A Multi-Armed Bandit to Smartly Select a Training Set from Big Medical Data
With the availability of big medical image data, the selection of an adequate
training set is becoming more important to address the heterogeneity of
different datasets. Simply including all the data does not only incur high
processing costs but can even harm the prediction. We formulate the smart and
efficient selection of a training dataset from big medical image data as a
multi-armed bandit problem, solved by Thompson sampling. Our method assumes
that image features are not available at the time of the selection of the
samples, and therefore relies only on meta information associated with the
images. Our strategy simultaneously exploits data sources with high chances of
yielding useful samples and explores new data regions. For our evaluation, we
focus on the application of estimating the age from a brain MRI. Our results on
7,250 subjects from 10 datasets show that our approach leads to higher accuracy
while only requiring a fraction of the training data.Comment: MICCAI 2017 Proceeding
Importance of dose-schedule of 5-aza-2'-deoxycytidine for epigenetic therapy of cancer
<p>Abstract</p> <p>Background</p> <p>The inactivation of tumor suppressor genes (TSGs) by aberrant DNA methylation plays an important role in the development of malignancy. Since this epigenetic change is reversible, it is a potential target for chemotherapeutic intervention using an inhibitor of DNA methylation, such as 5-aza-2'-deoxycytidine (DAC). Although clinical studies show that DAC has activity against hematological malignancies, the optimal dose-schedule of this epigenetic agent still needs to be established.</p> <p>Methods</p> <p>Clonogenic assays were performed on leukemic and tumor cell lines to evaluate the <it>in vitro </it>antineoplastic activity of DAC. The reactivation of TSGs and inhibition of DNA methylation by DAC were investigated by reverse transcriptase-PCR and Line-1 assays. The <it>in vivo </it>antineoplastic activity of DAC administered as an i.v. infusion was evaluated in mice with murine L1210 leukemia by measurement of survival time, and in mice bearing murine EMT6 mammary tumor by excision of tumor after chemotherapy for an <it>in vitro </it>clonogenic assay.</p> <p>Results</p> <p>Increasing the DAC concentration and duration of exposure produced a greater loss of clonogenicity for both human leukemic and tumor cell lines. The reactivation of the TSGs (<it>p57KIP2 </it>in HL-60 leukemic cells and <it>p16CDKN2A </it>in Calu-6 lung carcinoma cells) and the inhibition of global DNA methylation in HL-60 leukemic cells increased with DAC concentration. In mice with L1210 leukemia and in mice bearing EMT6 tumors, the antineoplastic action of DAC also increased with the dose. The plasma level of DAC that produced a very potent antineoplastic effect in mice with leukemia or solid tumors was > 200 ng/ml (> 1 μM).</p> <p>Conclusion</p> <p>We have shown that intensification of the DAC dose markedly increased its antineoplastic activity in mouse models of cancer. Our data also show that there is a good correlation between the concentrations of DAC that reduce <it>in vitro </it>clonogenicity, reactivate TSGs and inhibit DNA methylation. These results suggest that the antineoplastic action of DAC is related to its epigenetic action. Our observations provide a strong rationale to perform clinical trials using dose intensification of DAC to maximize the chemotherapeutic potential of this epigenetic agent in patients with cancer.</p
Escape rate and Hausdorff measure for entire functions
The escaping set of an entire function is the set of points that tend to
infinity under iteration. We consider subsets of the escaping set defined in
terms of escape rates and obtain upper and lower bounds for the Hausdorff
measure of these sets with respect to certain gauge functions.Comment: 24 pages; some errors corrected, proof of Theorem 2 shortene
Tuning a Circular p-n Junction in Graphene from Quantum Confinement to Optical Guiding
The motion of massless Dirac-electrons in graphene mimics the propagation of
photons. This makes it possible to control the charge-carriers with components
based on geometrical-optics and has led to proposals for an all-graphene
electron-optics platform. An open question arising from the possibility of
reducing the component-size to the nanometer-scale is how to access and
understand the transition from optical-transport to quantum-confinement. Here
we report on the realization of a circular p-n junction that can be
continuously tuned from the nanometer-scale, where quantum effects are
dominant, to the micrometer scale where optical-guiding takes over. We find
that in the nanometer-scale junction electrons are trapped in states that
resemble atomic-collapse at a supercritical charge. As the junction-size
increases, the transition to optical-guiding is signaled by the emergence of
whispering-gallery modes and Fabry-Perot interference. The creation of tunable
junctions that straddle the crossover between quantum-confinement and
optical-guiding, paves the way to novel design-architectures for controlling
electronic transport.Comment: 16 pages, 4 figure
Gendered Representations of Male and Female Social Actors in Iranian Educational Materials
This research investigates the representations of gendered social actors within the subversionary discourse of equal educational opportunities for males and females in Iranian English as a Foreign Language (EFL) books. Using critical discourse analysis (CDA) as the theoretical framework, the authors blend van Leeuwen’s (Texts and practices: Readings in critical discourse analysis, Routledge, London, 2003) ‘Social Actor Network Model’ and Sunderland’s (Gendered discourses, Palgrave Macmillan, Hampshire, 2004) ‘Gendered Discourses Model’ in order to examine the depictions of male and female social actors within this gendered discourse. The gendered discourse of equal opportunities was buttressed by such representations within a tight perspective in proportion to gender ideologies prevailing in Iran. Resorting to CDA, we can claim that resistance against such gendered discourse in Iranian EFL textbooks militates against such gender norms. These representations of male and female social actors in school books are indicative of an all-encompassing education, reinforcing that the discourse of equal opportunities is yet to be realized in the education system of Iran
The magic nature of 132Sn explored through the single-particle states of 133Sn
Atomic nuclei have a shell structure where nuclei with 'magic numbers' of
neutrons and protons are analogous to the noble gases in atomic physics. Only
ten nuclei with the standard magic numbers of both neutrons and protons have so
far been observed. The nuclear shell model is founded on the precept that
neutrons and protons can move as independent particles in orbitals with
discrete quantum numbers, subject to a mean field generated by all the other
nucleons. Knowledge of the properties of single-particle states outside nuclear
shell closures in exotic nuclei is important for a fundamental understanding of
nuclear structure and nucleosynthesis (for example the r-process, which is
responsible for the production of about half of the heavy elements). However,
as a result of their short lifetimes, there is a paucity of knowledge about the
nature of single-particle states outside exotic doubly magic nuclei. Here we
measure the single-particle character of the levels in 133Sn that lie outside
the double shell closure present at the short-lived nucleus 132Sn. We use an
inverse kinematics technique that involves the transfer of a single nucleon to
the nucleus. The purity of the measured single-particle states clearly
illustrates the magic nature of 132Sn.Comment: 19 pages, 5 figures and 4 table
Criminal and Noncriminal Psychopathy: The Devil is in the Detail
Brooks, NS ORCiD: 0000-0003-1784-099XPsychopathy is prevalent and problematic in criminal populations, but is also found to be present in noncriminal populations. In 1992, Robert Hare declared that psychopaths may also “be found in the boardroom”, which has since been followed by an interest in the issue of noncriminal, or even successful, psychopathy. In this chapter, the paradox of criminal and noncriminal psychopathy is discussed with specific attention given to the similarities and differences that account for psychopathic personality across contexts. That psychopathy is a condition typified by a constellation of traits and behaviours requires wider research across diverse populations, and thus the streams of research related to criminal and noncriminal psychopathy are presented and the implications of these contrasting streams are explored
Isoforms of U1-70k control subunit dynamics in the human spliceosomal U1 snRNP
Most human protein-encoding genes contain multiple exons that are spliced together, frequently in alternative arrangements, by the spliceosome. It is established that U1 snRNP is an essential component of the spliceosome, in human consisting of RNA and ten proteins, several of which are post- translationally modified and exist as multiple isoforms. Unresolved and challenging to investigate are the effects of these post translational modifications on the dynamics, interactions and stability of the particle. Using mass spectrometry we investigate the composition and dynamics of the native human U1 snRNP and compare native and recombinant complexes to isolate the effects of various subunits and isoforms on the overall stability. Our data reveal differential incorporation of four protein isoforms and dynamic interactions of subunits U1-A, U1-C and Sm-B/B’. Results also show that unstructured post- ranslationally modified C-terminal tails are
responsible for the dynamics of Sm-B/B’ and U1-C and that their interactions with the Sm core are controlled by binding to different U1-70k isoforms and their phosphorylation status in vivo. These results therefore provide the important functional link between proteomics and structure as well as insight into the dynamic quaternary structure of the native U1 snRNP important for its function. This work was funded by: BBSRC (OVM), BBSRC and EPSRC (HH and NM), EU Prospects (HH), European Science Foundation (NM), the Royal Society (CVR), and fellowship from JSPS and HFSP (YM and DAPK respectively)
