Framework For Improving Complex System Performance
This paper introduces a framework for improvement of complex system performance. Complex systems are besieged with conditions marked by increasing uncertainty, emergence, and ambiguity. Additionally, demands for increased productivity, resource efficiencies, and performance improvement make new approaches paramount for modern systems engineers. In response, a framework to improve complex system performance is developed. Following an introduction, the paper pursues four objectives: (1) introduction of Complex System Governance (CSG) as a foundation to describe essential system functions, (2) suggest system `pathologies\u27 as an explanation for deep system performance issues, (3) exploration of system performance improvement as a function of `requisite variety\u27 to compensate for deep system issues, and (4) introduce a framework for complex system performance improvement using system pathologies as `unab-sorbed variety\u27. The paper closes with some challenges for further development of the framework for deployment and application guidance for practitioners

A genomic island integrated into recA of Vibrio cholerae contains a divergent recA and provides multi-pathway protection from DNA damage
© 2015 Society for Applied Microbiology and John Wiley & Sons Ltd. Lateral gene transfer (LGT) has been crucial in the evolution of the cholera pathogen, Vibrio cholerae. The two major virulence factors are present on two different mobile genetic elements, a bacteriophage containing the cholera toxin genes and a genomic island (GI) containing the intestinal adhesin genes. Non-toxigenic V.cholerae in the aquatic environment are a major source of novel DNA that allows the pathogen to morph via LGT. In this study, we report a novel GI from a non-toxigenic V.cholerae strain containing multiple genes involved in DNA repair including the recombination repair gene recA that is 23% divergent from the indigenous recA and genes involved in the translesion synthesis pathway. This is the first report of a GI containing the critical gene recA and the first report of a GI that targets insertion into a specific site within recA. We show that possession of the island in Escherichia coli is protective against DNA damage induced by UV-irradiation and DNA targeting antibiotics. This study highlights the importance of genetic elements such as GIs in the evolution of V.cholerae and emphasizes the importance of environmental strains as a source of novel DNA that can influence the pathogenicity of toxigenic strains
Perceptual Context in Cognitive Hierarchies
Cognition does not only depend on bottom-up sensor feature abstraction, but
also relies on contextual information being passed top-down. Context is higher
level information that helps to predict belief states at lower levels. The main
contribution of this paper is to provide a formalisation of perceptual context
and its integration into a new process model for cognitive hierarchies. Several
simple instantiations of a cognitive hierarchy are used to illustrate the role
of context. Notably, we demonstrate the use context in a novel approach to
visually track the pose of rigid objects with just a 2D camera
A 15.65 solar mass black hole in an eclipsing binary in the nearby spiral galaxy Messier 33
Stellar-mass black holes are discovered in X-ray emitting binary systems,
where their mass can be determined from the dynamics of their companion stars.
Models of stellar evolution have difficulty producing black holes in close
binaries with masses >10 solar masses, which is consistent with the fact that
the most massive stellar black holes known so all have masses within 1 sigma of
10 solar masses. Here we report a mass of 15.65 +/- 1.45 solar masses for the
black hole in the recently discovered system M33 X-7, which is located in the
nearby galaxy Messier 33 (M33) and is the only known black hole that is in an
eclipsing binary. In order to produce such a massive black hole, the progenitor
star must have retained much of its outer envelope until after helium fusion in
the core was completed. On the other hand, in order for the black hole to be in
its present 3.45 day orbit about its 70.0 +/- 6.9 solar mass companion, there
must have been a ``common envelope'' phase of evolution in which a significant
amount of mass was lost from the system. We find the common envelope phase
could not have occured in M33 X-7 unless the amount of mass lost from the
progenitor during its evolution was an order of magnitude less than what is

Joseph Benson Mather was the eldest son of Robert Mather (1782-1855) and Ann (d.1831) daughter of Rev. Joseph Benson. Robert Mather arrived in VDL in 1822 and set up in business at the corner of Elizabeth & Liverpool Sts in Hobart, and became a trustee of the Wesleyan Church. He received a grant of land at Lauderdale but his farm was unsuccessful and he went into business with his two elder sons, Joseph Benson (1814-1890) and Robert Andrew (1815-1884). In 1834 he and some of his family joined the Society of Friends, and his daughter Sarah Benson married the Quaker missionary George Washington Walker (1800-1859) in 1840. Joseph Benson married Anna Maria Cotton and had a son, (Joseph) Francis (d. 1925) and three daughters, one of whom married C H Robey. Robert Mather's second wife, whom he married in 1842, was Esther Dixon
The Association between Asthma and Allergic Symptoms in Children and Phthalates in House Dust: A Nested Case–Control Study
Global phthalate ester production has increased from very low levels at the end of World War II to approximately 3.5 million metric tons/year. The aim of the present study was to investigate potential associations between persistent allergic symptoms in children, which have increased markedly in developed countries over the past three decades, and the concentration of phthalates in dust collected from their homes. This investigation is a case–control study nested within a cohort of 10,852 children. From the cohort, we selected 198 cases with persistent allergic symptoms and 202 controls without allergic symptoms. A clinical and a technical team investigated each child and her or his environment. We found higher median concentrations of butyl benzyl phthalate (BBzP) in dust among cases than among controls (0.15 vs. 0.12 mg/g dust). Analyzing the case group by symptoms showed that BBzP was associated with rhinitis (p = 0.001) and eczema (p = 0.001), whereas di(2-ethylhexyl) phthalate (DEHP) was associated with asthma (p = 0.022). Furthermore, dose–response relationships for these associations are supported by trend analyses. This study shows that phthalates, within the range of what is normally found in indoor environments, are associated with allergic symptoms in children. We believe that the different associations of symptoms for the three major phthalates—BBzP, DEHP, and di-n-butyl phthalate—can be explained by a combination of chemical physical properties and toxicologic potential. 15-deoxy-Delta(12,14)-Prostaglandin J(2) inhibits human soluble epoxide hydrolase by a dual orthosteric and allosteric mechanism
Human soluble epoxide hydrolase (hsEH) is an enzyme responsible for the inactivation of bioactive epoxy fatty acids, and its inhibition is emerging as a promising therapeutical strategy to target hypertension, cardiovascular disease, pain and insulin sensitivity. Here, we uncover the molecular bases of hsEH inhibition mediated by the endogenous 15-deoxy-Δ12,14-Prostaglandin J2 (15d-PGJ2). Our data reveal a dual inhibitory mechanism, whereby hsEH can be inhibited by reversible docking of 15d-PGJ2 in the catalytic pocket, as well as by covalent locking of the same compound onto cysteine residues C423 and C522, remote to the active site. Biophysical characterisations allied with in silico investigations indicate that the covalent modification of the reactive cysteines may be part of a hitherto undiscovered allosteric regulatory mechanism of the enzyme. This study provides insights into the molecular modes of inhibition of hsEH epoxy-hydrolytic activity and paves the way for the development of new allosteric inhibitors
Mosquito Abundance, Bed net Coverage and Other Factors Associated with Variations in Sporozoite Infectivity Rates in Four Villages of Rural Tanzania.
Entomological surveys are of great importance in decision-making processes regarding malaria control strategies because they help to identify associations between vector abundance both species-specific ecology and disease intervention factors associated with malaria transmission. Sporozoite infectivity rates, mosquito host blood meal source, bed net coverage and mosquito abundance were assessed in this study. A longitudinal survey was conducted in four villages in two regions of Tanzania. Malaria vectors were sampled using the CDC light trap and pyrethrum spray catch methods. In each village, ten paired houses were selected for mosquitoes sampling. Sampling was done in fortnight case and study was undertaken for six months in both Kilimanjaro (Northern Tanzania) and Dodoma (Central Tanzania) regions. A total of 6,883 mosquitoes were collected including: 5,628 (81.8%) Anopheles arabiensis, 1,100 (15.9%) Culex quinquefasciatus, 89 (1.4%) Anopheles funestus, and 66 (0.9%) Anopheles gambiae s.s. Of the total mosquitoes collected 3,861 were captured by CDC light trap and 3,022 by the pyrethrum spray catch method. The overall light trap: spray catch ratio was 1.3:1. Mosquito densities per room were 96.5 and 75.5 for light trap and pyrethrum spray catch respectively. Mosquito infectivity rates between villages that have high proportion of bed net owners and those without bed nets was significant (P < 0.001) and there was a significant difference in sporozoite rates between households with and without bed nets in these four villages (P < 0.001). Malaria remains a major problem in the study areas characterized as low transmission sites. Further studies are required to establish the annual entomological inoculation rates and to observe the annual parasitaemia dynamics in these communities.
