Greb1 is required for axial elongation and segmentation in vertebrate embryos
During vertebrate embryonic development, the formation of axial structures is driven by a population of stem-like cells that reside in a region of the tailbud called the chordoneural hinge (CNH). We have compared the CNH transcriptome with those of surrounding tissues and shown that the CNH and tailbud mesoderm are transcriptionally similar, and distinct from the presomitic mesoderm. Amongst CNH-enriched genes are several that are required for axial elongation, including Wnt3a, Cdx2, Brachyury/T and Fgf8 , and androgen/estrogen receptor nuclear signalling components such as Greb1 . We show that the pattern and duration of tailbud Greb1 expression is conserved in mouse, zebrafish, and chicken embryos, and that Greb1 is required for axial elongation and somitogenesis in zebrafish embryos. The axial truncation phenotype of Greb1 morphant embryos is explained by much reduced expression of No tail ( Ntl/Brachyury ) which is required for axial progenitor maintenance. An integrated modelling framework to assess cascade water reuse in urban areas
In the recent years water scarcity has been an increasing problem for many countries worldwide. For this reason, there is currently a strong focus on increasing reclaimed wastewater reuse, especially for agriculture purposes (Fernandes and Cunha Marques, 2023). Besides, the cost of energy from conventional resources is increasing, thus the energy sector is moving towards more distributed and efficient use of heat sources across urban areas. Typical applications are heat pumps using local groundwater reservoirs and subsequently discharging in the nearby surface water bodies/artificial channels (recipients). Furthermore, for a better quality of these recipients and for a better performance of wastewater treatment plants (WWTP), stormwater can be collected in separated sewers discharging only the urban runoff to the recipient. In this context, water is subjected to multiple uses, with potential cross-contaminations across different compartments, posing a risk for the environment. Hence, there is a strong need for tools capable of supporting stakeholders towards a wiser and safer use of water resources, to ensure long-term resilience, stability, sustainability and security of the society with regard to water use. A decade of monitoring micropollutants in urban wet-weather flows: What did we learn?
Urban wet-weather discharges from combined sewer overflows (CSO) and stormwater outlets (SWO) are a potential pathway for micropollutants (trace contaminants) to surface waters, posing a threat to the environment and possible water reuse applications. Despite large efforts to monitor micropollutants in the last decade, the gained information is still limited and scattered. In a metastudy we performed a data-driven analysis of measurements collected at 77 sites (683 events, 297 detected micropollutants) over the last decade to investigate which micropollutants are most relevant in terms of 1) occurrence and 2) potential risk for the aquatic environment, 3) estimate the minimum number of data to be collected in monitoring studies to reliably obtain concentration estimates, and 4) provide recommendations for future monitoring campaigns. We highlight micropollutants to be prioritized due to their high occurrence and critical concentration levels compared to environmental quality standards. These top-listed micropollutants include contaminants from all chemical classes (pesticides, heavy metals, polycyclic aromatic hydrocarbons, personal care products, pharmaceuticals, and industrial and household chemicals). Analysis of over 30,000 event mean concentrations shows a large fraction of measurements (> 50%) were below the limit of quantification, stressing the need for reliable, standard monitoring procedures. High variability was observed among events and sites, with differences between micropollutant classes. The number of events required for a reliable estimate of site mean concentrations (error bandwidth of 1 around the “true" value) depends on the individual micropollutant. The median minimum number of events is 7 for CSO (2 to 31, 80%-interquantile) and 6 for SWO (1 to 25 events, 80%-interquantile). Our analysis indicates the minimum number of sites needed to assess global pollution levels and our data collection and analysis can be used to estimate the required number of sites for an urban catchment. Our data-driven analysis demonstrates how future wet-weather monitoring programs will be more effective if the consequences of high variability inherent in urban wet-weather discharges are considered
Covid-19 patient management in outpatient setting: A population-based study from southern italy
Evidence on treatments for early-stage COVID-19 in outpatient setting is sparse. We explored the pattern of use of drugs prescribed for COVID-19 outpatients’ management in Southern Italy in the period February 2020–January 2021. This population-based cohort study was conducted using COVID-19 surveillance registry from Caserta Local Health Unit, which was linked to claims databases from the same catchment area. The date of SARS-CoV-2 infection diagnosis was the index date (ID). We evaluated demographic and clinical characteristics of the study drug users and the pattern of use of drugs prescribed for outpatient COVID-19 management. Overall, 40,030 patients were included in the analyses, with a median (IQR) age of 44 (27–58) years. More than half of the included patients were asymptomatic at the ID. Overall, during the study period, 720 (1.8%) patients died due to COVID-19. Azithromycin and glucocorticoids were the most frequently prescribed drugs, while oxygen was the less frequently prescribed therapy. The cumulative rate of recovery from COVID-19 was 84.2% at 30 days from ID and it was lower among older patients. In this study we documented that the drug prescribing patterns for COVID-19 treatment in an outpatient setting from Southern Italy was not supported from current evidence on beneficial therapies for early treatment of COVID-19, thus highlighting the need to implement strategies for improving appropriate drug prescribing in general practice
Modeling Water Quality in Watersheds: From Here to the Next Generation
In this synthesis, we assess present research and anticipate future development needs in modeling water quality in watersheds. We first discuss areas of potential improvement in the representation of freshwater systems pertaining to water quality, including representation of environmental interfaces, in-stream water quality and process interactions, soil health and land management, and (peri-)urban areas. In addition, we provide insights into the contemporary challenges in the practices of watershed water quality modeling, including quality control of monitoring data, model parameterization and calibration, uncertainty management, scale mismatches, and provisioning of modeling tools. Finally, we make three recommendations to provide a path forward for improving watershed water quality modeling science, infrastructure, and practices. These include building stronger collaborations between experimentalists and modelers, bridging gaps between modelers and stakeholders, and cultivating and applying procedural knowledge to better govern and support water quality modeling processes within organizations
A major clinical event following transperineal prostate biopsy: An unexpected duodenal perforation
A 73-year-old Caucasian male underwent an ultrasound-guided transperineal prostate biopsy in an outpatient setting. Prostate cancer was suspected because of persistently rising prostate-specific antigen levels. The procedure was concluded early because the patient complained of extreme discomfort after the transrectal probe was inserted. After less than an hour, the patient described an acute abdominal pain that began suddenly after completion of the biopsy. Results of a computed tomography scan led to an emergency exploratory laparotomy. This procedure revealed a small duodenal perforation that was caused by a peptic ulcer and immediately repaired. The major clinical event of duodenal perforation following the execution of a transperineal prostate biopsy posed an immediate diagnostic challenge. The closely related timing of these 2 distinct and unlinked events represents the most unusual aspect of the case. It is a reminder that a concurrent disease always has to be considered and excluded despite the temporal closeness of reckoned occurrences. © 2010
