    Application of the linear principle for the strongly-correlated variables: Calculations of differences between spectra

    In this paper the authors suggest a new method of detection of possible differences between similar near infrared (NIR) spectra based on the self-similar (fractal) property. This property is a general characteristic that belongs to a wide class of the strongly-correlated systems. As an example we take a set of NIR spectra measured for three systems: (1) glassy carbon (GC) electrodes, (2) GC electrodes affected by azobenzene (AB) substance and finally (3) films (AB-FILM). Besides the physical model that should describe the intrinsic properties of these substances we found the fitting function that follow from the linear principle for the strongly-correlated variables. This function expressed in the form of linear combination of 4 power-law functions describes with the high accuracy the integrated curves that were obtained from the averaged values of the initially measured spectra. The nine fitting parameters can be considered as the quantitative "finger prints" for detection of the differences between similar spectra. Besides this result we established the self-similar behavior of the remnant functions. In other words, the difference between the initially integrated function and its fitting function can be expressed in the form of linear combinations of periodical functions having a set of frequencies following to relationship ω(k)=ω0ξk, where the initial frequency ω0 and scaling factor ξ are determined by the eigen-coordinates method. This behavior in the NIR spectra was discovered in the first time and physical reasons of such behavior merit an additional research. © 2011

    Elevated levels of protein AMBP in cerebrospinal fluid of women with preeclampsia compared to normotensive pregnant women

    Purpose: To investigate the cerebrospinal fluid (CSF) proteome of patients with preeclampsia (PE) and normotensive pregnant women, in order to provide a better understanding of brain involvement in PE. Experimental design: Ninety-eight CSF samples (43 women with PE and 55 normotensive controls) were analyzed by LC-MS/MS proteome profiling. CSF was obtained during the spinal puncture before caesarean delivery. Results: Eight proteins were higher abundant and 17 proteins were lower abundant in patients with PE. The most significantly differentially abundant protein was protein AMBP (alpha-1-microglobulin/bikunin precursor). This finding was validated by performing an ELISA experiment (p = 0.002). Conclusions and clinical relevance: The current study showed a clear difference between the protein profiles of CSF from patients with PE and normotensive pregnant women. Protein AMBP is a precursor of a heme-binding protein that counteracts the damaging effects of free hemoglobin, which may be related to the presence of free hemoglobin in CSF. Protein levels showed correlations with clinical symptoms during pregnancy and postpartum.     Proteomic alterations in early stage cervical cancer

    Laser capture microdissection (LCM) allows the capture of cell types or welldefined structures in tissue. We compared in a semi-quantitative way the proteomes from an equivalent of 8,000 tumor cells from patients with squamous cell cervical cancer (SCC, n = 22) with healthy epithelial and stromal cells obtained from normal cervical tissue (n = 13). Proteins were enzymatically digested into peptides which were measured by high-resolution mass spectrometry and analyzed by "all-ornothing" analysis, Bonferroni, and Benjamini-Hochberg correction for multiple testing. By comparing LCM cell type preparations, 31 proteins were exclusively found in early stage cervical cancer (n = 11) when compared with healthy epithelium and stroma, based on criteria that address specificity in a restrictive "all-or-nothing" way. By Bonferroni correction for multiple testing, 30 proteins were significantly up-regulated between early stage cervical cancer and healthy control, including six members of the MCM protein family. MCM proteins are involved in DNA repair and expected to be participating in the early stage of cancer. After a less stringent Benjamini-Hochberg correction for multiple testing, we found that the abundances of 319 proteins were significantly different between early stage cervical cancer and healthy controls.     It has now been well established that in Rheumatoid Arthritis early treatment with disease modifying antirheumatic drugs (DMARDs) should be initiated to control the inflammatory activity, structural damage and function. Toxicity of the drugs should be monitored carefully. In cases with insufficient response aggressive treatment options can create a difference so additional strategies should be taken into consideration in time. This review mentions about the more commonly prescribed DMARDs such as methotrexate, sulphasalazine and leflunomide. Practical tips on special situations like pregnancy, lactation and perioperative period are given, first drug choice and combination therapies are summarized

    During pregnancy 30gr of calcium (Ca) is transferred to the fetus and during lactation about 300-400 mg of Ca is lost through breast milk daily. Normally this condition is compensated by physiologic adaptations and a near full recovery is expected with weaning. Occasionally some women experience severe osteoporosis with fragility fractures during pregnancy or shortly after delivery. Although some cases with pregnancy and lactation- associated osteoporosis have "traditional" risk factors for osteoporosis, there are cases without any known risk factors as well. There is no consensus about the ideal treatment but weaning, correction of vitamin D deficiency, Ca-vitamin D supplementation and thorocolomber orthosis are usually prescribed. Treatment strategy might include addition of a bisphosphonate. Treatment with strontium ranelate or teriparatide has been recommended in case reports.
