Forschungsdatenmanagement in Brandenburg (FDM-BB): Aufbau einer Landesinitiative
Bundeslandinitiativen mit dem Schwerpunkt Forschungsdatenmanagement ( FDM ) spielen neben der Nationalen Forschungsdateninfrastruktur ( NFDI ) eine wichtige Rolle, um Kompetenzen, Beratungsangebote, technologische Dienstleistungen und dergleichen âin die FlĂ€che zu bringenâ. Der Beitrag stellt am Beispiel FDM-BB vor, wie in wissenschaftsgeleiteter Weise Handlungsempfehlungen erarbeitet und die Entwicklung einer Landesstrategie zu Forschungsdatenmanagement unterstĂŒtzt werden kann. Mit einer solchen Strategie sollen alle brandenburgischen Hochschulen â unter BerĂŒcksichtigung des Diskussionsund Entwicklungsstandes der auĂeruniversitĂ€ren Forschungseinrichtungen â in zwei Bereichen befĂ€higt werden: (1) dem Aufbau von FDM -AktivitĂ€ten vor Ort und (2) der kooperativen (Weiter-) Entwicklung gemeinsamer FDM -Dienste. Damit wird die AnschlussfĂ€higkeit der brandenburgischen Hochschulen zu den nationalen Entwicklungen ( NFDI ) und der European Open Science Cloud ( EOSC ) hergestellt und das koordinierte Vorgehen der Einrichtungen unterstĂŒtzt
Genotype-phenotype correlation at codon 1740 ofSETD2
The SET domain containing 2, histone lysine methyltransferase encoded by SETD2 is a dual-function methyltransferase for histones and microtubules and plays an important role for transcriptional regulation, genomic stability, and cytoskeletal functions. Specifically, SETD2 is associated with trimethylation of histone H3 at lysine 36 (H3K36me3) and methylation of α-tubulin at lysine 40. Heterozygous loss of function and missense variants have previously been described with Luscan-Lumish syndrome (LLS), which is characterized by overgrowth, neurodevelopmental features, and absence of overt congenital anomalies. We have identified 15 individuals with de novo variants in codon 1740 of SETD2 whose features differ from those with LLS. Group 1 consists of 12 individuals with heterozygous variant c.5218C>T p.(Arg1740Trp) and Group 2 consists of 3 individuals with heterozygous variant c.5219G>A p.(Arg1740Gln). The phenotype of Group 1 includes microcephaly, profound intellectual disability, congenital anomalies affecting several organ systems, and similar facial features. Individuals in Group 2 had moderate to severe intellectual disability, low normal head circumference, and absence of additional major congenital anomalies. While LLS is likely due to loss of function of SETD2, the clinical features seen in individuals with variants affecting codon 1740 are more severe suggesting an alternative mechanism, such as gain of function, effects on epigenetic regulation, or posttranslational modification of the cytoskeleton. Our report is a prime example of different mutations in the same gene causing diverging phenotypes and the features observed in Group 1 suggest a new clinically recognizable syndrome uniquely associated with the heterozygous variant c.5218C>T p.(Arg1740Trp) in SETD2
Assessing the Quality of Digital Coproduction: An Interdisciplinary Model
The digital transformation of society raises high hopes for the improvement of the design and implementation of coproduction processes, which may lead to higher levels of acceptance, satisfaction, and trust with respect to public services. Yet and despite a growing number of studies on the role of digital technology on coproduction, our knowledge on how to assess the quality of digital coproduction is still very limited. The difficulty of defining quality indicators relates to three issues: the complexity of defining quality in public services, the lack of a clear understanding of quality in a coproduction process and the tendency to evaluate coproduction using the same approach as with other digitalised services. In this article, we adopt an interdisciplinary perspective to develop a conceptual model that builds on and bridges theoretical approaches commonly used to assess the quality of digital services, while adding elements that are specific to coproduction
VEGF-Rezeptor-spezifische Signaltransduktion ĂŒber Ras und Stickstoffmonoxid in Endothelzellen
Die intrazellulĂ€re Signaltransduktion des vaskulĂ€ren Endothelwachstumsfaktors (VEGF) wurde in humanen primĂ€ren und spontan transformierten Endothelzellen der Nabelschnurvene (HUVEC bzw. T-HUVEC) untersucht. Die VEGF-vermittelten Signalwege zu Zellteilung, Wanderung und Ăberleben wurden durch eine Rezeptor-SchimĂ€re, Rezeptor-spezifische Liganden und Signalweg-Inhibitoren im Vergleich analysiert. Das kleine G-Protein und Proto-Onkogen Ras sowie der gasförmige SignalĂŒbertrĂ€ger Stickstoffmonoxid wurde als wichtige Signaltransduktionsschaltstellen fĂŒr die Regulation der Endothelzellfunktionen durch VEGF identifiziert. Die Rolle von Ras bei der Entwicklung von T-HUVEC abgeleiteten Sarkomen im Nacktmaus-Tumorimplantationsmodell wurde mit einem funktionsblockierenden rekombinanten anti-Ras-Einzelketten-Antikörper untersucht. Die Inhibition von Ras reduzierte dabei die mitotische AktivitĂ€t, die Wachstumsgeschwindigkeit und das Endvolumen der T-HUVEC-Sarkome in NacktmĂ€usen

Verwaltung als Akteur â Akteure in der Verwaltung : aktuelle BeitrĂ€ge zur Debatte
Dieser Beitrag analysiert die jĂŒngere deutschsprachige Verwaltungswissenschaft im Hinblick darauf, mit welchen empirischen, theoretischen und methodischen Schwerpunktsetzungen sie Verwaltungen als kollektive Akteure einerseits und individuelle Akteure in der Verwaltung andererseits untersucht. Eine Verortung aktueller Studien ist geboten, um zu verstehen, wie Verwaltungswissenschaft nach Antworten auf die bestimmenden Fragen der Disziplin nach den Formen, ErklĂ€rungs- und Einflussfaktoren sowie Effekten von Verwaltungshandeln sucht. DafĂŒr fĂŒhren wir eine quantitative Analyse aller in der Zeitschrift âDer moderne Staat â Zeitschrift fĂŒr Public Policy, Recht und Managementâ (dms) von 2013 bis 2018/1 erschienenen Artikel durch. Im Ergebnis lĂ€sst sich eine groĂe Bandbreite von theoretischen Konzepten, aber eine geringe Zahl an theorietestenden und theoriegenerierenden Untersuchungen feststellen. Methodisch ĂŒberwiegen qualitative und deskriptive Studien. Der empirische Fokus der BeitrĂ€ge liegt auf einem LĂ€nderfokus auf Deutschland und auf Mehrebenenanalysen. Basierend auf identifizierten ForschungslĂŒcken schlieĂt der Beitrag mit Forschungsdesideraten fĂŒr zukĂŒnftige verwaltungswissenschaftliche Untersuchungen und fordert einen stĂ€rkeren Beitrag zur Weiterentwicklung von Theorien, mehr Ausgeglichenheit bei den methodischen AnsĂ€tzen und eine Erweiterung des empirischen Fokus der zukĂŒnftigen Forschung

Beyond the Great Wall: Gold of the Silk Roads and the First Empire of the Steppes
Fingerprinting ancient gold work requires the use of
nondestructive
techniques with high spatial resolution (down to 25 ÎŒm) and
good detection limits (micrograms per gram level). In this work experimental
setups and protocols for synchrotron radiation induced X-ray fluorescence
(SRXRF) at the BAMline of the Berlin electron storage ring company
for synchrotron radiation (BESSY) in Berlin for the measurement of
characteristic trace elements of gold are compared considering the
difficulties, shown in previous works, connected to the quantification
of Pt. The best experimental conditions and calculation methods were
achieved by using an excitation energy of 11.58 keV, a silicon drift
chamber detector (SDD) detector, and pure element reference standards.
A detection limit of 3 ÎŒg/g has been reached. This newly developed
method was successfully applied to provenancing the Xiongnu gold from
the Gol Mod necropolis, excavated under the aegis of the United Nations
Educational, Scientific and Cultural Organization (UNESCO). The composition
of the base alloys and the presence of Pt and Sn showed that, contrary
to what is expected, the gold foils from the first powerful empire
of the steppes along the Great Wall were produced with alluvial gold
from local placer deposits located in Zaamar, Boroo, and in the Selenga
River
