    MiST 3.0: an updated microbial signal transduction database with an emphasis on chemosensory systems

    Bacteria and archaea employ dedicated signal transduction systems that modulate gene expression, second-messenger turnover, quorum sensing, biofilm formation, motility, host-pathogen and beneficial interactions. The updated MiST database provides a comprehensive classification of microbial signal transduction systems. This update is a result of a substantial scaling to accommodate constantly growing microbial genomic data. More than 125 000 genomes, 516 million genes and almost 100 million unique protein sequences are currently stored in the database. For each bacterial and archaeal genome, MiST 3.0 provides a complete signal transduction profile, thus facilitating theoretical and experimental studies on signal transduction and gene regulation. Newsoftware infrastructure and distributed pipeline implemented in MiST 3.0 enable regular genome updates based on the NCBI RefSeq database. A novel MiST feature is the integration of unique profile HMMs to link complex chemosensory systems with corresponding chemoreceptors in bacterial and archaeal genomes

    Adaptive fast multipole methods on the GPU

    We present a highly general implementation of fast multipole methods on graphics processing units (GPUs). Our two-dimensional double precision code features an asymmetric type of adaptive space discretization leading to a particularly elegant and flexible implementation. All steps of the multipole algorithm are efficiently performed on the GPU, including the initial phase which assembles the topological information of the input data.     Quality functions modeling of industrial enterprises products

One of the integral aspects of the quality management systems of industrial enterprises is the requirement of a continuous positive development of all its processes. In this regard, it is necessary to implement the principle of “continuous improvement” and in terms of products produced by the enterprise, thereby reducing loss of product’s quality. The purpose of the paper is to identify possible loss of quality in the stages of the product’s life cycle through the functions’ modeling of product’s quality. The authors carried out the functions’ modeling of product’s quality based on Quality Function Deployment methodology taking into account existing production, technological and material resources of the enterprise, allowing to formulate the condition for structuring of the quality functions and to increase the degree of the consumer’s satisfaction. Modern conditions upgrade issues concerning the search of ways to develop and increase activity efficiency of large-scale industrial associations that possess a high concentration of material and scientific resources, and influence significantly both a certain economic sector or region and the country’s development in general. This paper aims to substantiate author’s techniques to define the impact of corporate entities on a regional social-economic sphere. The authors have highlighted main features of Russian corporate entities and possible forms of ownership. Types of regions and their features the consideration of which is necessary to evaluate regional development are presented on the basis of the author’s estimation procedure. The factor analysis made the foundation for a rating assessment of corporate entities impact on the region; it allowed to estimate quantitatively corporations’ activity and the level of regional development at a certain time period.     Is anticoagulant therapy necessary after hospitalization with COVID-19 pneumonia?

    The coronavirus disease 2019 (COVID-19) pandemic is associated with high virulence, mortality and healthcare burden around the world. One of its features is procoagulant activity, which leads to a high incidence of thromboembolic events in the lungs and other organs. Therefore, from the very onset of the moderate COVID-19, low molecular weight heparins began to be used as anticoagulants, which proved to have a beneficial effect on mortality and the disease course and were included in all guidelines. However, the question on anticoagulant therapy need after discharge from the hospital is controversial. The opinions of various medical professional communities on this issue are divided. In particular, some of them, including the Russian Ministry of Health guidelines recommend 30-45day anticoagulation using novel oral anticoagulants (dabigatran, rivaroxaban, apixaban), but other sources do not provide such recommendations. This review discusses the effectiveness of anticoagulant therapy after COVID-19, as well as the need to use stratification scales to assess this therapy

    Isolation, Purification and Evaluation of Serological Activity of Rabies Virus Antigens

    Objective of the study is to evaluate the serological activity of rabies virus antigens isolated from the brain tissue of mice by homogenization on FastPrep followed by ultracentrifugation. Materials and methods. Producer strain of the rabies virus “Ovechiy” GNKI. The rabies virus was isolated from the brain tissue of experimentally infected mice, followed by the study of the electrophoretic profile. The serological activity of the virus components was assessed by immunoblot and ELISA using specific anti-rabies sera.Results and conclusions. In the course of comparing the methods of isolation and purification of the rabies virus antigen, it was found that most optimal one is to use a homogenization on FastPrep-24, followed by fractionation in a sucrose gradient. As a result of fractionation in a graded sucrose density gradient with a concentration of 15–50 % at 25000 g for 120 min, five fractions of the rabies virus components were obtained. The maximum purified protein fraction was from 15–20 % sucrose zone, which corresponded to a molecular weight of 67 kDa. The specific antigen activity of the fraction in ELISA reached up the titers of 1:1280 (Specificity coefficient 2.2). Using immunoblot of antigens, obtained from the sucrose gradient in the range of 40–45 % and 20–35 % after ultracentrifugation, one major fraction of polypeptides (54 kDa) was detected, which showed the highest antigenic activity. The results obtained will be useful in the design of test systems for rabies screening and monitoring the effectiveness of anti-epizootic measures
