    Feeding barley grain-rich diets altered electrophysiological properties and permeability of the ruminal wall in a goat model

    High-producing ruminants are commonly fed large amounts of concentrate to meet their high energy demands for rapid growth or high milk production. However, this feeding strategy can severely impair rumen functioning, leading to subacute ruminal acidosis. Subacute ruminal acidosis might have consequences for electrophysiological properties by changing the net ion transfer and permeability of ruminal epithelia, which may increase the uptake of toxic compounds generated in the rumen into the systemic circulation. The objective of the present study was to investigate the effects of excessive barley feeding on the electrophysiological and barrier functions of the ruminal epithelium and serum inflammation and ketogenesis markers after a long-term feeding challenge, using growing goats as a ruminant model. A feeding trial was carried out with growing goats allocated to 1 of the 3 groups (n=5-6 animals/group), with diets consisting exclusively of hay (control diet) or hay with 30 or 60% barley grain. Samples of the ventral ruminal epithelium were taken after euthanasia and instantly subjected to Ussing chamber experiments, where electrophysiological properties of the epithelium were measured in parallel with the permeability of marker molecules of different sizes [fluorescein 5(6)-isothiocyanate and horseradish peroxidase] from luminal to apical side. Additionally, ruminal fluid and blood samples were taken at the beginning of the experiment as well as shortly before euthanasia. Ruminal fluid samples were analyzed for volatile fatty acids and pH, whereas blood samples were analyzed for lipopolysaccharide, serum amyloid A, and β-hydroxybutyrate. Electrophysiological data indicated that barley feeding increased the epithelial short-circuit current compared with the control. Tissue conductance also increased with dietary barley inclusion. As shown with both marker molecules, permeability of ruminal epithelia increased with barley inclusion in the diet. Despite a lowered ruminal pH associated with increased volatile fatty acids (such as propionate and butyrate) concentrations as well as altered epithelial properties in response to high-grain feeding, no signs of inflammation became apparent, as blood serum amyloid A concentrations remained unaffected by diet. However, greater amounts of grain in the diet were associated with a quadratic increase in lipopolysaccharide concentration in the serum. Also, increasing the amounts of barley grain in the diet resulted in a tendency to quadratically augment serum concentrations of β-hydroxybutyrate and, hence, the alimentary ketogenesis. Further studies are needed to clarify the role of barley inclusion in the development of subacute ruminal acidosis in relation to ruminal epithelial damage and the translocation of toxic compounds in vivo.     Diversity of methicillin-resistant coagulase-negative Staphylococcus spp. and methicillin-resistant Mammaliicoccus spp. isolated from ruminants and New World camelids

    Information about livestock carrying methicillin-resistant coagulase-negative staphylococci and mammaliicocci (MRCoNS/MRM) is scarce. The study was designed to gain knowledge of the prevalence, the phenotypic and genotypic antimicrobial resistance and the genetic diversity of MRCoNS/MRM originating from ruminants and New World camelids. In addition, a multi-locus sequence typing scheme for the characterization of Mammaliicoccus (formerly Staphylococcus) sciuri was developed. The study was conducted from April 2014 to January 2017 at the University Clinic for Ruminants and the Institute of Microbiology at the University of Veterinary Medicine Vienna. Seven hundred twenty-three nasal swabs originating from ruminants and New World camelids with and without clinical signs were examined. After isolation, MRCoNS/MRM were identified by MALDI-TOF, rpoB sequencing and typed by DNA microarray-based analysis and PCR. Antimicrobial susceptibility testing was conducted by agar disk diffusion. From all 723 nasal swabs, 189 MRCoNS/MRM were obtained. Members of the Mammaliicoccus (M.) sciuri group were predominant (M. sciuri (n = 130), followed by M. lentus (n = 43), M. fleurettii (n = 11)). In total, 158 out of 189 isolates showed phenotypically a multi-resistance profile. A seven-loci multi-locus sequence typing scheme for M. sciuri was developed. The scheme includes the analysis of internal segments of the house-keeping genes ack, aroE, ftsZ, glpK, gmk, pta1 and tpiA. In total, 28 different sequence types (STs) were identified among 92 selected M. sciuri isolates. ST1 was the most prevalent ST (n = 35), followed by ST 2 (n = 15), ST3 and ST5 (each n = 5), ST4 (n = 3), ST6, ST7, ST8, ST9, ST10 and ST11 (each n = 2)

    Untersuchung der Serokonversion auf Chlamydia abortus von Schafen aus der Region Vorarlberg vor und nach der Alpung

    Insgesamt 796 Serumproben von gesömmerten Schafen in der Region Vorarlberg wurden mittels eines kommerziellen ELISA-Kits auf Antikörper gegen Chlamydia (C.) abortus, den Erreger des enzootischen Schafabortes, untersucht. Ziel dieser Studie war es die Seroprävalenz dieser Region zu eruieren, ein Vergleich zu der Seroprävalenz des benachbarten Kanton Graubündens zu erstellen sowie die Serokonversion während der Alpung festzustellen. 421 Proben wurden vor der Alpung, die restlichen 375 nach der Alpung gesammelt, wobei von 359 Tieren korrelierende Serumpaare vorhanden waren. Für das untersuchte Gebiet Vorarlberg ergab sich eine mittlere Seroprävalenz von 9.2 % bei einem Schwellenwert von 60 %; bei 5.0 % der Tiere mit korrelierenden Serumproben konnte von vor der Alpung seronegativen Tieren nach der Alpung C. abortus-Antikörper nachgewiesen werden. Die Seroprävalenzwerte entsprechen denen schweizerischer Gebiete, die sowohl geografisch als auch in der Art der Sömmerung der untersuchten Region ähnlich sind. Nur der benachbarte Kanton Graubünden wies mit 43 % bedeutend höhere Werte auf. Die Resultate dieser Studie zeigen, dass der traditionelle Tierverkehr zwischen diesen beiden Regionen die Verbreitung von C. abortus nicht begünstigt. In total, 796 serum samples of sheep on commune alpine pastures in the region of Vorarlberg were investigated by a commercial ELISA kit for antibodies against Chlamydia abortus, the agent of ovine enzootic abortion. The aim of the study was to determine the seroprevalence within this region and to compare these results with the seroprevalence in the neighboring canton Graubünden as well as to obtain data on the seroconversion after alpine pasturing. Therefore, 421 samples were collected before and 375 samples after alpine pasturing, whereas corresponding serum samples were available from 359 sheep. Within the region of Vorarlberg, a mean seroprevalence of 9.2 % was calculated with a threshold of 60 %. Seroconversion for C. abortus occurred in 5.0 % of animals with corresponding serum samples. Seroprevalence values were comparable to Swiss regions with similar management systems, although the neighboring canton Graubünden is known to have a much more higher seroprevalence of 43 %. In conclusion, the traditional animal exchange between these two regions is not significantly favoring the spread of C. abortus
