    Subcellular optogenetic activation of Cdc42 controls local and distal signaling to drive immune cell migration

    Migratory immune cells use intracellular signaling networks to generate and orient spatially polarized responses to extracellular cues. The monomeric G protein Cdc42 is believed to play an important role in controlling the polarized responses, but it has been difficult to determine directly the consequences of localized Cdc42 activation within an immune cell. Here we used subcellular optogenetics to determine how Cdc42 activation at one side of a cell affects both cell behavior and dynamic molecular responses throughout the cell. We found that localized Cdc42 activation is sufficient to generate polarized signaling and directional cell migration. The optically activated region becomes the leading edge of the cell, with Cdc42 activating Rac and generating membrane protrusions driven by the actin cytoskeleton. Cdc42 also exerts long-range effects that cause myosin accumulation at the opposite side of the cell and actomyosin-mediated retraction of the cell rear. This process requires the RhoA-activated kinase ROCK, suggesting that Cdc42 activation at one side of a cell triggers increased RhoA signaling at the opposite side. Our results demonstrate how dynamic, subcellular perturbation of an individual signaling protein can help to determine its role in controlling polarized cellular responses

    Retinal dysplasia represents a congenital disorder characterized by abnormal proliferation of retinal tissue causing leukocoria. We present a case of an infant with bilateral leukocoria, clinically diagnosed as retinoblastoma, followed by enucleation of the left eye. Microscopy however, demonstrated retinal dysplasia consisting of a disorderly proliferation of retinal tissue with formation of rosettes, mimicking retinoblastoma. Microscopic features that aid in differentiating this lesion from retinoblastoma are discussed

    With the exponential growth of mobile users, there is a massive growth of data as well as novel services to support such data management. However, the existing 4G network is absolutely not meant for catering up such higher demands of bandwidth utilization as well as servicing massive users with similar Quality of service. Such problems are claimed to be effectively addressed by the adoption of 5G networking system. Although the characteristics of 5G networking are theoretically sound, still it is under the roof of the research. Therefore, this paper presents a discussion about the conventional approach as well as an approach using cognitive radio network towards addressing the frequently identified problems of energy, resource allocation, and spectral efficiency. The study collects the existing, recent researches in the domain of 5G communications from various publications. Different from existing review work, the paper also contributes towards identifying the core research findings as well as a significant research gap towards improving the communication in the 5G network system

    Adaptations of the antioxidant system in erythrocytes of trained adult rats: Impact of intermittent hypobaric-hypoxia at two altitudes

    We have investigated the effects of daily exposure to intermittent hypobaric-hypoxia to two simulated altitudes (5700 m and 6300 m) in adult male rats that had been regularly swim trained in normoxia at sea level prior to exposures. Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) along with the oxidative stress (OS) indices, malondialdehyde (MDA) and protein carbonyl content were measured in erythrocytes and their membranes. Hemoglobin increased in the trained animals exposed to 5700 m and in untrained rats exposed to 6300 m. Osmotic fragility in terms of hemolysis increased in altitude exposed animals. SOD increased in those exposed to 6300 m, while CAT increased in trained rats exposed to 5700 m and to 6300 m unlike in untrained rats where CAT increased only at 6300 m. GSH-Px showed varying degrees of elevation in all animals exposed to both altitudes. Erythrocyte membranes showed significant elevations in malondialdehyde (MDA) at 6300 m, while elevated protein carbonyls were noticeable at both altitudes in whole cells and membranes. These results suggest a positively associated elevation in protein oxidation with altitude in trained rats. At 5700 m, animals were less stressed, unlike at 6300 m, as seen from the magnitude of elevations in the OS indices and from the responses of the antioxidant enzymes. © 2005 Elsevier Inc.     Insulin resistance and its relation to inflammatory status and serum lipids among young women with PCOS

    Background: The incidence of polycystic ovarian syndrome (PCOS) is increasing among young women. PCOS women have decreased insulin sensitivity independent of body mass index with increase in lipid levels. Studies on measuring inflammatory status in PCOS showed varying results. The inter-relationship between inflammatory status, insulin resistance and lipid levels among PCOS women was studied. Methods: Twenty PCOS women and 20 healthy controls of age 18-25 years were recruited. Fasting blood samples were collected for estimation of serum glucose, insulin levels, lipid levels and C-reactive protein (CRP) concentration. Insulin resistance was determined by homeostasis model assessment (HOMA-IR) index. Results: PCOS women had significant increase in fasting insulin, HOMA-IR and triglycerides compared to healthy controls. HOMA-IR was positively associated with serum triglycerides, VLDL levels and CRP levels among PCOS subjects. Total Cholesterol was positively associated with CRP. Regression analysis showed HOMA-IR as a sole parameter strongly linked with PCOS women. This indicates that, IR is an independent pathogenic variable linked with PCOS which in turn showed positive association with CRP and triglycerides. Conclusion: IR is the hallmark of PCOS among young adolescent women. IR is associated with elevated CRP and triglyceride levels. Taking measures to increase insulin sensitivity, may help in altering dyslipidemia and inflammatory status, thereby reducing CVD risk among young women with PCOS

    Four Element Microstrip Antenna Array with Electromagnetic Band Gap Structure and Silver Material Deposition for Reduced Mutual Coupling

    This paper presents the performance of four element microstrip antenna array. The overall bandwidth of the microstrip antenna array with silver material coating is equal to 228.53 % as compared to 75.91 % without silver material coating. The microstrip antenna array with silver material coating of 30 mm thickness is producing good reduction in mutual coupling values at the resonant frequency of 5.53 GHz. Moreover, the radiation properties of antenna array are improved with good reduction in power radiated in the undesired direction. FR-4 glass epoxy substrate is used as dielectric substrate which has a dielectric constant of 4.2 and loss tangent of 0.0245. The microstrip antenna arrays are designed using Mentor Graphics IE3D software and the measured results are obtained using vector network analyzer
