Colheita foliar da cancorosa [Sorocea bonplandii (Baill.) Burg., Lanj. & W. Boer]: uma espinheira-santa da Mata Atlântica.
Sorocea bonplandii, conhecida por cancorosa ou falsa-espinheira-santa, Ă© uma pequena árvore abundante no sub-bosque florestal da Mata Atlântica. A espĂ©cie tem reconhecido valor medicinal e Ă© amplamente usada na medicina popular. Neste contexto o objetivo deste trabalho foi avaliar o potencial de produção foliar e estratĂ©gias para a colheita foliar de S. bonplandii em florestas nativas visando subsidiar estratĂ©gias de uso sustentável da espĂ©cie. Os resultados indicaram que a espĂ©cie permite a poda de ramos, pois nĂŁo foram identificados problemas quanto Ă regeneração pĂłs-poda. As avaliações nas florestas secundárias de Santa Catarina demonstraram ainda a superioridade das formações de Floresta Estacional Decidual (FED) sobre a Floresta OmbrĂłfila Densa (FOD), em termos de quantidade e qualidade de folhas produzidas por planta. Na FED, alĂ©m de ser dominante no sub-bosque, a espĂ©cie ainda apresenta qualidade foliar superior em comparação Ă FOD, pois na FED aproximadamente menos de 10% da biomassa de folhas colhidas foram classificadas de pĂ©ssima qualidade, comparativamente a 30%, na FOD. A estimativa de produtividade foi de 64 kg de massa foliar seca por hectare, revelando o grande potencial para manejo da espĂ©cie. Embora estes resultados sejam os primeiros com o manejo desta espĂ©cie, a sua utilização de forma sustentável colaborará para a conservação da Mata Atlântica, pois isto poderá diminuir a pressĂŁo exploratĂłria das demais etno-espĂ©cies denominadas espinheiras santas. Estrutura e composição florĂstica de dois fragmentos da floresta estacional decidual do Alto-Uruguai, SC.
Em dois fragmentos florestais da Floresta Estacional Decidual (FED) do Alto-Uruguai, SC, foram aplicados os mĂ©todos de parcelas (MP) e de pontos-quadrantes (MQ) visando a comparar os mĂ©todos amostrais e caracterizar a riqueza florĂstica e composição estrutural. Foram inventariadas 91 espĂ©cies, sendo que 52 foram encontradas no sub-bosque (sem valor madeireiro), com destaque para Sorocea bonplandii, Gymnanthes concolor e Trichilia claussenii. As outras 39 espĂ©cies emergentes no dossel florestal sĂŁo de uso madeireiro, sendo as mais abundantes Apuleia leiocarpa, Nectandra megapotamica, Cabralea canjerana e Holocalyx balansae. As espĂ©cies de maior densidade e área basal foram praticamente as mesmas em ambos os fragmentos florestais, independente do mĂ©todo usado. O MP detectou uma maior diversidade de espĂ©cies e uma maior densidade de plantas por área comparativamente ao MQ, embora ambos os mĂ©todos detectassem espĂ©cies diferentes. Todavia, o uso simultâneo dos dois mĂ©todos possibilitou em uma maior cobertura da diversidade de espĂ©cies nas duas áreas, com redução no tempo para o inventário. Em ambos os mĂ©todos, detectaram-se erros amostrais acima do esperado e quanto menores os valores do erro amostral, maior foi a similaridade dos resultados das avaliações entre os mĂ©todos. Paradoxalmente a riqueza de espĂ©cies presentes na FED contrasta com o pequeno nĂşmero de fragmentos que restaram neste ecossistema. Importante ressaltar que esses fragmentos sĂŁo as Ăşltimas fontes de sementes para a recolonização de áreas abandonadas ou degradadas, como tambĂ©m para conservação e uso em programas de melhoramento genĂ©tico de essĂŞncias florestais nativas e banco de sementes in situ
Clinical utility of tolvaptan in the management of hyponatremia in heart failure patients
Hyponatremia is an electrolyte disorder frequently observed in several clinical settings and common in hospitalized patients with decompensated heart failure (HF). It is caused by deregulation of arginine vasopressin (AVP) homeostasis associated with water retention in hypervolemic or in euvolemic states. While hypervolemic hypotonic hyponatremia is also seen in advanced liver cirrhosis, renal failure, and nephrotic syndrome, the bulk of evidence associating this electrolyte disorder to increasing morbidity and mortality can be found in the HF literature. Hospitalized HF patients with low serum sodium concentration have lower short-term and long-term survival, longer hospital stay and increased readmission rates. Conventional therapeutic approaches, ie, restriction of fluid intake, saline and diuretics, can be effective, but often the results are unpredictable. Recent clinical trials have demonstrated the effectiveness of nonpeptide AVP receptor antagonists (vaptans) in the treatment of hyponatremia. The vaptans induce aquaresis, an electrolyte-sparing excretion of free water resulting in the correction of serum sodium concentrations and plasma osmolality, without activation of the renin-angiotensin-aldosterone system (RAAS) or changes in renal function and blood pressure. Further prospective studies in a selected congestive HF population with hyponatremia, using clinical-status titrated dose of tolvaptan, are needed to determine whether serum sodium normalization will be translated into a better long-term prognosis. This review will focus on recent clinical trials with tolvaptan, an oral V2 receptor antagonist, in HF patients. The ability of tolvaptan to safely increase serum sodium concentration without activating the RAAS or compromising renal function and electrolyte balance makes it an attractive agent for treating hyponatremic HF patients
The genetic status of Sorocea bonplandii in the highly fragmented forest in southern Brazil.
Sorocea bonplandii is a dioecious sub canopy plant species with an abundant and widespread distribution in the Atlantic Forest, Southern Brazil. It is traditionally used for art crafts and popular medicine. We have investigated the genetic status of S. bonplandii in one large and six small populations of the fragmented Atlantic Forest in southern Brazil. In spite of population fragmentation, the allozyme electrophoretic analysis involving 23 loci of 12 enzymes from 420 plants revealed unusually high frequencies of heterozygous genotypes and a balanced distribution, in agreement with Hardy-Weinberg proportions. Very little genetic differentiation among populations was detected. The observed excess of heterozygotes suggests that selection favors heterozygous plants. Although S. bonplandii is a dominant sub-canopy species which strongly depends on ombrophilous environmental conditions, its genetic structure and diversity do not appear to suffer with forest fragmentation. All of these characteristics, in addition to its medicinal uses, suggest that this species is a potential resource for sustainable management of tropical forests in southern Brazil
Caracterização e dinâmica de duas fases sucessionais em floresta secundária da mata atlântica.
Em Santa Catarina, foi observado aumento da cobertura florestal nas Ăşltimas dĂ©cadas, o que vem construindo uma paisagem florestal retalhada por um grande mosaico de fragmentos de vários estádios sucessionais. Neste trabalho, buscou-se avaliar o dinamismo do processo sucessional de dois diferentes estádios sucessionais. Em área florestal de 40 ha localizada no MunicĂpio de SĂŁo Pedro de Alcântara, SC, abandonada pelo uso agropecuário em meados de 1970, foram estabelecidas aleatoriamente parcelas permanentes (50 x 50 m), duas em estádio florestal secundário mĂ©dio (SM) e quatro em estádio secundário avançado (SA). As avaliações anuais durante o perĂodo de 1994 a 2000 de todas as plantas arbĂłreas com DAP >5 cm revelaram que no SM os valores da densidade de plantas, residentes, recrutadas, mortas e ramificadas foram superiores em relação aos no SA. No entanto, a riqueza de espĂ©cies, área basal e distribuição diamĂ©trica foram superiores no SA. Botanicamente, foram observadas com muita clareza as espĂ©cies e as famĂlias dominantes de cada estádio e igualmente o dinamismo sucessional desse grupo de espĂ©cies, aumento explosivo e posterior declĂnio e substituição, evidenciando-se perfeitamente a funcionalidade dos grupos ecolĂłgicos nessa tipologia florestal. Por fim, destacou-se que a densidade de plantas ramificadas Ă© notadamente superior em estádios florestais secundários iniciais, embora as taxas de incremento corrente anual fossem similares. As avaliações florĂsticas mostraram, ainda, que as florestas no litoral catarinense se encontravam em dinâmica sucessional, em que espĂ©cies climáxicas vĂŞm substituindo paulatinamente o grupo de espĂ©cies pioneiras, elevando a diversidade de espĂ©cies e a biomassa florestal

Effects of combined drug treatments on Plasmodium falciparum : in vitro assays with doxycycline, ivermectin and efflux pump inhibitors
There is great concern regarding the rapid emergence and spread of drug-resistance in Plasmodium falciparum, the parasite responsible for the most severe form of human malaria. Parasite populations resistant to some or all the currently available antimalarial treatments are present in different world regions. Considering the need for novel and integrated approaches to control malaria, combinations of drugs were tested on P. falciparum. The primary focus was on doxycycline, an antibiotic that specifically targets the apicoplast of the parasite. In combination with doxycycline, three different drugs known to inhibit efflux pumps (verapamil, elacridar and ivermectin) were tested, with the assumption that they could increase the intracellular concentration of the antibiotic and consequently its efficacy against P. falciparum. We emphasize that elacridar is a third-generation ABC transporters inhibitor, never tested before on malaria parasites. In vitro experiments were performed on asexual stages of two strains of P. falciparum, chloroquine-sensitive (D10) and chloroquineresistant (W2). Incubation times on asynchronous or synchronous cultures were 72h or 96h, respectively. The antiplasmodial effect (i.e. the IC50) was determined by measuring the activity of the parasite lactate dehydrogenase, while the interaction between drugs was determined through combination index (CI) analyses. Elacridar achieved an IC50 concentration comparable to that of ivermectin, approx. 10-fold lower than that of verapamil, the other tested ABC transporter inhibitor. CI results showed synergistic effect of verapamil plus doxycycline, which is coherent with the starting hypothesis, i.e. that ABC transporters represent potential targets, worth of further investigations, towards the development of companion molecules useful to enhance the efficacy of antimalarial drugs. At the same time, the observed antagonistic effect of doxycycline in combination with ivermectin or elacridar highlighted the importance of drug testing, to avoid the de-facto generation of a sub-dosage, a condition that facilitates the development of drug resistance
