    Beta-amyloid peptide blocks the fast-inactivating K+ current in rat hippocampal neurons

    Deposition of beta-amyloid peptide (A beta) in senile plaques is a hallmark of Alzheimer disease neuropathology. Chronic exposure of neuronal cultures to synthetic A beta is directly toxic, or enhances neuronal susceptibility to excitotoxins. Exposure to A beta may cause a loss of cellular calcium homeostasis, but the mechanism by which this occurs is uncertain. In this work, the acute response of rat hippocampal neurons to applications of synthetic A beta was measured using whole-cell voltage-clamp techniques. Pulse application of A beta caused a reversible voltage-dependent decrease in membrane conductance. A beta selectively blocked the voltage-gated fast-inactivating K+ current, with an estimated KI < 10 microM. A beta also blocked the delayed rectifying current, but only at the highest concentration tested. The response was independent of aggregation state or peptide length. The dynamic response of the fast-inactivating current to a voltage jump was consistent with a model whereby A beta binds reversibly to closed channels and prevents their opening. Blockage of fast-inactivating K+ channels by A beta could lead to prolonged cell depolarization, thereby increasing Ca2+ influx

    Carbon monoxide fluxes measured using the eddy covariance method from an intensively managed grassland in Ireland

    Carbon monoxide (CO) is classed as a secondary greenhouse gas (GHG) as it can extend the lifetime of GHGs such as methane and ozone by reacting with hydroxyl (OH) radicals and thus controlling the oxidizing capacity of the atmosphere. CO fluxes were measured from a fertilized and grazed temperate grassland in south-east Ireland using a high frequency quantum cascade laser (QCL) and the eddy covariance method. Measurements were carried out in 2019 (12 months) and 2020 (10 months). In both datasets, a diurnal pattern was observed where CO fluxes peaked between 8 am and 6 pm, and between 8 pm and 2 am during the colder months of the year. Quality controlled CO flux data was gap-filled using a general additive model (GAM) that incorporated photosynthetic photon flux density (PPFD), air temperature, water-filled pore space (WFPS) and month of the year. Modelled CO emissions showed less divergence with measured CO fluxes in 2020 compared to 2019, suggesting that the environmental variables in the GAM were the predominant driver of CO emissions in 2020 but not in 2019. Cumulative CO emissions in 2019 and 2020 were 39.7 ± 147.1 mgCO–C per m2 per month and 31.5 ± 75.2 mgCO–C per m2 per month, respectively. While soils are typically considered a sink of CO, our results show that managed pastures can act as a source of CO and thus warrant further investigation into the implications CO may have on GHG dynamics from agricultural landscapes

    Joint modelling of multiple network wiews

    Latent space models (LSM) for network data were introduced by Hoff et al. (2002) under the basic assumption that each node of the network has an unknown position in a D-dimensional Euclidean latent space: generally the smaller the distance between two nodes in the latent space, the greater their probability of being connected. In this paper we propose a variational inference approach to estimate the intractable posterior of the LSM. In many cases, different network views on the same set of nodes are available. It can therefore be useful to build a model able to jointly summarise the information given by all the network views. For this purpose, we introduce the latent space joint model (LSJM) that merges the information given by multiple network views assuming that the probability of a node being connected with other nodes in each network view is explained by a unique latent variable. This model is demonstrated on the analysis of two datasets: an excerpt of 50 girls from 'Teenage Friends and Lifestyle Study' data at three time points and the Saccharomyces cerevisiae genetic and physical protein-protein interactions

    Diffraction Symmetry in Crystalline, Close-Packed C60

    We have grown crystals of the carbon structure C60 by sublimation. In contrast to solution-grown crystals, the sublimed crystals have long range order with no evidence of solvent inclusions. Sublimed C60 forms three dimensional, faceted crystals with a close-packed, face-centered cubic unit cell. We have refined a crystal structure using the "soccer ball" model of the C60 molecule. The results indicate that the C60 molecule has the expected spherical shape, however the data are not sufficiently accurate to unambiguously determine atomic positions

    Targeted nasal vaccination provides antibody-independent protection against Staphylococcus aureus.

    Despite showing promise in preclinical models, anti-Staphylococcus aureus vaccines have failed in clinical trials. To date, approaches have focused on neutralizing/opsonizing antibodies; however, vaccines exclusively inducing cellular immunity have not been studied to formally test whether a cellular-only response can protect against infection. We demonstrate that nasal vaccination with targeted nanoparticles loaded with Staphylococcus aureus antigen protects against acute systemic S. aureus infection in the absence of any antigen-specific antibodies. These findings can help inform future developments in staphylococcal vaccine development and studies into the requirements for protective immunity against S. aureus

    Mott Transition in Degenerate Hubbard Models: Application to Doped Fullerenes

    The Mott-Hubbard transition is studied for a Hubbard model with orbital degeneracy N, using a diffusion Monte-Carlo method. Based on general arguments, we conjecture that the Mott-Hubbard transition takes place for U/W \propto \sqrt{N}, where U is the Coulomb interaction and W is the band width. This is supported by exact diagonalization and Monte-Carlo calculations. Realistic parameters for the doped fullerenes lead to the conclusion that stoichiometric A_3 C_60 (A=K, Rb) are near the Mott-Hubbard transition, in a correlated metallic state.Comment: 4 pages, revtex, 1 eps figure included, to be published in Phys.Rev.B Rapid Com
