    Oral salmon calcitonin reduces cartilage and bone pathology in an osteoarthritis rat model with increased subchondral bone turnover

    SummaryObjectivesTraumatic osteoarthritis (OA) is possibly augmented by effects from loss of sex hormones. Salmon calcitonin is shown to reduce OA pathogenesis and bone resorption. We investigated the effects of oral salmon calcitonin treatment and ovariectomy on cartilage and bone pathology in a traumatic OA model.MethodsSix groups with 10 7-month-old female Sprague Dawley rats each were subjected to bilateral meniscectomy (MNX), ovariectomy (OVX) or Sham surgery and treated for 8 weeks with oral salmon calcitonin (CT) or vehicle (V) in the following way: (1) Sham+V; (2) MNX+V; (3) MNX+CT; (4) OVX+V; (5) MNX/OVX+V; (6) MNX/OVX+CT. Weights were recorded weekly and CTX-II was measured in serum. At termination 56 days post-surgery, the right tibia was analyzed for changes in articular cartilage thickness, extent of cartilage damage and subchondral bone changes in predefined zones, as recommended in the novel OARSI histopathology score.ResultsThe combined MNX/OVX model produced a significantly reduced cartilage thickness (P=0.033) in the outer zone (Z1) of the tibial plateau and increased calcified cartilage damage (P=0.0004) and serum CTX-II (P=0.003). Addition of OVX to MNX significantly increased the width of matrix damage at the surface (P=0.025) and 50% cartilage depth (P=0.004). Treatment with oral salmon calcitonin counteracted the loss of cartilage thickness (P=0.055), significantly reduced subchondral bone damage score (P=0.019) and reduced the type II collagen degradation (P=0.009).ConclusionsAddition of ovariectomy augmented site-specific traumatic OA pathology, which was reduced by oral salmon calcitonin treatment. Treatments for OA might ideally affect both bone and cartilage

    Phenomenological theory of superradiation from crystals of high-spin molecules is suggested. We show that radiation friction can cause a superradiation pulse and investigate the role of magnetic anisotropy, external magnetic field and dipole-dipole interactions. Depending on the contribution of all these factors at low temperature, several regimes of magnetization of crystal sample are described.     The Middle Way: East Asian masters students’ perceptions of critical argumentation in U.K. universities.

    The paper explores the learning experiences of East Asian masters students in dealing with Western academic norms of critical thinking in classroom debate and assignment writing. The research takes a cultural approach, and employs grounded theory and case study methodology, the aims being for students to explain their perceptions of their personal learning journeys. The data suggest that the majority of students interviewed rejected full academic acculturation into Western norms of argumentation. They instead opted for a ‘Middle Way’ that synergizes the traditional cultural academic values held by many East Asian students with those elements of Western academic norms that are perceived to be aligned with these. This is a relatively new area of research which represents a challenge for British lecturers and students

    Helioseismic Holography of an Artificial Submerged Sound Speed Perturbation and Implications for the Detection of Pre-Emergence Signatures of Active Regions

    We use a publicly available numerical wave-propagation simulation of Hartlep et al. 2011 to test the ability of helioseismic holography to detect signatures of a compact, fully submerged, 5% sound-speed perturbation placed at a depth of 50 Mm within a solar model. We find that helioseismic holography as employed in a nominal "lateral-vantage" or "deep-focus" geometry employing quadrants of an annular pupil is capable of detecting and characterizing the perturbation. A number of tests of the methodology, including the use of a plane-parallel approximation, the definition of travel-time shifts, the use of different phase-speed filters, and changes to the pupils, are also performed. It is found that travel-time shifts made using Gabor-wavelet fitting are essentially identical to those derived from the phase of the Fourier transform of the cross-covariance functions. The errors in travel-time shifts caused by the plane-parallel approximation can be minimized to less than a second for the depths and fields of view considered here. Based on the measured strength of the mean travel-time signal of the perturbation, no substantial improvement in sensitivity is produced by varying the analysis procedure from the nominal methodology in conformance with expectations. The measured travel-time shifts are essentially unchanged by varying the profile of the phase-speed filter or omitting the filter entirely. The method remains maximally sensitive when applied with pupils that are wide quadrants, as opposed to narrower quadrants or with pupils composed of smaller arcs. We discuss the significance of these results for the recent controversy regarding suspected pre-emergence signatures of active regions

    Azithromycin Failure in Mycoplasma genitalium Urethritis

    We report significant failure rates (28%, 95% confidence interval 15%–45%) after administering 1 g azithromycin to men with Mycoplasma genitalium–positive nongonococcal urethritis. In vitro evidence supported reduced susceptibility of M. genitalium to macrolides. Moxifloxacin administration resulted in rapid symptom resolution and eradication of infection in all cases. These findings have implications for management of urethritis

    Tunneling and the Spectrum of the Potts Model

    The three-dimensional, three-state Potts model is studied as a paradigm for high temperature quantum chromodynamics. In a high statistics numerical simulation using a Swendson-Wang algorithm, we study cubic lattices of dimension as large as 64364^3 and measure correlation functions on long lattices of dimension 202×12020^2\times 120 and 302×12030^2\times 120. These correlations are controlled by the spectrum of the transfer matrix. This spectrum is studied in the vicinity of the phase transition. The analysis classifies the spectral levels according to an underlying S3S_3 symmetry. Near the phase transition the spectrum agrees nicely with a simple four-component hamiltonian model. In the context of this model, we find that low temperature ordered-ordered interfaces nearly always involve a disordered phase intermediate.     Fecal Immunochemical Test Screening and Risk of Colorectal Cancer Death.

    The fecal immunochemical test (FIT) is widely used for colorectal cancer (CRC) screening, but evidence of its effectiveness is limited. To evaluate whether FIT screening is associated with a lower risk of dying from CRC overall, according to cancer location, and within demographic groups. This nested case-control study in a cohort of screening-eligible people was conducted in 2 large, integrated health systems of racially, ethnically, and socioeconomically diverse members with long-term programs of mailed FIT screening outreach. Eligible participants included people aged 52 to 85 years who died from colorectal adenocarcinoma between 2011 and 2017 (cases); cases were matched in a 1:8 ratio based on age, sex, health-plan membership duration, and geographic area to randomly selected persons who were alive and CRC-free on case's diagnosis date (controls). Data analysis was conducted from January 2002 to December 2017. Completing 1 or more FIT screenings in the 5-year period prior to the CRC diagnosis date among cases or the corresponding date among controls; in secondary analyses, 2- to 10-year intervals were evaluated. The primary study outcome was CRC death overall and by tumor location. Secondary analyses were performed to assess CRC death by race and ethnicity. From a cohort of 2 127 128 people, a total of 10 711 participants (3529 aged 60-69 years [32.9%]; 5587 male [52.1%] and 5124 female [47.8%]; 1254 non-Hispanic Asian [11.7%]; 973 non-Hispanic Black [9.1%]; 1929 Hispanic or Latino [18.0%]; 6345 non-Hispanic White [59.2%]) was identified, including 1103 cases and 9608 controls. Among controls during the 10-year period prior to the reference date, 6101 (63.5%) completed 1 or more FITs with a cumulative 12.6% positivity rate (768 controls), of whom 610 (79.4%) had a colonoscopy within 1 year. During the 5-year period, 494 cases (44.8%) and 5345 controls (55.6%) completed 1 or more FITs. In regression analysis, completing 1 or more FIT screening was associated with a 33% lower risk of death from CRC (adjusted odds ratio [aOR], 0.67; 95% CI, 0.59-0.76) and 42% lower risk in the left colon and rectum (aOR, 0.58; 95% CI, 0.48-0.71). There was no association with right colon cancers (aOR, 0.83; 95% CI, 0.69-1.01) but the difference in the estimates between the right colon and left colon or rectum was statistically significant (P = .01). FIT screening was associated with lower CRC mortality risk among non-Hispanic Asian (aOR, 0.37; 95% CI, 0.23-0.59), non-Hispanic Black (aOR, 0.58; 95% CI, 0.39-0.85) and non-Hispanic White individuals (aOR, 0.70; 95% CI, 0.57-0.86) (P for homogeneity = .04 for homogeneity). In this nested case-control study, completing FIT was associated with a lower risk of overall death from CRC, particularly in the left colon, and the associations were observed across racial and ethnic groups. These findings support the use of FIT in population-based screening strategies

    Control of an Energy Efficient Hydraulic Cylinder Drive with Multiple Pressure Lines

    This paper concerns a novel energy efficient hydraulic cylinder drive concept, consisting of three pressure lines used to supply a hydraulic cylinder(s). The proposed concept allows for reduced throttling losses compared to conventional solutions, while maintaining accurately cylinder control. One application where the concept shows great potential is as the actuation system for life tests of large mechanical structures, where structure deflections is made through cylinder force control. This work contributes to the development and control of this hydraulic concept, where the purpose is to obtain a high energy efficiency and an accurate cylinder force control. The final concept design is implemented in simulation models, where the performance of the developed control system is investigated to verify that the control performance is satisfactory. Furthermore, the energy efficiency is compared to a conventional hydraulic concept to verify the feasibility. Through simulation models, control performance similar to a conventional hydraulic concept has been obtained with the proposed control structure, while reducing the hydraulic energy consumption by up to 50%. Based on the findings in this paper it is assessed that the concept is ready for experimental validation

    Dynamics of tournaments: the soccer case

    A random walk-like model is considered to discuss statistical aspects of tournaments. The model is applied to soccer leagues with emphasis on the scores. This competitive system was computationally simulated and the results are compared with empirical data from the English, the German and the Spanish leagues and showed a good agreement with them. The present approach enabled us to characterize a diffusion where the scores are not normally distributed, having a short and asymmetric tail extending towards more positive values. We argue that this non-Gaussian behavior is related with the difference between the teams and with the asymmetry of the scores system. In addition, we compared two tournament systems: the all-play-all and the elimination tournaments.Comment: To appear in EPJ
