    WHEN THE SAP FLOWS: AFFECTION AND INDUSTRY IN THE MAPLE WOODS

    This thesis arises from my time waiting for, collecting, and boiling sap from maple trees into syrup. I spent four months in the Adirondack Mountains of upstate New York working for a modern commercial sugaring operation and sugaring in the old-fashioned manner—with buckets and a wood-fired evaporator. The narrative follows my journey as a lifelong Westerner traveling east to learn an old tradition with my hands. Instead of observing how the warmth of a changing climate was affecting maple sugaring, I was thrust into a landscape defined by cold, during one of the coldest winters on record in the Northeast. The pith of the story centers around my inquiry into what Wendell Berry calls “the real work” and Robert Bringhurst names “vocation.” What calls to me in life? What jolts me into feeling alive? What do I value? Based on what I value how and where should I live? In many episodes, I begin to craft ways I would like to live in relation to the earth and its communities, my loved ones, and myself. I share my own struggle to approach the world affectionately, graciously and deliberately. I investigate what defines home for me, and how I can come to know that home more intimately. And I question the intentions behind collecting sap from the maple trees. Should sap be received as a gift or taken as a commodity? Through a long grip of winter and a splash of early spring, this story seeks to understand place through work, love through play, and purpose through question

    To efficiently bud off from infected cells, HIV and other enveloped viruses hijack the host cellular machinery that is normally involved in vacuolar protein sorting and multivesicular body (MVB) biogenesis. The HIV Gag protein mimics hepatocyte growth factor–regulated tyrosine kinase substrate (Hrs), a modular adaptor protein that links membrane cargo recognition to its degradation after delivery to MVBs. In contrast to T cells, where HIV budding occurs at the plasma membrane, virus buds into vacuoles of macrophages, a process that may facilitate its spread within the infected host

    Severe B Cell Deficiency in Mice Lacking the Tec Kinase Family Members Tec and Btk

    The cytoplasmic protein tyrosine kinase Tec has been proposed to have important functions in hematopoiesis and lymphocyte signal transduction. Here we show that Tec-deficient mice developed normally and had no major phenotypic alterations of the immune system. To reveal potential compensatory roles of other Tec kinases such as Bruton's tyrosine kinase (Btk), Tec/Btk double-deficient mice were generated. These mice exhibited a block at the B220+CD43+ stage of B cell development and displayed a severe reduction of peripheral B cell numbers, particularly immunoglobulin (Ig)MloIgDhi B cells. Although Tec/Btknull mice were able to form germinal centers, the response to T cell–dependent antigens was impaired. Thus, Tec and Btk together have an important role both during B cell development and in the generation and/or function of the peripheral B cell pool. The ability of Tec to compensate for Btk may also explain phenotypic differences in X-linked immunodeficiency (xid) mice compared with human X-linked agammaglobulinemia (XLA) patients

    Plasticity of CD4+ T Cell Lineage Differentiation

    The differentiation of naive CD4+ T cells into lineages with distinct effector functions has been considered to be an irreversible event. T helper type 1 (Th1) cells stably express IFN-γ, whereas Th2 cells express IL-4. The discovery and investigation of two other CD4+ T cell subsets, induced regulatory T (iTreg) cells and Th17 cells, has led to a rethinking of the notion that helper T cell subsets represent irreversibly differentiated endpoints. Accumulating evidence suggests that CD4+ T cells, particularly iTreg and Th17 cells, are more plastic than previously appreciated. It appears that expression of Foxp3 by iTreg cells or IL-17 by Th17 cells may not be stable and that there is a great degree of flexibility in their differentiation options. Here, we will discuss recent findings that demonstrate the plasticity of CD4+ T cell differentiation and the biological implications of this flexibility

    This article pursues two distinct, but related hypotheses. First, as total LSAT takers decline, we expect to see a decline in the number of new attorneys admitted to the patent bar. Second, as the number of new patent attorneys shrinks and the number of women pursuing engineering degrees increases, we expect that the patent bar will become more female. In order to test these hypotheses, we gathered and collated data from the Law School Admission Counsel (LSAC) regarding students taking the Law School Admissions Test (LSAT), the United States Patent and Trademark Office (USPTO), the Society of Women Engineers (SWE), and the American Bar Association (ABA). The data establishes that the first hypothesis is true, but the second one is false. That is, the number of new entrants to the patent bar will drop precipitously. By 2018, new entrants will number one half of what they were in 2008. However, the number of female patent attorneys compared to the number of male patent attorneys will not change in this same time period. That is, even though the patent bar will shrink, the patent bar will not become more female
