.00, 11.47, 9.50, 10.90 and 10.87 cm), root length (12.56, 12.93, 12.83, 12.60 11.50 and 13.00 cm), seedling dry weight (85.73, 87.40, 88.47, 87.70, 88.60 and 88.27 g) and seedling vigour index (2223, 2164, 2397, 2143, 2217 and 2354) as compared to untreated seeds and macro insecticides. Therefore, it is very clear that nano based insecticides has a significant (0.1 %) impact on the seed quality improvement

    In recent years, people are considering healthy diet habits and many of them are trying to track and maintain their daily diet and consumption. To assist them, there are many applications available online and those applications are capable of recording calories for the ingredients consumed, but users must check individual calories and calculate total calories manually. In this paper, we propose a new technique to calculate calories for a given recipe in multiple formats. The new technique uses tokenization, hashing techniques and fuzzy matching for entity extraction and finally does the unit conversion to calculate calories. We compared the results of the proposed technique with the outcomes of the existing applications. These results proved that the new technique has the capacity to produce similar results compared to that of the existing applications and able to calculate calories for recipes in the different formats available on the internet
